BINA SARANA SUKSES JOBSITE PT. MANAMBANG MUARA ENIM

Disposal area pada tambang terbuka merupakan tempat penimbunan material tanah
penutup yang harus direncanakan secara cermat agar lereng tetap stabil dan aman dari
longsoran. Stabilitas lereng disposal dipengaruhi oleh karakteristik material timbunan,
perubahan kondisi material, serta curah hujan yang dapat memicu terbentuknya bidang
gelincir. Penelitian ini dilakukan di PT. Bina Sarana Sukses Jobsite PT. Manambang
Muara Enim, Sumatera Selatan, dengan tujuan menganalisis kestabilan timbunan
menggunakan metode Waste Dump and Stockpile Stability Rating and Hazard
Classification System (WSRHC). Hasil analisis menunjukkan bahwa timbunan
memiliki bobot kestabilan sebesar 20 dan termasuk kategori IV (risiko tinggi), dengan
nilai Engineering Geology Index (EGI) sebesar -4 dan Design and Performance Index
(DPI) sebesar 24. Upaya yang direkomendasikan meliputi investigasi bawah
permukaan, pengujian laboratorium, analisis stabilitas terperinci, pengaturan tahapan
konstruksi, serta peningkatan monitoring untuk meminimalkan risiko longsor pada
disposal area.
A disposal area in open-pit mining is a location for dumping overburden material that
must be carefully planned to ensure slope stability and prevent landslides. The stability
of disposal slopes is influenced by the characteristics of the fill material, changes in
material conditions, and rainfall that can trigger the formation of slip surfaces. This
study was conducted at PT. Bina Sarana Sukses Jobsite PT. Manambang Muara Enim,
South Sumatra, with the aim of analyzing dump stability using the Waste Dump and
Stockpile Stability Rating and Hazard Classification (WSRHC) System method. The
analysis results showed that the dump has a stability score of 20 and is classified as
Category IV (high risk), with an Engineering Geology Index (EGI) value of -4 and a
Design and Performance Index (DPI) value of 24. Recommended efforts include
subsurface investigations, laboratory testing, detailed stability analysis, proper
construction stage management, and increased monitoring to minimize the risk of
landslides in the disposal area.
5090254319J1E022046A Narrative Inquiry into English Culinary Project Presentations in Enhancing Students' Speaking SkillsPenelitian ini mengeksplorasi integrasi metode Project-Based Learning (PjBL) ke dalam English for Specific Purposes (ESP) dalam konteks Sekolah Menengah Kejuruan (SMK). Studi ini mengkaji bagaimana program Presentasi Proyek Kuliner Bahasa Inggris meningkatkan keterampilan berbicara siswa kelas sebelas jurusan Kuliner di SMK Mardikenya Purwokerto tahun ajaran 2024/2025. Dengan pendekatan naratif kualitatif, data dikumpulkan melalui wawancara semiterstruktur dengan guru bahasa Inggris sebagai partisipan utama dan koordinator proyek sebagai informan, serta analisis dokumen panduan proyek dan modul ajar. Data dianalisis menggunakan analisis tematik dan analisis isi untuk menjawab tiga pertanyaan penelitian mengenai: (1) bagaimana proyek dilaksanakan, (2) bagaimana PjBL diintegrasikan ke dalam ESP, dan (3) apa persepsi guru terhadap dampak proyek pada keterampilan berbicara siswa. Temuan penelitian menunjukkan bahwa (1) proyek mengikuti proses implementasi lima tahap yang terstruktur; (2) integrasi bahasa Inggris ke dalam proyek kuliner dicapai melalui persiapan kelas yang sistematis, berfokus pada imperative sentences, sequence words, measurements, descriptive adjectives, dan frasa presentasi yang diselaraskan dengan tugas proyek; dan (3) guru mempersepsikan peningkatan signifikan pada keterampilan berbicara siswa, khususnya dalam aspek kemauan berbicara, kepercayaan diri, pelafalan, kosakata, dan kemampuan berbicara di depan umum. Secara keseluruhan, studi ini membuktikan bahwa program Presentasi Proyek Kuliner Bahasa Inggris berfungsi sebagai model efektif untuk mengintegrasikan PjBL ke dalam konteks ESP di SMK.This research explores the integration of Project-Based Learning (PjBL) into English for Specific Purposes (ESP) within a vocational education context. The study investigates how the English Culinary Project Presentations program enhanced the speaking skills of eleventh-grade Culinary Arts students at SMK Mardikenya Purwokerto during the 2024/2025 academic year. Guided by a qualitative narrative inquiry design, data were collected via semi-structured interviews with the English teacher as the main participant and the project coordinator as the informant, alongside a document analysis of project guidelines and lesson plans. Data were analyzed using thematic and content analysis to answer three research questions regarding: (1) how the project was carried out, (2) how PjBL was integrated into ESP, and (3) what are the teacher’s perceptions of the project's impact on students' speaking skills. The findings reveal that: (1) the project followed a structured five-stage implementation process; (2) English was integrated into the culinary project through systematic classroom preparation, focusing on imperative sentences, sequence words, measurements, descriptive adjectives, and presentation phrases aligned with project tasks; and (3) the teacher perceived significant enhancements in students' speaking skills, specifically regarding willingness to speak, confidence, pronunciation, vocabulary, and public speaking ability. Overall, this study demonstrates that the English Culinary Project Presentations program serves as an effective model for integrating PjBL into vocational ESP contexts.

Curug Cilember merupakan salah satu destinasi wisata alam yang terletak di Desa Jogjogan, Kecamatan Cisarua, Kabupaten Bogor, yang dikenal memiliki tujuh tingkatan air terjun di kawasan hutan pinus. Meskipun memiliki potensi wisata yang besar, pengembangannya masih menghadapi kendala infrastruktur pendukung, pengelolaan, dan promosi. Penelitian ini bertujuan untuk menganalisis kontribusi objek wisata Curug Cilember terhadap Pendapatan Asli Daerah (PAD) Kabupaten Bogor serta merumuskan strategi pengembangan destinasi yang tepat.Cilember Waterfall is a natural tourist destination located in Jogjogan Village, Cisarua District, Bogor Regency, known for its seven-tiered waterfalls and pine forest. Despite its significant tourism potential, development still faces obstacles such as limited supporting infrastructure, suboptimal destination management, and limited tourism promotion. This study aims to analyze the contribution of the Cilember Waterfall tourist attraction to Bogor Regency's Local Own-source Revenue (PAD) and to formulate an appropriate tourism destination development strategy.

Wanprestasi dalam perjanjian pemborongan furniture dapat terjadi apabila
pekerjaan tidak diselesaikan sesuai waktu maupun spesifikasi yang disepakati.
Salah satunya terdapat dalam Putusan Pengadilan Negeri Denpasar Nomor
9/Pdt.G.S/2025/PN Dps antara PT Hadi Investama Abadi sebagai Penggugat dan
KD Dody Budi Sayoga sebagai Tergugat yang tidak menyelesaikan pekerjaan
furniture sesuai waktu yang diperjanjikan. Penelitian ini bertujuan untuk
menganalisis pertimbangan hukum hakim dalam menetapkan wanprestasi serta
akibat hukum ganti rugi dalam Putusan Pengadilan Negeri Denpasar Nomor
9/Pdt.G.S/2025/PN Dps. Penelitian ini menggunakan metode yuridis normatif
dengan pendekatan perundang-undangan (statute approach) dan pendekatan kasus
(case approach), bersifat preskriptif, menggunakan data sekunder melalui studi
kepustakaan dan dianalisis secara normatif kualitatif. Hasil penelitian
menunjukkan bahwa hakim menetapkan wanprestasi berdasarkan perjanjian yang
sah, tidak dipenuhinya kewajiban penyelesaian pekerjaan dalam jangka waktu
yang diperjanjikan, dilakukannya somasi namun kewajiban tetap tidak terpenuhi,
serta tidak terbuktinya keadaan memaksa. Namun, pertimbangan hakim lebih
menitikberatkan pada keterlambatan pekerjaan dan belum menguraikan secara
mendalam ketidaksempurnaan hasil pekerjaan sebagai bentuk wanprestasi. Akibat
hukum yang timbul adalah dikabulkannya gugatan Penggugat dan dihukumnya
Tergugat membayar ganti rugi materiil sebesar Rp33.901.400,00. Menurut
penulis, putusan tersebut telah sesuai dengan pasal 1243 KUHPdt, karena kerugian
yang dialami Penggugat merupakan akibat langsung dari wanprestasi yang
dilakukan Tergugat sehingga hubungan kausal antara wanprestasi dan kerugian
telah terpenuhi.
Breach of contract in a furniture construction agreement may occur when the work
is not completed within the agreed period or in conformity with the agreed
specifications. Such a case is reflected in the Denpasar District Court Decision
Number 9/Pdt.G.S/2025/PN Dps between PT Hadi Investama Abadi as the Plaintiff
and KD Dody Budi Sayoga as the Defendant, who failed to complete the furniture
work within the agreed period. This study aims to analyze the judge's legal
considerations in establishing the breach of contract and the legal consequences of
the award of damages in the said decision. The research employs a normative
juridical method with statutory and case approaches, is prescriptive in nature, using
secondary data obtained through library research and analyzed using qualitative
normative analysis. The findings show that the judge established the breach based
on a valid agreement, the failure to complete the work within the stipulated period,
the issuance of formal notices of default (somasi) which nevertheless remained
unfulfilled, and the unproven existence of force majeure. However, the
considerations emphasized the delay and did not thoroughly elaborate on the
defective workmanship as a form of breach of contract. The Plaintiff's claim was
granted, and the Defendant was ordered to pay material damages of IDR
33,901,400.00. In the author's opinion, the decision accords with Article 1243 of the
Indonesian Civil Code, as the losses suffered by the Plaintiff were a direct result of
the Defendant's breach, thereby satisfying the causal link between the breach and
the loss.

STUDI KASUS PERATURAN GUBERNUR DKI JAKARTA NOMOR 63 TAHUN 2020
Artikel ini menjelaskan dimensi politik di balik implementasi Peraturan Gubernur DKI Jakarta Nomor 63 Tahun 2020 tentang Penugasan Kepada Badan Usaha Milik Daerah Untuk Menyelenggarakan Sistem Integrasi Pembayaran Antar Moda Transportasi, dengan berfokus pada dua pertanyaan, yaitu bagaimana dinamika kontestasi koalisi advokasi berlangsung dalam tahap implementasi sistem integrasi pembayaran transportasi di bawah Pergub Nomor 63 Tahun 2020, dan sejauh mana belief system serta strategi politik dari koalisi mendikte output serta outcome kebijakan. Menggunakan kerangka Advocacy Coalition Framework yang dipadukan dengan Model Implementasi Kebijakan Top-Down, artikel ini membedah bagaimana koalisi yang ada berkontestasi dengan belief system masing-masing pada fase formulasi hingga fase implementasi. Menggunakan pendekatan kualitatif deskriptif-analitis melalui wawancara semiterstruktur, studi dokumentasi, dan observasi lapangan, penelitian ini mengidentifikasi tiga koalisi advokasi, yaitu Koalisi Pro-Integrasi Teknologi (Pemprov DKI, PT Jaklingko, operator modern), Koalisi Operator Tradisional (Organda, sopir angkot), dan Koalisi Fokus Pengguna (konsumen, lembaga riset, dan akademisi). Temuan menunjukkan bahwa dominasi Koalisi Pro Integrasi Teknologi menghasilkan output implementasi yang positif di moda modern, tercermin dari pertumbuhan penumpang Transjakarta 11,21%, MRT Jakarta 12,95%, dan LRT Jakarta 8,97% per-tahun 2025, resistensi Koalisi Operator Tradisional melalui strategi slow compliance memperlambat integrasi di moda konvensional. Pada level outcome, penelitian ini menemukan kesenjangan, keberhasilan output belum berhasil menekan kemacetan Jakarta yang melonjak ke peringkat ke-24 kota termacet di dunia tahun 2025 menurut TomTom Traffic Index, naik tajam dari peringkat 90 pada tahun sebelumnya. Penelitian ini memperkaya literatur ACF dengan menunjukkan bahwa kontestasi koalisi tidak berhenti pada fase formulasi, melainkan terus berlangsung secara dinamis sepanjang implementasi kebijakan berjalan.
This article examines the political dimensions underlying the implementation of Jakarta Governor Regulation No. 63 of 2020 concerning the Assignment of Regional Owned Enterprises to Organize an Integrated Payment System Across Transportation Modes. It focuses on two research questions: how the dynamics of advocacy coalition contestation unfold during the implementation stage of the transportation payment integration system under Governor Regulation No. 63 of 2020, and to what extent the belief systems and political strategies of these coalitions dictate policy output and outcomes. Using the Advocacy Coalition Framework combined with the Top-Down Policy Implementation Model, this article dissects how the existing coalitions contest one another based on their respective belief systems, from the formulation stage through to implementation. Using a descriptive-analytical qualitative approach involving semi-structured interviews, documentary study, and field observation, this research identifies three advocacy coalitions: the Pro-Technology Integration Coalition (DKI Provincial Government, PT Jaklingko, modern operators), the Traditional Operator Coalition (Organda, angkot drivers), and the User-Focused Coalition (consumers, research institutions, and academics). The findings show that the dominance of the Pro-Technology Integration Coalition produced positive implementation outputs in modern modes, reflected in passenger growth of 11.21% for Transjakarta, 12.95% for MRT Jakarta, and 8.97% for LRT Jakarta over the course of 2025, while resistance from the Traditional Operator Coalition through a slow compliance strategy hindered integration in conventional modes. At the outcome level, this research finds a gap: the success at the output level has not managed to reduce Jakarta's traffic congestion, which instead surged to the 24th
most congested city in the world in 2025 according to the TomTom Traffic Index, a sharp rise from 90th place the previous year. This research enriches the ACF literature by demonstrating that coalition contestation does not stop at the formulation stage but continues dynamically throughout the course of
policy implementation.

Kualitas air merupakan faktor yang mendukung kelangsungan hidup organisme budidaya seperti suhu, Power of Hydrogen (pH), alkalinitas, Total Organic Matter (TOM), Total Dissolved Solid (TDS), dan nitrit. Faktor keberhasilan budidaya ikan lele ini salah satunya yaitu dalam hal manajemen kualitas air yang ramah lingkungan. Tujuan dalam penelitian ini untuk mengetahui manfaat manajemen kualitas air dengan inovasi yang ramah lingkungan yaitu ekoenzim, serta dosis efektif ekoenzim yang digunakan. Metode yang digunakan pada penelitian ini yaitu metode eksperimental pada pemeliharaan ikan lele. Penambahan ekoenzim kurang signifikan dalam memperbaiki kualitas air yang meliputi suhu, Power of Hydrogen (pH), Total Dissolved Solid (TDS), alkalinitas, nitrit, dan Total Organic Matter (TOM). Hasil pengukuran menunjukkan kisaran suhu 23 ± 290 C, Power of Hydrogen (pH) 7.4-9.0, Total Dissolved Solid (TDS) 347- 1042 mg/L, alkalinitas 40-80 mg/L, nitrit 1 mg/L, dan Total Organic Matter (TOM) 170.64 – 2907.2 mg/L. Meskipun ekoenzim tidak berpengaruh dalam memperbaiki kualitas air pada parameter suhu, Total Dissolved Solid (TDS), alkalinitas, nitrit, dan Total Organic Matter (TOM), namun ekoenzim dapat menstabilkan parameter Power of Hydrogen (pH) wadah budidaya. Ekoenzim dosis 10 mL merupakan yang paling efektif dalam menstabilkan kualitas air dan menekan tingkat kematian harian ikan budidaya.Water quality is a factor that supports the survival of cultivated organisms such as temperature, Power of Hydrogen (pH), alkalinity, Total Organic Matter (TOM), Total Dissolved Solid (TDS), and nitrite. One of the factors for the success of catfish cultivation is environmentally friendly water quality management. The purpose of this study was to determine the benefits of water quality management with environmentally friendly innovations, namely ecoenzymes, as well as the effective dosage of ecoenzymes used. The method used in this study was an experimental method in catfish cultivation. The addition of ecoenzymes was less significant in improving water quality which included temperature, Power of Hydrogen (pH), Total Dissolved Solid (TDS), alkalinity, nitrite, and Total Organic Matter (TOM). The measurement results showed a temperature range of 23 ± 290 C, Power of Hydrogen (pH) 7.4-9.0, Total Dissolved Solid (TDS) 347-1042 mg/L, alkalinity 40-80 mg/L, nitrite 1 mg/L, and Total Organic Matter (TOM) 170.64 – 2907.2 mg/L. Although ecoenzyme has no effect in improving water quality on temperature, Total Dissolved Solid (TDS), alkalinity, nitrite, and Total Organic Matter (TOM) parameters, ecoenzyme can stabilize the Power of Hydrogen (pH) parameter of the cultivation container. Ecoenzyme dose of 10 mL is the most effective in stabilizing water quality and reducing the daily mortality rate of cultivated fish.
5090754293K1A022081Aktivitas Antioksidan dan Antiinflamasi Hidrolisat Protein Ikan Gabus (Channa striata) oleh Fraksi Enzim Protease Bacillus subtilis B209Radikal bebas yang berlebihan dapat memicu stres oksidatif dan inflamasi sehingga meningkatkan resiko berbagai penyakit degeneratif. Sumber senyawa bioaktif alami yang berpotensi sebagai antioksidan dan antiinflamasi adalah peptida hasil hidrolisis protein ikan gabus (Channa striata). Ikan gabus merupakan salah satu jenis ikan yang mempunyai kandungan protein yang tinggi, daging ikan gabus mengandung protein sebesar 70%. Penelitian ini bertujuan untuk mendapatkan fraksi protease dengan aktivitas spesifik tertinggi yang digunakan untuk hidrolisis protein ikan gabus. Hidrolisat protein yang dihasilkan kemudian diuji aktivitas antioksidan dan antiinflamasi. Ekstrak kasar enzim protease dari Bacillus subtilis B209 difraksinasi bertingkat menggunakan ammonium sulfat, kemudian fraksi dengan aktivitas spesifik tertinggi dikarakterisasi dan digunakan untuk menghidrolisis protein ikan gabus. Aktivitas antioksidan dianalisis dengan metode DPPH, aktivitas antiinflamasi menggunakan metode penghambatan denaturasi BSA, profil protein dianalisis menggunakan SDS-PAGE, dan uji hemolisis. Hasil penelitian menunjukkan bahwa protease fraksi F15 memiliki aktivitas spesifik tertinggi sebesar 0,186 U/mg dan faktor purifikasi sebesar 9,239 dengan kondisi optimum pada suhu 40 °C dan pH 8. Nilai derajat hidrolisis tertinggi hidrolisat protein diperoleh pada waktu 60 menit sebesar 17,566%. Aktivitas antioksidan tertinggi hidrolisat protein diperoleh pada waktu hidrolisis 10 menit dengan nilai persentase inhibisi sebesar 40,589%. Hidrolisat protein ikan gabus diperoleh nilai IC50 sebesar 3740,9 ppm dan AAI sebesar 0,005 menunjukkan aktivitas antioksidan lemah. Konsentrasi hidrolisat protein yang memiliki aktivitas antiinflmasi berada pada kisaran 0,3 – 0,9 mg/mL dengan persentase inhibisi > 20%. Persentase hemolisis diperoleh sebesar 5,102% menunjukkan bahwa hidrolisat protein ikan gabus lisis terhadap sel darah merah. Hasil analisis SDS-PAGE menunjukkan pita protein membentuk smear.Excessive free radical can trigger oxidative stress and inflammation, thereby increasing the risk of various degenerative diseases. One of the natural bioactive compounds with potential antioxidant and anti-inflammatory properties is peptides derived from the hydrolysis of snakehead fish (Channa striata) protein. Snakehead fish is a freshwater fish species with a high protein content, containing approximately 70% protein in its flesh. This study aimed to obtain the protease fraction with the highest specific activity to be used for the hydrolysis of snakehead fish protein. The resulting protein hydrolysate was subsequently evaluated for its antioxidant and anti-inflammatory activities. Crude protease extract from Bacillus subtilis B209 was fractionated stepwise using ammonium sulfate, and the fraction with the highest specific activity was characterized and subsequently used to hydrolyze snakehead fish protein. Antioxidant activity was determined using the DPPH method, anti-inflammatory activity was evaluated by the bovine serum albumin (BSA) denaturation inhibition assay, protein profile was analyzed by SDS-PAGE, and hemolytic activity was assessed by a hemolysis assay. The results showed that the F15 protease fraction exhibited the highest specific activity of 0.186 U/mg with a purification factor of 9.239 and optimum conditions at 40 °C and pH 8. The highest degree of hydrolysis was obtained after 60 minutes of hydrolysis, reaching 17.566%. The highest antioxidant activity was observed in the hydrolysate produced after 10 minutes of hydrolysis, with a percentage inhibition of 40.589%. The snakehead fish protein hydrolysate exhibited an IC50 value of 3740.9 ppm and an Antioxidant Activity Index (AAI) of 0.005 indicating weak antioxidant activity. The hydrolysate showed anti-inflammatory activity at concentrations ranging from 0.3 to 0.9 mg/mL, with inhibition percentage exceeding 20%. The hemolysis percentage was 5.102%, indicating that the snakehead fish protein hydrolysate caused lysis of red blood cells. SDS-PAGE analysis revealed that the protein bands exhibited a smeared pattern.
5090854322K1A022045Penurunan Kadar BOD, COD, dan TSS Limbah Cair Pembuatan Tahu Menggunakan Proses Elektrokoagulasi Dibantu Biokoagulan Biji PepayaLimbah cair industri tahu mengandung bahan organik, senyawa nitrogen, fosfor, minyak dan lemak, serta padatan tersuspensi yang tinggi sehingga dapat mencemari lingkungan apabila dibuang tanpa pengolahan. Kandungan pencemar tersebut berasal dari proses pengolahan kedelai yang dapat meningkatkan pencemaran. Penelitian ini bertujuan untuk mengetahui pengaruh berat dan ukuran biokoagulan biji pepaya, kecepatan pengadukan, serta lama waktu pengadukan pada proses elektrokoagulasi terhadap penurunan kadar BOD (Biochemical Oxygen Demand), COD (Chemical Oxygen Demand), dan TSS (Total Suspended Solid) limbah cair pembuatan tahu. Penelitian ini menguji pengaruh variasi tersebut menggunakan elektroda aluminium dengan variasi berat biokoagulan 0, 1, 2, dan 3 g, ukuran biokoagulan 60, 80, 100, dan 120 mesh, kecepatan pengadukan 300, 400, 500, dan 600 rpm, serta waktu pengadukan 20, 30, 40, dan 50 menit. Hasil penelitian menunjukkan bahwa kondisi optimum diperoleh pada penggunaan biokoagulan 3 g, ukuran biokoagulan 120 mesh, kecepatan pengadukan 500 rpm, dan waktu pengadukan 50 menit dengan efisiensi penurunan BOD sebesar 87,17%, COD sebesar 86,28%, dan TSS sebesar 97,69%. Kombinasi proses elektrokoagulasi dibantu biokoagulan biji pepaya mampu menurunkan kadar pencemar limbah cair pembuatan tahu dengan persentase penurunan yang tinggi. Industrial wastewater from tofu production contains organic matter, nitrogen compounds, phosphorus, oils and fats, as well as high levels of suspended solids, which can pollute the environment if discharged without treatment. These pollutants originate from the soybean processing, which can exacerbate pollution. This study aims to determine the effects of the weight and size of papaya seed biocoagulant, stirring speed, and stirring duration in the electrocoagulation process on the reduction of BOD (Biochemical Oxygen Demand), COD (Chemical Oxygen Demand), and TSS (Total Suspended Solids) levels in tofu production wastewater. This study examined the effects of these variables using aluminum, with bio-coagulant weights of 0, 1, 2, and 3 gs, bio-coagulant particle sizes of 60, 80, 100, and 120 mesh, stirring speeds of 300, 400, 500, and 600 rpm, and stirring times of 20, 30, 40, and 50 minutes. The results showed that optimal conditions were achieved using 3 gs of biocoagulant, a 120-mesh particle size, a stirring speed of 500 rpm, and a stirring time of 50 minutes, resulting in BOD removal efficiency of 87.17%, COD removal efficiency of 86.28%, and TSS removal efficiency of 97.69%. Pertumbuhan yang cepat tersebut, diduga didukung oleh bakteri endofit yang berperan dalam merangsang pertumbuhan tanaman (plant growh promoting) E. crassipes. Dua sifat penting bakteri endofit yang mendukung pertumbuhan tanaman antara lain penambatan nitrogen dan penghasil hormon pertumbuhan Indole Acetic Acid (IAA). Eksplorasi bakteri endofit penambat nitrogen dan penghasil IAA dari akar E. crassipes yang tumbuh di lingkungan Bendung Gerak Sungai Serayu belum pernah dilakukan. Penelitian ini bertujuan untuk mengisolasi bakteri endofit akar E. crassipes dari lingkungan Bendung Gerak Sungai Serayu yang memiliki kemampuan penambatan nitrogen dan menghasilkan IAA.

Penelitian telah dilakukan selama 5 bulan dimulai pada bulan Februari-Juni 2026 dengan metode survei. Variabel bebas dalam penelitian ini adalah isolat bakteri dari akar eceng gondok, sedangkan variabel terikat meliputi kemampuan bakteri menambat nitrogen dan menghasilkan IAA. Parameter yang diamati meliputi perubahan warna media NFB, konsentrasi amonium, perubahan warna uji IAA, konsentrasi IAA, serta karakteristik isolat bakteri terpilih. Pengambilan sampel dilakukan di lingkungan Sungai Serayu. Isolasi, pengujian kemampuan bakteri dalam menambat nitrogen dan menghasilkan IAA, serta karakterisasi bakteri dilakukan di Laboratorium Mikrobiologi Fakultas Biologi Universitas Jenderal Soedirman. Data aktivitas penambatan nitrogen dan aktivitas produksi IAA dianalisis secara deskriptif. Hasil karakterisasi isolat bakteri mengacu pada Bergey’s Manual of Determinative Bacteriology untuk mengetahui identitas isolat bakteri.

Sebanyak lima isolat mampu menambat nitrogen (isolat EGS04, EGS05, EGS07, EGS09, dan EGS11) berkisar antara 0,808–1,591 ppm, dengan nilai tertinggi ditunjukkan oleh isolat EGS05 sebesar 1,591 ppm. Selain itu, dua isolat mampu menghasilkan IAA (EGS02 dan EGS05) berkisar 26,892-61,363 ppm. Aktivitas penambatan nitrogen dan produksi IAA tertinggi dihasilkan oleh isolat EGS05. Hasil pengujian karakterisasi isolat EGS05 secara fenotipik menunjukkan bahwa isolat EGS05 termasuk dalam genus Bacillus. Penelitian ini membuktikan akar E. crassipes merupakan habitat potensial bagi bakteri endofit yang mampu menambat nitrogen dan menghasilkan IAA.
Water hyacinth (Eichhornia crassipes) is an aquatic weed characterized by its rapid growth rate. This rapid growth is presumed to be supported by endophytic bacteria that function as plant growth-promoting bacteria (PGPB). Two important traits of endophytic bacteria that contribute to plant growth are nitrogen fixation and the production of the phytohormone indole-3-acetic acid (IAA). The exploration of nitrogen-fixing and IAA-producing endophytic bacteria from the roots of E. crassipes growing in the Serayu River Barrage area has not previously been reported. Therefore, this study aimed to isolate endophytic bacteria from the roots of E. crassipes collected from the Serayu River Barrage area and to observe their ability to fix nitrogen and produce IAA.
This study was conducted over a five-month period, from February to June 2026, using a survey method. The independent variable was the bacterial isolates obtained from the roots of water hyacinth, while the dependent variables were the ability of the isolates to fix nitrogen and produce IAA. The observed parameters included changes in the color of the nitrogen-free bromothymol blue (NFB) medium, ammonium concentration, color changes in the IAA assay, IAA concentration, and the characteristics of the selected bacterial isolate. Root samples of E. crassipes were collected from the Serayu River. Isolation of endophytic bacteria, observation of their nitrogen-fixing and IAA-producing abilities, and bacterial characterization were carried out at the Microbiology Laboratory, Faculty of Biology, Universitas Jenderal Soedirman. Data on nitrogen-fixing activity and IAA production were analyzed descriptively. The characterization results were compared with the criteria described in Bergey's Manual of Determinative Bacteriology to determine the identity of the bacterial isolates.
Five isolates (isolates EGS04, EGS05, EGS07, EGS09, and EGS11) demonstrated nitrogen-fixing ability, with concentrations ranging from 0.808-1.591 ppm, and the highest value was observed in isolate EGS05 (1.591 ppm). In addition, two isolates (EGS02 and EGS05) were able to produce IAA, with concentrations ranging from 26.892-61.363 ppm. The highest nitrogen-fixing activity and IAA production were exhibited by isolate EGS05. Phenotypic characterization of isolate EGS05 indicated that it belongs to the genus Bacillus. This study demonstrates that the roots of E. crassipes constitute a potential habitat for endophytic bacteria capable of nitrogen fixation and IAA production.

ABSTRAK
Restless Leg Syndrome (RLS) merupakan masalah serius yang sering dialami oleh pasien Hemodialisis (HD). Beberapa terapi non farmakologi dapat dilakukan untuk mengurangi gejala RLS yaitu latihan peregangan, aerobic, terapi cool dialysate, Terapi Aktifitas Fisik (TAF), refleksiologi, dan pijat aromaterapi. Literature review ini bertujuan untuk mengetahui pengaruh Intervensi Physical Exercise(IPE) dalam mengurangi gejala RLS pada pasien yang menjalani HD meliputi jenis gerakan, durasi latihan serta frekuensi. Data dikumpulkan dari database elektronik PUBMED, Science Direct, Proquest, Ebscohost, CNBI dan e-resources dengan jumlah total 522 artikel. Artikel dibatasi pada kriteria berbahasa Inggris dan desain eksperimen dalam rentang waktu 10 tahun terakhir. Berdasarkan 7 artikel yang terpilih didapatkan bahwa IPE yang dapat diterapkan pada pasien dengan RLS antara lain latihan peregangan dan latihan aerobic. Sementara itu untuk durasi IPE yang paling tepat adalah selama 30-40 menit dan frekuensi IPE yang paling seuai adalah 3.kali per minggu. Instrument yang paling banyak dipakai untuk menilai derajat RLS adalah International Restless Legs Syndrome Severity Scale (IRLS). TAK efektif untuk mengurangi RLS pada pasien Hemodialisis. Faktor yang paling berperan dalam efektifitas IPE meliputi jenis gerakan, durasi latihan dan frekuensi latihan yang dilakukan.
ABSTRACT
Restless Leg Syndrome (RLS) is a serious problem that is often experienced by Hemodialysis (HD) patients. Some nonpharmacological therapies can be done to reduce RLS symptoms such as stretching exercises, aerobics, cool dialysate therapy, Physical Activity Therapy (PAT) , reflexology, and aromatherapy massage. This literature review aims to determine the effect of Physical Exercise Intervention (PEI) in reducing RLS symptoms in patients undergoing HD including the type of movement, exercise duration, and frequency. Data were collected from PUBMED, Science Direct, Proquest, Ebscohost, CNBI, and e-resources with a total of 522 articles. These articles were limited to the criteria of English language and experimental design within the last 10 years. Physical Exercise Intervention could be applied with RLS including stretching exercises and aerobic exercises based on seven selected articles. Meanwhile, the most appropriate PEI duration was 30-40 minutes and the most appropriate PEI frequency was three times per week. The most widely used instrument to assess the degree of RLS was the international Restless Legs Syndrome Severity Scale (IRLS). PEI is effective in reducing RLS of hemodialysis patients. The most factors contribute to the effectiveness of PEI are the type of movement, the duration of exercises, and the frequency of exercises performed.
