Abstrak (Bhs. Indonesia)Captopril merupakan inhibitor Angiotensin Converting Enzyme (ACE) yang memiliki waktu paruh eliminasi yang cepat berkisar 2-3 jam. Namun absorbsinya dapat menurun 30-50% dengan adanya makanan. Penelitian ini bertujuan memformulasikan patch transdermal captopril menggunakan polietilen glikol 400, asam oleat, dan isopropil alkohol dalam polimer HPMC dan eudragit RL100 yang diharapkan dapat menghasilkan karakteristik fisik dan formula optimum yang baik.
Patch transdermal captopril dirancang dengan metode simplex lattice design (SLD) dengan bantuan software Design Expert v.7. Evaluasi karakteristik meliputi uji visual, ketebalan, bobot, susut pengeringan, daya serap kelembaban, dan ketahanan pelipatan sediaan patch. Penentuan formula optimum menggunakan software Design Expert v.7 dan diverifikasi menggunakan software OpenStat v.2012.
Karakteristik fisik formula optimum patch transdermal captopril menghasilkan kesan visual yang baik, ketebalan 0,54 mm, bobot 272 mg, presentase susut pengeringan 4,34 %, daya serap kelembaban 4,35 %, dan ketahanan terhadap pelipatan >300 kali. Proporsi formula optimum patch transdermal captopril adalah polietilen glikol 400 1,4%, asam oleat dan isopropil alkohol masing-masing 1%. Kandungan zat aktif formula optimum adalah 95,22%.
Abtrak (Bhs. Inggris)Captopril is an angiotensin-converting enzyme inhibitors (ACE), which has a rapid elimination half-life ranges from 2-3 hours. However, the absorbtion may decline 30-50% in the presence of food. This study aims to formulate a transdermal patch captopril using polyethylene glycol 400, oleic acid, and isopropyl alcohol in the polymer HPMC and Eudragit RL100 is expected to results a good physical characteristics and a optimum formula.
Simplex Lattice Design (SLD) method designs the transdermal patch of captopril using Design Expert v.7 software. The evaluation includes visual characteristics, thickness, weight, loss on drying, moisture uptake, and folding endurance. Determination of the optimum formula using Design Expert v.7 software and verified by OpenStat v.2012 software.
Physical characteristics of the optimum formula transdermal patch of captopril results a good visual impression, thickness is 0.54 mm, weights is 272 mg, loss on drying is 4.34%, 4.35% of moisture uptake, and folding endurance >300 times. The proportion of the optimum formula transdermal patch of captopril is 14% polyethylene glycol 400, oleic acid and isopropyl alcohol 1%, respectively. The drug content of optimum formula is 95.224%.
