| Abstrak (Bhs. Indonesia) | Penelitian ini bertujuan untuk 1) mengidentifikasi R. solanacearum asal tomat dataran rendah dan 2) menguji kisaran inang R. solanacearum asal tomat dataran rendah. Penelitian ini dilaksanakan di Laboratorium Perlindungan Tanaman, Fakultas Pertanian, Universitas Jenderal Soedirman, dan di rumah plastik. Penelitian dilakukan dalam dua tahap, tahap 1 identifikasi R. solanacearum asal tomat dataran rendah, tahap 2 uji kisaran inang menggunakan Rancangan Acak Lengkap dengan 3 jenis tanaman (tomat, terung, cabai merah), masing-masing perlakuan ditanam dalam bak plastik (12 tanaman) dan diulang sebanyak 8 kali. Hasil penelitian menunjukkan bahwa R. solanacearum memiliki beberapa ciri yaitu, aliran masa bakteri berwarna putih keruh, pada medium CPG-TTC koloni bakteri berbentuk tidak beraturan, berlendir, dan berwarna putih susu dengan warna merah di tengahnya, sedangkan pada medium YPGA koloni bakteri berbentuk tidak beraturan, berwarna putih susu dan fluidal. Hasil uji Gram dan katalase, menunjukkan bahwa bakteri R. solanacearum merupakan bakteri Gram negatif dan katalase positif, hasil uji hipersensitivitas pada daun tembakau dan cocor bebek menunjukkan bahwa bakteri bersifat virulen. Pada uji kisaran inang tanaman yang menunjukkan gejala layu adalah tomat dan terung, masa inkubasi masing-masing 17 hari dan 24 hari, intensitas penyakit 50% dan 0,01% dengan laju infeksi 0,0771 unit/hari dan 0,0092 unit/hari, dan nilai AUDPC (Area Under Disease Progress Curve) 282,5% dan 0,005%. |
| Abtrak (Bhs. Inggris) | This aims of the research were 1) identify of R. solanacearum from lowlands tomato and 2) test the host range of R. solanacearum from lowlands tomato. This research was conducted at the Laboratory of Plant Protection, Faculty of Agriculture, Jenderal Soedirman University, and in plastic house. The study was conducted in two phases, the first phase was identification of R. solanacearum from lowland tomato, second phase is to test the host range using completely randomized design with 3 types of plants (tomato, eggplant and red pepper), each were planted in tray (12 plants) and repeated 8 times. The results showed that R. solanacearum has some characteristics of bacterial mass flow cloudy white, the medium of CPG-TTC the colonies shaped of bacteria is irregular, fluidal, and has white milk colour with red in the middle, while the bacterial colonies on the medium YPGA shaped irregular, has color white milk and fluidal. Gram and catalase test results of R. solanacearum showed that R. solanacearum is a negative Gram bacteria and positive catalase, the result of hypersensitivity test on tobacco leaves and cocor bebek (Kalanchoe pinnata) showed that bacteria are virulent. In host range tests showed symptoms of wilting plants are tomato and eggplant, the incubation period each 17 days and 24 days, the intensity of diseases 50% and 0,01% with an infection rate of 0,0771unit/day and 0,0092 unit/day, and AUDPC (Area Under Disease Progress Curve) value 282,5% and 0,005%. |
