| Abstrak (Bhs. Indonesia) | Captopril, obat golongan Angiotensin Converting Enzyme Inhibitor memiliki waktu paruh eliminasi 1,6-1,9 jam, adanya makanan menurunkan absorbsi menjadi 30-50%. Formulasi patch transdermal captopril menggunakan polietilen glikol, propilen glikol, dan isopropil alkohol dalam polimer HPMC dan eudragit RL100 diharapkan dapat menjadi alternatif sistem penghantaran captopril dengan bioavailabilitas baik. Tujuan penelitian ini adalah mengetahui karakteristik fisik dan formula optimum patch transdermal captopril. Patch transdermal captopril dibuat dengan mengkombinasikan polietilen glikol, propilen glikol, dan isopropil alkohol yang dirancang berdasarkan simplex lattice design. Evaluasi karakteristik fisik meliputi visual, ketebalan, bobot, susut pengeringan, daya serap kelembaban, dan ketahanan terhadap pelipatan patch transdermal. Formula optimum ditentukan menggunakan software Design Expert v.7, diverifikasi menggunakan software OpenStat v.2012 dan diuji drug content. Karakteristik fisik patch transdermal captopril dengan plasticizer polietilen glikol, enhancer propilen glikol, dan cosolvent isopropil alkohol menghasilkan patch dengan tampilan visual baik, bobot 122-268 mg, ketebalan 0,20-0,63 mm, susut pengeringan 2,59-18,20%, daya serap kelembaban 0-9,27%, dan ketahanan terhadap pelipatan 52-300 kali. Proporsi formula optimum yaitu 5% polietilen glikol, 1% propilen glikol, dan 10% isopropil alkohol dengan karakteristik visual baik, bobot 147,67 mg, ketebalan 0,25 mm, persentase susut pengeringan 7,56%, daya serap kelembaban 5,35%, dan ketahanan terhadap pelipatan lebih dari 300 kali. Kandungan obat formula optimum 100,62%. |
