Indonesia) | Tumbuhan benalu teh (Scurrula oortiana) merupakan salah satu jenis tumbuhan obat (herba medicina) yang terbilang sulit dan memiliki beberapa kendala dalam pemanfaatannya secara langsung sehingga diperlukan pendekatan lain melalui pemanfaatan kapang endofit yang hidup di dalam jaringan tubuh tumbuhan benalu teh. Penelitian ini bertujuan untuk mengkaji keragaman kapang endofit pada tumbuhan benalu teh, mengkaji kemampuan kapang endofit tumbuhan benalu teh dalam menghasilkan senyawa antimikroba, mengkaji keefektifan senyawa antimikroba yang dihasilkan oleh ekstrak kapang endofit tumbuhan benalu teh dalam menekan pertumbuhan bakteri MRSA secara in vitro, dan mengkaji golongan senyawa antimikroba yang dihasilkan oleh ekstrak kapang endofit tumbuhan benalu teh. Metode yang digunakan dalam penelitian ini adalah metode eksperimental laboratorium. Hasil penelitian menunjukkan sebanyak 17 isolat kapang endofit telah berhasil diisolasi dari organ akar, batang, dan daun tumbuhan S. oortiana, diketahui 5 isolat yaitu isolat BK-S1 (A) (Zopfiella curvata), BK-S2 (B) (Curvularia lunata), BR-S1 (A) (Helicodendron sp.), BR-L (A) (Humicola fuscoatra), dan BR-L (C) (Basidiomycetes) memiliki kemampuan dalam menghasilkan senyawa antimikroba terhadap bakteri MRSA, senyawa antimikroba dari ekstrak etil asetat kapang endofit isolat BR-S1 (A) paling efektif dalam menekan pertumbuhan bakteri MRSA secara in vitro, dan senyawa antimikroba dari ekstrak etil asetat kapang endofit isolat BR-S1 (A) termasuk dalam golongan flavonoid dan tanin. |
| Abtrak (Bhs. Inggris) | Parasite plant of tea is one kind of plant which has been explored and exploited as raw material source of drugs or medication. However, this effort is dificult due to the limitation of the source plant. To extract the active substances from the plant, it takes large amounts of biomass. In this regard, an efford should be found by the use of endophytic fungi from parasite plat which hipothetically has influence to active compounds production, such as antimicrobe compound to various pathogenic bacteria such as Methicillin-Resistant Staphylococcus aureus (MRSA). This study aimed to assess 1). diversity of endophytic fungi on tea parasite plant, 2). ability of the endophytic fungi to produce antimicrobial compounds, 3). effectiveness of the antimicrobial compounds in suppressing the growth of MRSA bacteria In Vitro, and 4). class of antimicrobial compounds produced by the endophytic fungi. This research was conducted experimentally. Samples of tea plant parasite obtained from the Purwodadi, Cendana village, Kutasari Purbalingga. The samples were identified and determined, the endophytic fungi then waere isolated from the organs of roots, stems and leaves of the tea plant parasite. The the pure isolates were then characterized and identified to genus / species and preserved into two kinds of stocks i.e. labor and stock (preserved) cultures. The following steps were micro-scale cultivation and initial selection of endophytic fungi's ability to produce antimicrobial compounds using a crude extract of endophytic fungi and choose the best one to do cultivation in macro-scale, extracted using 4 kinds of solvents different, n-hexane, ethyl acetate, 96% ethanol, and methanol, and tested the ability of each extract in suppressing the growth of MRSA bacteria using the agar diffusion method. Each extract which contained active compounds were also tested both qualitatively and quantitatively. The results showed that the samples of tea plant parasite belong to the species of Scurrula oortiana (Korth.) Dans. Isolation results obtained 17 isolates of endophytic fungi consisting of 4 isolates on root, 9 isolates on stem, and 4 isolates in leaf organs respectively. Initial selection of endophytic fungi's ability to produce antimicrobial compounds showed that of the 17 isolates isolated, only 5 isolates were indicated the ability to produce antimicrobial compounds which were BK-S1 (A) (Zopfiella curvata), BK-S2 (B) (Curvularia lunata), BR-S1 (A) (Helicodendron sp.), BR-L (A) (Humicola fuscoatra), and BR-L (C) (Basidiomycetes). Extracts of endophytic fungi isolates BR-S1 (A) had a various ability to suppress the growth of MRSA bacteria in vitro and most effective extract was an extract of ethyl acetate followed by methanol and ethanol extracts, while the n-hexane extract was not effective at all. Test results are qualitatively showed that the four kinds of extracts of endophytic fungi isolates BR-S1 (A) did not contain alkaloids. In addition, flavonoids was found in ethyl acetate extracts only, tannins was found in ethyl acetate and ethanol extracts of 96 %, and saponins was found in the methanol extract only. Quantitative test showed that total flavonoids of antimicrobial compounds from the ethyl acetate extract of endophytic fungi isolates BR-S1 (A) belonged to the class of flavonoids or tannins. Conclusions could be drawn from this study i.e. 1). endophytic fungi obtained 17 isolates of S. oortiana plant consisting of four isolates on root organ, the organ stem 9 isolates, and 4 isolates in leaf organs, 2). Out of the 17 isolates of endophytic fungi were obtained, only 5 isolates of endophytic fungi were able to produce antimicrobial compounds that isolate BK-S1 (A) (Zopfiella curvata), BK-S2 (B) (Curvularia lunata), BR-S1 (A) (Helicodendron sp.), BR-L (A) (Humicola fuscoatra), and BR-L (C) (Basidiomycetes), 3). Antimicrobial compounds from the ethyl acetate extract of endophytic fungi BR-S1 (A) the most effective in suppressing the growth of MRSA bacteria in vitro, 4). Antimicrobial compounds from the ethyl acetate extract of endophytic fungi BR-S1 (A) included in the class of flavonoids or tannins. |
