Optimasi Suhu dan Waktu Pemanasan Transesterifikasi Pada Pembuatan Medium Chain Tryglyceride Oil Dari Virgin Coconut Oil Menggunakan Response Surface Methodology

Virgin Coconut Oil (VCO) mengandung asam lemak rantai sedang atau Medium-Chain Triglycerides (MCT) yang berpotensi dikembangkan menjadi minyak MCT bernilai ekonomi tinggi. Namun, kandungan MCT dalam VCO masih bercampur dengan asam lemak rantai panjang dan senyawa lainnya sehingga diperlukan beberapa tahap pengolahan, salah satunya transesterifikasi. Tahap ini dipengaruhi oleh suhu dan waktu pemanasan, karena kondisi yang tidak sesuai dapat menurunkan rendemen akibat terjadinya reaksi samping, seperti saponifikasi dan reaksi balik pembentukan trigliserida. Penelitian ini bertujuan mengoptimasi suhu dan waktu pemanasan pada tahap transesterifikasi dalam pembuatan minyak MCT dari VCO, menganalisis pengaruh kedua faktor terhadap rendemen, serta menentukan karakteristik kimia produk yang dihasilkan. Proses penelitian meliputi netralisasi VCO, transesterifikasi VCO netral, isolasi medium-chain fatty acids (MCFAs), dan sintesis MCT. Optimasi dilakukan menggunakan Response Surface Methodology (RSM) dengan rancangan Central Composite Design (CCD) melalui perangkat lunak Design Expert versi 13. Hasil optimasi menunjukkan kondisi optimum transesterifikasi pada suhu 51,667°C selama 2,244 jam dengan prediksi rendemen sebesar 82,347%. Suhu dan waktu pemanasan menunjukkan interaksi berbanding terbalik, yaitu suhu yang lebih rendah memerlukan waktu pemanasan lebih lama, sedangkan suhu yang lebih tinggi memerlukan waktu lebih singkat. Kondisi suhu maupun waktu yang terlalu rendah atau terlalu tinggi menurunkan rendemen. Minyak MCT hasil sintesis memiliki bilangan asam 0,34 mg KOH/g, bilangan peroksida 0,333 meq O₂/kg, dan bilangan iod 0,697 g I₂/100 g. Komposisi asam lemak produk masih didominasi oleh asam laurat dan asam miristat.

Virgin Coconut Oil (VCO) contains medium-chain fatty acids, or Medium-Chain Triglycerides (MCTs), which have the potential to be developed into high-value MCT oil. However, the MCT content in VCO is still mixed with long-chain fatty acids and other compounds, requiring several processing steps, one of which is transesterification. This stage is influenced by heating temperature and time, as inappropriate conditions can reduce yield due to side reactions, such as saponification and the reverse reaction leading to triglyceride formation. This study aims to optimize the heating temperature and time during the transesterification stage in the production of MCT oil from VCO, analyze the effects of these two factors on yield, and determine the chemical characteristics of the resulting product. The research process included the neutralization of VCO, the transesterification of neutralized VCO, the isolation of medium-chain fatty acids (MCFAs), and the synthesis of MCT. Optimization was performed using Response Surface Methodology (RSM) with a Central Composite Design (CCD) via Design Expert software version 13. The optimization results indicate that the optimal conditions for transesterification are a temperature of 51.667°C for 2.244 hours, with a predicted yield of 82.347%. Temperature and heating time exhibit an inverse relationship: lower temperatures require longer heating times, while higher temperatures require shorter heating times. Temperatures or heating times that are too low or too high reduce the yield. The synthesized MCT oil has an acid value of 0.34 mg KOH/g, a peroxide value of 0.333 meq O₂/kg, and an iodine value of 0.697 g I₂/100 g. The fatty acid composition of the product is still dominated by lauric acid and myristic acid.
