Abstrak (Bhs. Indonesia)Aeromonas hydrophila merupakan bakteri patogen penyebab Motile Aeromonas
Septicemia (MAS) pada ikan air tawar yang pengendaliannya masih banyak
menggunakan antibiotik. Penggunaan antibiotik secara berlebihan berpotensi
menyebabkan resistensi bakteri sehingga diperlukan alternatif antibakteri alami,
salah satunya eco-enzyme. Penelitian ini bertujuan mengetahui kemampuan ecoenzyme berbahan dasar campuran limbah buah jeruk dan buah pepaya dalam
menghambat pertumbuhan A. hydrophila serta mengetahui konsentrasi yang
memberikan daya hambat terhadap bakteri tersebut. Penelitian menggunakan
Rancangan Acak Lengkap (RAL) dengan empat konsentrasi eco-enzyme, yaitu
25%, 50%, 75%, dan 100%, kontrol positif amoxicillin, serta kontrol negatif
akuades asam, masing-masing tiga ulangan. Parameter yang diamati meliputi
karakteristik eco-enzyme, kandungan senyawa aktif, dan aktivitas antibakteri
menggunakan metode difusi sumuran. Data dianalisis menggunakan uji
Kruskal–Wallis yang dilanjutkan dengan uji Dunn–Bonferroni. Eco-enzyme
memiliki karakteristik suhu 25–30°C, pH 2,9–6,5, dan TDS 2.450–4.450 ppm serta
mengandung tanin, flavonoid, dan steroid. Zona hambat terbentuk pada
konsentrasi 50%, 75%, dan 100% dengan diameter berturut-turut 3,50 ± 0,50 mm,
5,33 ± 0,577 mm, dan 6,75 ± 1,392 mm. Hasil uji Kruskal–Wallis menunjukkan
perbedaan signifikan (p = 0,038), sedangkan uji Dunn–Bonferroni menunjukkan
perbedaan nyata antara konsentrasi 50% dan 100% (p = 0,033). Eco-enzyme
campuran limbah jeruk dan pepaya memiliki aktivitas antibakteri terhadap A.
hydrophila, tetapi belum dapat ditetapkan satu konsentrasi sebagai konsentrasi
optimal berdasarkan hasil uji statistik.
Abtrak (Bhs. Inggris)Aeromonas hydrophila is a pathogenic bacterium that causes Motile Aeromonas
Septicemia (MAS) in freshwater fish, which is commonly controlled using antibiotics.
Excessive use of antibiotics may lead to bacterial resistance, creating a need for natural
antibacterial alternatives, such as eco-enzyme. This study aimed to determine the ability
of eco-enzyme produced from mixed orange and papaya fruit waste to inhibit the growth
of A. hydrophila and to determine the concentrations that exhibited antibacterial activity
against the bacterium. A Completely Randomized Design (CRD) was applied using four
eco-enzyme concentrations, namely 25%, 50%, 75%, and 100%, a positive control of
amoxicillin, and a negative control of acidified distilled water, with three replications
each. The observed parameters included eco-enzyme characteristics, active compounds,
and antibacterial activity using the agar well diffusion method. Data were analyzed using
the Kruskal–Wallis test followed by the Dunn–Bonferroni test. The eco-enzyme had
temperatures of 25–30°C, pH values of 2.9–6.5, and Total Dissolved Solids (TDS) of
2,450–4,450 ppm, and contained tannins, flavonoids, and steroids. Inhibition zones were
observed at concentrations of 50%, 75%, and 100%, with diameters of 3.50 ± 0.50 mm,
5.33 ± 0.577 mm, and 6.75 ± 1.392 mm, respectively. The Kruskal–Wallis test showed a
significant difference (p = 0.038), while the Dunn–Bonferroni test showed a significant
difference between the 50% and 100% concentrations (p = 0.033). Eco-enzyme produced
from mixed orange and papaya fruit waste exhibited antibacterial activity against A.
hydrophila, but a single optimal concentration could not be determined based on the
statistical analysis.
