| Judul Artikel | Aktivitas Ekstrak Etanol Daun Jatropha curcas sebagai Anti Inflamasi pada Tikus Adjuvant-Induced Arthritis |
| Abstrak (Bhs. Indonesia) | Berdasarkan penelitian sebelumnya, daun jarak pagar (Jatropha curcas) berpotensi sebagai anti inflamasi dan terbukti mampu menghambat edema akut. Tujuan penelitian ini adalah untuk mengetahui aktivitas anti inflamasi kronis dari ekstrak etanol daun J. curcas terhadap edema pada tikus adjuvant induced arthritis (AIA) yang diinduksi complete freund’s adjuvant (CFA). Tikus wistar jantan dibagi menjadi 6 kelompok (n=6), terdiri dari kelompok normal (NaCl 0,9%), kelompok kontrol (CFA), kelompok CFA+Na-diklofenak dengan dosis 6,75 mg/kg BB (p.o), dan kelompok CFA+ekstrak etanol daun J. curcas dengan dosis 150 mg/kg BB, 300 mg/kg BB dan 600 mg/kg BB (p.o). Setiap kelompok diinduksi dengan 0,2 ml CFA (1 mg/ml) pada hari ke-1 dan diberikan injeksi booster dengan 0,1 ml CFA (1 mg/ml) pada hari ke-5. Ekstrak diberikan pada hari ke 14-28. Keliling edema diamati pada hari ke 0, 1, 4, 8, 12, 16, 20, 24, dan 28. Rekruitmen neutrofil dan histopatologi sendi tibioarsal diamati pada hari ke-29 serta dianalisis secara deskriptif. Data kenaikan keliling edema dihitung sebagai persentasi kenaikan edema dan persentasi penghambatan edema serta dianalisis dengan menggunakan analisis statistika Kruskal-Wallis yang dilanjut dengan uji Mann-Whitney (data berdistribusi tidak normal) dan ANOVA satu arah yang dilanjut dengan uji LSD (data berdistribusi normal). Hasil analisis statistika menunjukkan bahwa kelompok ekstrak etanol daun J. curcas menunjukkan nilai mean pada dosis 150 mg/kg BB, 300 mg/kg BB, dan 600 mg/kg BB masing-masing adalah sebesar 0,233±0,052, 0,283±0,075, dan 0,267±0,052 (p < 0,05). Hasil analisis deskriptif menunjukkan bahwa kelompok ekstrak pada ketiga dosis tersebut mampu menghambat inflamasi dan destruksi kartilago pada jaringan sendi, namun tidak menunjukkan penurunan terhadap rekruitmen neutrofil. Hasil ini mengindikasikan bahwa ekstrak etanol daun J. curcas memiliki aktivitas anti inflamasi kronis dan berpotensi sebagai anti anthritis. |
| Abtrak (Bhs. Inggris) | Based on previous study, Jatropha (Jatropha curcas) leaf potentially as an anti-inflammatory and has been shown to inhibit acute edema. The purpose of this study was to determine the anti-inflammatory chronic activity of ethanolic extract of J. curcas leaf against edema on adjuvant induced arthritis (AIA) in male wistar rats induced by complete freund's adjuvant (CFA). Male wistar rats were divided into 6 groups (n=6), consists of normal group (0.9% NaCl), control group (CFA), CFA+Na-diclofenac group at a dose of 6.75 mg/kg (p.o), and CFA+ethanolic extract group of J. curcas leaf with a dose of 150 mg/kg, 300 mg /kg and 600 mg/kg (p.o). Each group was induced by 0.2 ml CFA (1 mg/ml) on day 1 and a booster injection with 0.1 ml of CFA (1 mg/ml) was administered on day 5. Extract was treated on days 14-28. Circumference of edema was observed on days 0, 1, 4, 8, 12, 16, 20, 24, and 28. Recruitments of neutrophils and histopathology of tibiotarsal joint was observed on day 29, and analyzed descriptively. Increase of edema circumference were calculated as percentage increase and percentage inhibition of edema, and analyzed by Kruskal-Wallis followed by Mann-Whitney test and one-way ANOVA followed by LSD test. Results of statistical analysis indicate that the ethanolic extract of J. curcas leaves group showed mean value for dose 150 mg/kg, 300 mg/kg, and 600 mg/kg were 0.233±0.052, 0.283±0.075 and 0.267±0.052 (p < 0,05), respectively. Result of descriptive analysis showed that the extract at a dose group is able to inhibit inflammation and cartilage destruction in the joint tissues, but did not show a decline in the recruitment of neutrophils. These results indicate that the ethanol extract of J. curcas leaf has anti-inflammatory chronic activity and potentially as an anti anthritis. |
