Abstrak (Bhs. Indonesia)Penelitian ini bertujuan untuk: (1) menguji zona hambat lima isolat B. subtilis (B1, B46, B209, B211, dan B315) terhadap R. solanacearum in vitro, (2) mempelajari mekanisme penghambatan lima isolat B. subtilis terhadap R. solanacearum, (3) mendapatkan B. subtilis yang paling berpotensi sebagai agens hayati dan pemacu pertumbuhan pada tomat. Penelitian dilaksanakan di rumah kaca dan Laboratorium Perlindungan Tanaman Fakultas Pertanian, Universitas Jenderal Soedirman, Purwokerto. Penelitian dilaksanakan dari bulan Juni sampai November 2013. Penelitian ini terdiri atas dua tahap. Tahap 1 uji antagonis in vitro menggunakan rancangan acak lengkap (RAL), dengan 5 perlakuan dan 5 ulangan. Perlakuan yang dicoba yaitu B. subtilis (B1, B46, B209, 211, dan B315) diuji dengan R. solanacearum. Tahap 2 uji bakteri sebagai pemacu pertumbuhan tomat in planta menggunakan rancangan acak kelompok (RAK), dengan 6 perlakuan dan 4 ulangan. Perlakuan yang dicoba meliputi: K (Kontrol/tanpa B. subtilis), B1(B. subtilis B1), B2 (B. subtilis B46), B3 (B. subtilis B209), B4 (B. subtilis B211), B5 (B. subtilis B315). Variabel yang diamati adalah zona hambatan, mekanisme hambatan, tinggi tanaman, jumlah daun, diameter batang, panjang akar, bobot tanaman segar, bobot tanaman kering, bobot akar segar, bobot akar kering, dan analisis C/N ratio. Hasil penelitian menunjukkan bahwa lima B. subtilis mampu menghambat pertumbuhan R. solanacearum secara in vitro dengan zona hambatan terbesar ditunjukkan oleh B. subtilis B209 sebesar 13,70 mm. Hasil ektraksi B. subtilis baik dengan metanol maupun etil asetat mampu menghambat R. solanacearum secara in vitro dengan zona hambatan terbesar ditunjukkan oleh ekstraksi etil asetat sebesar >1,5 mm. Kelima isolat B. subtilis menunjukkan mekanisme penghambatan secara bakteriostatik. Hasil uji antagonisme in vitro menggunakan koloni B. subtilis menunjukkan aktivitas antibiosis yang lebih baik terhadap R. solanacearum dibandingkan ekstrak B. subtilis. B. subtilis B209 adalah yang terbaik dalam menghambat R. solanacearum in vitro dan meningkatkan pertumbuhan tanaman tomat.

Inggris)This research aims to: (1) test of inhibition zone of B. subtilis (B1, B46, B209, B211, and B315) to R. solanacearum in vitro, (2) to study inhibition mechanism of five B. subtilis isolates to R. solanacearum, and 3) to get one of the most potential of B. subtilis as biocontrol agent and growth promoting of tomatoes in planta. The research was conducted in a greenhouse and Laboratory of Plant Protection, Faculty of Agriculture, Jenderal Soedirman University, Purwokerto. The research was conducted from Juni 2013 to November 2013. The research consisted of two step. Step 1: antagonist in vitro test using Completely Randomized Design (CRD), arranged by 5 treatments and 5 replications. Treatment tested was B. subtilis (B1, B46, B209, 211, and B315) were tested with R. solanacearum. Step 2: test of B. subtilis as a growth promoter in planta tomatoes using Randomized Completely Block Design (RCBD), arranged by 6 treatments and 4 replications. Treatment tested was K (Control/without B. subtilis), B1 (B. subtilis B1), B2 (B. subtilis B46), B3 (B. subtilis B209), B4 (B. subtilis B211), B5 (B. subtilis B315). Variables observed were inhibition zone, inhibition mechanism, plant height, leaves numbers per crop, diameter of rod, root length, plant fresh weight, plant dry weight, root fresh weight, root dry weight, and analysis of C/N ratio. The results of this research showed that five B. subtilis are able to inhibit the growth of R. solanacearum in vitro with the largest inhibition zone indicated by B. subtilis B209 amounted to 13,70 mm. The results of extraction of B. subtilis both in methanol and ethyl acetate were able to inhibit R. solanacearum in vitro with the largest inhibition zone was shown by ethyl acetate extraction of > 1.5 mm.These five B. subtilis isolates showed inhibition mechanism in bakteriostatis. The results of in vitro antagonism test using colony B. subtilis showed better activity antibiosis against R. solanacearum compared extracts B. subtilis. The B. subtilis B209 is the best one in effected to inhibiton R. solanacearum in vitro and growth promoting of tomatoes.
