| Abstrak (Bhs. Indonesia) | Tujuan penelitian ini adalah mengetahui efek pemberian testosteron terhadap motilitas dan abnormalitas spermatozoa, serta interaksi antara pemberian testosteron, waktu penyimpanan dan umur terhadap motilitas dan abnormalitas Sapi Madura pada suhu 5o C. Penelitian dilakukan Laboratorium Balai Besar Inseminasi Buatan, Singosari, Malang. Materi penelitian menggunakan semen sapi Madura grade C umur ≤ 5 tahun sampai ≥ 5 tahun, dengan bahan pengencer tris kuning telur ditambah testosteron. Penelitian menggunakan metode eksperimental laboratorium dengan rancangan penelitian Rancangan Acak Kelompok (RAK) pola factorial. Perlakuan terdiri atas blok (penyadapan 2 kali), umur (U1 & U2), level testosteron dalam pengencer (P0= kontrol, P1= 750 µg testosteron , P2= 500 µg testosteron, dan P3= 250 µg testosteron), dan lama penyimpanan (H1, H2, H3, H4, H5 & H6). Data dianalisis variansi dan dilanjutkan dengan uji beda nyata jujur (BNJ). Hasil analisis variansi menunjukan bahwa blok tidak berpengaruh nyata (P>0.05) terhadap motilitas spermatozoa. Perlakuan menunjukan pengaruh sangat nyata (P<0.01) terhadap motilitas spermatozoa. Umur berpengaruh nyata terhadap motilitas spermatozoa sapi Madura (P>0.05), berdasarkan hasil uji Beda Nyata Jujur (BNJ) U2 memiliki tingkat motilitas yang lebih tinggi daripada U1 yaitu U2 29.0 % dan U1 27.9 %. Pemberian hormon testosteron berpengaruh nyata terhadap abnormalitas spermatozoa sapi Madura (P < 0.05) hasil uji lanjut Beda Nyata Jujur (BNJ) P0 13,133 %, P1 14,655 %, P2 12,661 %, dan P3 14,173 %. Kesimpulan perlakuan terbaik pada penambahan testosteron dengan konsentrasi 500 µg terhadap motilitas dan abnormalitas spermatozoa sapi Madura. |

Inggris) | The purposes are the research was to determine the effect of testosterone on sperm motility and abnormalities, as well as the interaction between the added testosterone, storage time and age on motility and abnormalities of Madura Cattle at a temperature 5 ° C. The research was conducted at the Laboratory Center of Artificial Insemination, Singosari, Malang. The material of reasearch are grade C semen of ≤ 5 and ≥ 5 years old Madura bull. Randomized block design (RBD) with factorial methods is used in this experiment . Treatments consisted of P0, P1, P2, and P3, age of bull (U1 & U2), storage durations (H1, H2, H3, H4, H5 & H6) and as block twice tapping. Analysis of variance is used to evaluate reasearch date followed by a test of honestly significant difference (HSD). Results of analysis of variance showed that the block has no significanse effect (P>0.05) on the motility of spermatozoa. The testosteron treatments showed significant effect (P<0.01) on sperm motility, as well as age of Madura bull (P< 0.05). Based on Honestly Significant Difference test (HSD) showed that U2 (29.0% ) has a higher motility rate compared to U1 (27.9%). The addition of testosterone affects significantly on sperm’s abnormalities of Madura bull (P <0.05); the further results of Honestly Significant Difference (HSD) test showed that P0=13.133%, P1 = 14.655%, P2 = 12.661%, and P3 = 14.173%. it could be concluded that the best addition of testosteron in diluent to Madura bull sperm motlity and abnormality is 500 µg. |
