Judul ArtikelPERBEDAAN KADAR TESTOSTERON SERUM TIKUS PUTIH (Rattus norvegicus) JANTAN PADA MODEL STRES PARADOXICAL SLEEP DEPRIVATION (PSD) PASCA PEMBERIAN ANTIOKSIDAN
Abstrak (Bhs. Indonesia)Latar Belakang: Testosteron merupakan hormon androgen yang paling penting dan utama dalam sistem reproduksi pria. Penurunan kadar testosteron berhubungan dengan usia, obesitas, rendahnya High Density Lipoprotein (HDL) dan meningkatnya trigliserida. Penurunan kadar testosteron dapat dipercepat dengan adanya stres. Stres meningkatkan kadar glukokortikoid yang akan memicu peningkatan reactive oxygen species (ROS). Antioksidan adalah senyawa yang dapat melindungi sistem biologis contohnya adalah vitamin C dan E. Antioksidan akan mengurangi jumlah ROS sehingga kadar testosteron dapat terkontrol.
Tujuan: Tujuan penelitian ini adalah membandingkan perbedaan kadar testosteron serum tikus putih (Rattus norvegicus) jantan pada model stres PSD pasca pemberian antioksidan (vitamin C, E dan kombinasi vitamin C dan E).
Metode: Metode eksperimental post test only with control group design dengan Rancangan Acak Lengkap (RAL). Penelitian ini menggunakan 20 ekor tikus putih jantan (Rattus norvegicus) galur wistar usia 3-4 bulan, berat badan 200-300 gram. Tikus dibagi 4 kelompok, yaitu: yaitu kelompok kontrol dengan pemberian model stres PSD, kelompok perlakuan I dengan pemberian model stres PSD ditambah pemberian vitamin C (0,09 mg/grBB/hari), kelompok perlakuan II dengan pemberian model stres PSD ditambah pemberian vitamin E (0,054 mg/grBB/hari), dan kelompok perlakuan III dengan pemberian model stres PSD ditambah pemberian vitamin C dan vitamin E. Antioksidan diberikan dengan cari oral menggunakan sonde lambung. Lama perlakuan yaitu 7 hari. Testosteron serum diukur dengan metode Enzyme Linked Immunosorbent Assay (ELISA). Perbedaan kadar testosteron serum dianalisis menggunakan uji Kruskal-Wallis dan Post Hoc Mann Whitney.
Hasil: Hasil penelitian menunjukkan bahwa terdapat perbedaan rerata kadar testosteron serum yang bermakna antara kelompok kontrol (0,08±0,01 ng/ml) dengan kelompok perlakuan III (0,62±0,53 ng/ml) (p=0,021) dan antara kelompok perlakuan II (0,09±0,04 ng/ml) dengan kelompok perlakuan III (p=0,021).
Kesimpulan: Terdapat perbedaan kadar testosteron serum pasca pemberian antioksidan vitamin C, vitamin E, dan kombinasi keduanya pada tikus putih (Rattus norvegicus) jantan yang terpapar stres model paradoxical sleep deprivation (PSD)
Abtrak (Bhs. Inggris)Background: Testosterone is the most important and principal androgen hormone in male reproduction system. The decrease of testosterone was related to age, obesity, low HDL, and the rising of triglycerides. Decreasing of testosterone levels could be increased by stress. Stress could increase glucocorticoid level that would trigger the rising of ROS. Antioxidant is a chemical compound that could protect the biologycal system, such as vitamin C and E. It may decrease the level of ROS so then the testosterone serum level is controllable.
Objectives: This study aimed to know the difference of testosteron levels of male albino rats (Rattus norvegicus) on the stress PSD model after given antioxidants (Vitamins C, E and combination of those.
Method: Experimental method with post-test only with control group design and Completed Randomized Design (CRD). This study used 20 of rats (Rattus norvegicus) wistar strain 3-4 months old, 200-300 gram body weight. Rats were divided to 4 groups, which was: control group was given intervention the stress PSD model, group I was given intervention the stress PSD model with vitamin C(0,09 mg/grBW/day) treatment, group II was given intervention the stress PSD model with vitamin E (0,054 mg/grBW/day) treatment, and group III was given intervention the stress PSD model and also combination of vitamin C&E. Antioxidant was given per oral. The period of treatment was 7 days. Testosteron serum measured using Enzyme Linked Immunosorbent Assay (ELISA) method. The difference of testosteron serum analyzed by using Kruskal-Wallis and Post Hoc Mann Whitney statistical test.
Result: Result showed that there was a difference average that quite significant: control group (0,08±0,01 ng/ml) with group III (0,62±0,53 ng/ml) (p=0,021) and group II (0,09±0,04 ng/ml) with group III (p=0,021).
Conclusion: There was a difference of testosterone serum levels after being given antioxidant vitamin C &E, and combination of both on male albino rats (Rattus norvegicus) that experienced stress PSD model.
