Abstrak (Bhs. Indonesia)Jerawat merupakan salah satu masalah kulit yang umum dengan penyebab
utama infeksi bakteri patogen seperti S. aureus dan P. acnes. Resistensi antibiotik
sintetik yang semakin meningkat mendorong pencarian alternatif antibakteri
berbasis bahan alam. Penelitian ini bertujuan untuk mengisolasi senyawa bioaktif
dari ekstrak etil asetat rimpang kencur hitam (K. parviflora) serta menguji aktivitas
antibakterinya terhadap S. aureus dan P. acnes melalui metode difusi sumuran.
Tahapan penelitian meliputi ekstraksi dengan cara maserasi serbuk rimpang kencur
hitam secara bertahap menggunakan pelarut n-heksana dilanjutkan dengan etil
asetat, kromatografi cair vakum (KCV) menghasilkan 3 fraksi gabungan,
pemisahan lebih lanjut terhadap fraksi aktif melalui metode kromatografi radial
(kromatotron) menghasilkan 4 isolat gabungan. Isolat IG4 kemudian di murnikan
lebih lanjut menggunakan kromatotron menghasilkan 6 isolat gabungan. Isolat aktif
(IG4.1 dan IG4.3) kemudian diidentifikasi menggunakan Liquid Chromatography–
High Resolution Mass Spectrometry (LC-HRMS). Hasil identifikasi menunjukkan
bahwa senyawa utama dari isolat IG4.1 adalah senyawa 5,7,4'-trimetoksiflavon
(C18H16O5) dengan berat molekul 312, sedangkan senyawa utama isolat IG4.3
terdiri atas 5,6,7-trimetoksi-2-(4-metoksifenil) kromen-4-on (C19H18O6) dengan
berat molekul 342 dan 5,7-dimetoksiflavon (C17H14O4) dengan berat molekul 282.
Uji antibakteri dilakukan terhadap ekstrak, fraksi, dan isolat. Hasil uji diperoleh
diameter zona hambat (mm) untuk ekstrak etil asetat, fraksi FG2, FG3, isolat IG4.1,
dan IG4.3 berturut-turut sebesar 3,90; 0,00; 3,41; 0,25; 0,00 terhadap S. aureus dan
4,54; 0,00; 2,06; 0,61; 0,00 terhadap P. acnes.
Abtrak (Bhs. Inggris)Acne is a common skin condition primarily caused by pathogenic bacterial
infections such as S. aureus and P. acnes. The growing resistance to synthetic
antibiotics has spurred the search for natural antibacterial alternatives. This study
aims to isolate bioactive compounds from the ethyl acetate extract of black galangal
rhizome (K. parviflora) and to test their antibacterial activity against S. aureus and
P. acnes using the well diffusion method. The research stages included extraction
by gradually macerating black galangal rhizome powder using n-hexane followed
by ethyl acetate, yielding a yellowish-brown solid, which was then fractionated via
vacuum liquid chromatography (VLC) to produce 3 combined fractions. Further
separation of the active fractions via radial chromatography (Chromatotron)
yielded 4 combined isolates. Isolate IG4 was then further purified via
chromatotron, yielding 6 combined isolates. The active isolates (IG4.1 and IG4.3)
were subsequently identified using Liquid Chromatography–High Resolution Mass
Spectrometry (LC-HRMS). The identification results showed that the main
compound of isolate IG4.1 was 5,7,4'-trimethoxyflavone (C18H16O5) with a
molecular weight of 312, while the major compounds of isolate IG4.3 consist of
5,6,7-trimethoxy-2-(4-methoxyphenyl)chromen-4-one (C19H18O6) with a molecular
weight of 342 and 5,7-dimethoxyflavone (C17H14O4) with a molecular weight of 282.
Antibacterial tests were conducted on extracts, fractions, and isolates. The test
results yielded inhibition zone diameters (mm) for the ethyl acetate extract,
fractions FG2 and FG3, and isolates IG4.1 and IG4.3 of 3.90; 0.00; 3.41; 0.25;
0.00 against S. aureus and 4.54; 0.00; 2.06; 0.61; 0.00 against P. acnes.
