| Abstrak (Bhs. Indonesia) | Buah Talok (Muntingia calabura Linn) merupakan tumbuhan tropis yang banyak ditemui di Indonesia. Tumbuhan ini diduga memiliki khasiat imunomodulator karena mengandung saponin dan flavonoid. Penelitian ini bertujuan mengetahui fungsi imunomodulator ekstrak buah Talok terhadap respon imun tikus Wistar jantan yang diinduksi A. actinomycetemcomitans serta dosis yang efektif dalam peningkatan aktivitas fagositosis makrofag tikus Wistar jantan yang diinduksi A. actinomycetemcomitans. Penelitian ini adalah penelitian eksperimental laboratorium. Buah Talok diekstraksi menggunakan metode maserasi dengan pelarut etanol 70%. Filtrat yang diperoleh dipekatkan hingga diperoleh ekstrak kental kemudian dibuat larutan uji dengan empat variasi konsentrasi. Lima kelompok tikus Wistar jantan yang digunakan yaitu kelompok kontrol diberi aquadest, kelompok perlakuan 1 diberi ekstrak 0,01%, kelompok perlakuan 2 diberi ekstrak 0,1%, kelompok perlakuan 3 diberi ekstrak 1%, dan kelompok perlakuan 4 diberi ekstrak 10%. Data yang diperoleh dianalisis dengan one-way ANOVA dan dilanjutkan dengan Post Hoc test-Duncan dengan taraf kepercayaan 95%. Hasil penelitian menunjukkan bahwa aktivitas fagositosis makrofag terus meningkat pada konsentrasi 0,01%, 0,1%, dan 1%. Aktivitas fagositosis makrofag menurun pada konsentrasi 10% yang menunjukkan efek imunosupresan. Konsentrasi ekstrak buah Talok 1% menunjukkan aktivitas fagositosis makrofag tertinggi yang berarti sebagai imunostimulan yang optimal. Hal ini menunjukkan bahwa ekstrak buah Talok memiliki fungsi imunomodulator. |
| Abtrak (Bhs. Inggris) | Talok fruit (Muntingia calabura Linn) is a tropical plant commonly found in Indonesia. It is assumed to have some immunomodulatory properties because it contain saponins and flavonoids. This study intended to identify the function of immune-modulatory of the fruit extract on the immune response of the male Wistar rat induced with A. actinomycetemcomitans and to determine an effective dosage to increase the macrophage phagocytic activity of the male Wistar rat induced with A. actinomycetemcomitans. Talok fruit extracted using maceration method by dissolving them in ethanol 70%. The resulted filtrates were then condensed to produce clotted extracts. The extracts were then made into some solution made in four different concentrations. Five groups of Wistar rats were used to prove the function and the best dosage of the Talok fruit extracts. A control group was only provided with aquadest, whereas four experiment groups were given an extract of Talok fruits in the concentrations of 0,01%, 0,1%, 1%, and 10% respectively. The data collected were then analyzed using one-way ANOVA and continued with Post Hoc tes-Duncan in the reliability level of 95%. The results of the analysis show that the treatment of giving the Talok fruit extract influences the macrophage phagocytic activity of the male Wistar rat induced with A. actinomycetemcomitans. The study result showed that macrophage phagocytic activity was keep increased at concentration 0,01%, 0,1%, and 1%. Macrophage phagocytic activity was decreased at concentration 10% that showed immunosupresant effect. Extract concentration of Talok fruit 1% showed top-level of macrophage phagocytic activity that mean as optimal immunostimulatory. This is indicated that Talok fruit extract has immunomodulatory function. |
