Abstrak (Bhs. Indonesia)Kanker adalah pertumbuhan sel tubuh yang tidak normal diakibatkan oleh proliferasi yang berlebihan. Kanker payudara memiliki frekuensi kejadian yang tinggi di antara jenis kanker lain yang sering menyerang wanita. Secara empiris rimpang kunyit dan daun sirsak banyak digunakan dalam pengobatan kanker. Curcuma longa (kunyit) mengandung kurkumin. Annona muricata L (daun sirsak) mengandung acetogenin. Penggabungan bahan aktif dalam tanaman rimpang kunyit dan daun sirsak diharapkan dapat meningkatkan efektivitas anti kanker dari keduanya. Tujuan penelitian ini adalah mengetahui efek sitotoksik, aktivitas apoptosis dan ekspresi protein p53 kombinasi ekstrak etanol rimpang kunyit dan daun sirsak pada sel kanker payudara T47D.
Serbuk rimpang kunyit dan daun sirsak dimaserasi dengan etanol 96% selama 3x24 jam. Aktivitas sitotoksik diuji dengan Yellow MTT assay. Data persentase penghambatan dianalisis probit dengan SPSS untuk mendapat IC50. Nilai IC50 dianalisis dengan uji anova dan dilanjutkan dengan uji LSD. Apoptosis sel T47D diamati dengan metode doublestainning menggunakan etidium bromida-akridin orange dan dapat dihitung persentase apoptosis. Ekspresi protein p53 sel T47D diamati dengan metode imunositokimia dan dapat diperoleh persentase ekpresi protein p53.
Kombinasi ekstrak rimpang kunyit dan daun sirsak berpotensi memiliki efek sitotoksik dengan IC50 sebesar 17,650 µg/mL, sementara rimpang kunyit 12,992 µg/mL dan daun sirsak 22,009 µg/mL. Hasil analisis statistik dari ketiga nilai IC50 ekstrak memiliki efek sitotoksik dan menunjukkan perbedaan yang bermakna. Ekstrak rimpang kunyit, daun sirsak dan campuran ekstrak rimpang kunyit-daun sirsak (1:1) mampu menginduksi aktivitas apoptosis dengan persentase apoptosis masing-masing sebesar 32,86%; 12,25% dan 26,62%. Persentase pengaruh ekstrak rimpang kunyit, daun sirsak dan campuran ekstrak rimpang kunyit-daun sirsak (1:1) terhadap ekspresi protein p53 pada sel T47D masing-masing sebesar 47,76 %; 13,33% dan 24,00 %.
Abtrak (Bhs. Inggris)Cancer is a growing body of abnormal cells due to the excessive proliferation. Breast cancer had a high frequency of occurrence among other types of cancer affected women. Empirically, turmeric and soursop leaves has been widely used to treat cancer. Curcuma longa (turmeric) containes curcumin while Annonamuricata L (soursop leaf) containes acetogenin. The combination of turmeric plants and soursop leaves was expected to be more effective in treating cancer. The objectives of this study were to determine the effect of cytotoxic, apoptotic activity and p53 protein expression combined from ethanol extract of turmeric rhizome and sour sop leaves on breast cancer cell T47D.
Turmeric Rhizome powder and soursop leaves were macerated in 96% ethanol for 3x24 hours. Cytotoxic activity was tested with Yellow MTT assay. The probity of inhibition percentage data were analyzed by SPSS to obtain IC50. IC50 values were analyzed by one way ANOVA followed by LSD test. T47D cell apoptosis was evaluated by double stainning method using ethidium bromide-acridine orange and apoptosis percentage was calculated. p53 protein expression of T47D cells was evaluated by immunocytochemistry method and the proportion percentageof p53 protein expression could be acquired.
The combination of turmeric rhizome extract and soursop leaves potentially had a cytotoxic effect with IC50 of 17,650 µg/mL, while turmeric rhizome 12,992 µg/mL and soursop leaves 22,009 µg/mL. The statistical analysis results of these three IC50 extracts values had cytotoxic effects and showed significant differences. Turmeric rhizome extract, soursop leaves and the combination of turmeric rhizome-soursop leaves extract(1:1) was able to induce apoptotic activity with the apoptotic percentage of 32,86%; 12,25% dan 26,62% respectively. The effect of turmeric rhizome extract, soursop leaves and their combination (1:1) on the expression of p53 protein in T47D cells was 47,76%, 13,33% and 24,00% respectively.
