Abstrak (Bhs. Indonesia)Asam mefenamat merupakan golongan NSAID yang masuk ke dalam sistem BCS kelas II. Kelarutan asam mefenamat dalam air yang sangat kecil menimbulkan masalah pada laju disolusi obat yang merupakan tahap penentu pada proses absorpsi obat. Tujuan penelitian ini adalah untuk mengoptimasi formula tablet asam mefenamat dengan kombinasi eksipien karagenan, crospovidon, dan, polisorbat 80, sehingga akan menghasilkan suatu formula optimum yang memenuhi syarat sifat fisik dan disolusi tablet.
Metode penelitian adalah eksperimental laboratorium, dengan membuat 7 formula tablet asam mefenamat menggunakan kombinasi karagenan (A), crospovidon (B), dan polisorbat 80 (C), Formula I (100% A); Formula II (100% B); Formula III (100% C); Formula IV (50% A+50% B); Formula V (50% A+50% C); Formula VI (50% B+50% C); Formula VII (33,3% A+33,3% B+33,3% C). Keseragaman bobot, kekerasan, kerapuhan, waktu hancur, dan uji disolusi digunakan sebagai parameter optimasi. Analisis data dilakukan melalui pendekatan teoritis dengan membandingkan syarat-syarat yang terdapat dalam Famakope Indonesia III dan IV serta USP 32. Pemilihan formula optimum ditentukan dengan metode simplex lattice design melalui diagram desirability.
Berdasarkan diagram desirability diperoleh formula optimum teoritis dengan proporsi masing-masing bahan sebagai berikut: 44,06 % karagenan, 34,29 % crospovidon dan 21,65 % polisorbat 80, ditunjukkan dengan nilai desirability yang paling tinggi yaitu 1,000.
Abtrak (Bhs. Inggris)Mefenamic acid is a class of NSAID that include the system BCS class II. Mefenamic acid solubility in water is very small cause problems on the dissolution rate of drugs is a critical step in the process of absorption of the drug. The purpose of this study is to optimize formula of mefenamic acid tablet with a combination of excipients carrageenan, crospovidone, and polysorbate 80, so will result in an optimum formula qualifying physical properties and dissolution of the tablet.
The research method is experimental laboratory, by making 7 tablets mefenamic acid formula uses a combination of carrageenan (A), crospovidone (B), and polysorbate 80 (C), Formula I (100% A); Formula II (100% B); Formula III (100% C); Formula IV (50% A+50% B), Formula V (50% A+50% C); Formula VI (50% B+50% C); Formula VII (33.3% A+33.3% B+33,3% C). Weight uniformity, hardness, friability, disintegration time and dissolution test is used as a parameter optimization. Data analysis was performed by comparing the theoretical approaches with the terms contained in the Indonesian Pharmacopeia III and IV with USP 32. Selection of the optimum formula was determined by the method of simplex lattice design with diagrams desirability.
Based on the desirability diagram obtained with the optimum formula theoretical proportion of each of the following ingredients: 44,06 % carrageenan, 34,29 % crospovidon, and 21,65 % polysorbate 80, indicated by the highest desirability value is 1,000.
