Indonesia) | Tindakan dalam bidang kedokteran gigi seperti pencabutan, penanganan fraktur rahang, dan pemasangan implan dapat menimbulkan dampak terbentuknya perlukaan pada jaringan tulang. Saat ini banyak digunakan bone graft untuk membantu proses penyembuhan tulang. Selain bone graft saat ini banyak diteliti obat-obatan yang mampu menunjang penyembuhan tulang salah satunya simvastatin. Tujuan penelitian ini adalah untuk membuktikan pengaruh simvastatin per oral danDemineralized Bone Matrix pada proses penyembuhan tulang. Metode penelitian yang digunakan adalah metode eksperimental laboratoris. Dua puluh delapan ekor tikus putih (Rattus norvegicus) galur Wistar disiapkan dibagi ke dalam 4 kelompok dan dilakukan pengeboran di tulang kalvaria. Kelompok (1) diaplikasikan kombinasi simvastatin per oral 20 mg/kgBB dan± 5x10-4 cc Demineralized Bone Matrix; kelompok (2) diaplikasikan kombinasi simvastatin per oral 10 mg/kgBB dan± 5x10-4 cc Demineralized Bone Matrix; kelompok (3) diaplikasikan ± 5x10-4 cc Demineralized Bone Matrix; kelompok (4) tanpa aplikasi simvastatin per oral dan Demineralized Bone Matrix. Tikus dikorbankan 10 hari paska pembentukkan defek tulang selanjutnya dilakukan pembuatan sediaan jaringan histologi dengan pewarnaan Hematoksilin Eosin. Hasil yang diperoleh ditabulasikan menggunakan uji ANOVA satu arah dan LSD. Rerata osteoblas terbanyak dihasilkan oleh kelompok (1) sebanyak 35,00, jika dibandingkan dengan kelompok (2) sebanyak 30,86, kelompok (3) sebanyak 24,57, dan kelompok (4) sebanyak 19,86. Simpulan penelitian ini adalah terdapat pengaruh Demineralized Bone Matrix dan simvastatin terhadap proliferasi osteoblas. Rerata osteoblas terbanyak pada kelompok yang diaplikasikan simvastatin per oral 20 mg/kgBB dan± 5x10-4 cc Demineralized Bone Matrix. |
| Abtrak (Bhs. Inggris) | Dental Procedures such as tooth extraction, jaw fracture treatment, and implantation can cause injury on bone tissue. Nowadays bone graft is currently widely used bone graft to help the healing process of the bone. In addition, beside bone graft, there are several medicines investigated to support bone healing such as simvastatin. The purpose of this study was to demonstrate the influence of oral simvastatin and Demineralized Bone Matrix on bone healing process. The research method used was experimental laboratory method. Twenty eight white rats (Rattusnorvegicus) Wistar strain, were divided into 4 groups and drilled in calvaria. The treatments were group (1) the combination of oral simvastatin 20 mg/kg and ± 5x10-4 cc Demineralized Bone Matrix; group (2) the combination of oral simvastatin 10 mg/kg and ± 5x10-4 cc Demineralized Bone Matrix; group (3) ± 5x10-4 cc Demineralized Bone Matrix; group (4) without application of simvastatin and Demineralized Bone Matrix (as control). Rats were sacrificed 10 days post bone defect formation, histological tissue were preparared done making by hematoxylin eosin staining. The results were tabulated using one-way ANOVA and LSD. The mean osteoblasts were generated by the group (1) of 35.00, compared to the group (2) as much as 30.86, the group (3) as much as 24.57, and group (4) of 19.86. Conclusion of this study was that there was an effect of Demineralized Bone Matrix and simvastatin on proliferation of osteoblasts. The highest osteoblasts mean was in the group that applied with oral simvastatin 20 mg/kg and± 5x10-4 cc Demineralized Bone Matrix. |
