Abstrak (Bhs. Indonesia)Penelitian ini melaporkan efektivitas triploidisasi nilem dengan kejut dingin 40 C pada waktu dan lama kejut berbeda. Eksperimen ini menerapkan 10 perlakuan kombinasi waktu kejut (1, 3, atau 5 menit dari waktu fertilisasi) dan lama kejut (15, 20, atau 25 menit) berbeda pada telur nilem terfertilisasi. Variabel yang diamati yaitu fertilitas, daya tetas, abnormalitas larva, sintasan benih, panjang benih dan dimensi sel darah merah (eritrosit) benih. Hasilnya membuktikan bahwa perlakuan triploidisasi kejut dingin ini efektif dari bukti dimensi eritrosit benih perlakuan triploid secara sangat nyata (P<0.01) jauh lebih besar dibanding dimensi eritrosit benih kelompok diploid normal kontrolnya. Dimenis sel darah merah berupa ukuran panjang (major axes), lebar (minor axes), luas dan volume eritrosit. Rerata data fertilitas dan panjang tubuh benih perlakuan triploidisasi kejut dingin terbukti secara sangat nyata (P<0.01) dipengaruhi perlakuan, namun rerata persentase penetasan, persentase abnormalitas larva, dan sintasan tidak secara nyata (P>0.05) dipengaruhi perlakuan kejut dingin. Dengan demikian kejut dingin nilem yang dilaporkan ini siap diterapkan pada bidang akuakultur .
Abtrak (Bhs. Inggris)This study reports on shark minnow triploidization effectiveness from treatment of 4°C cold shocked at different time and different duration of the shock. This experiment applied 10 treatments of three different duration cold shock 4oC for 15, 20 or 25 minutes of fertilized eggs at three different time : 1, 3 or 5 minutes from fertilization time. Fertilization rate, hatching rate, percentage of larvae abnormalities, larvae and juvenile survival rate, juvenile body length and dimension of erythrocyte were assessed. Results showed that all triploidization treatments were effectively produce triploid juvenile as proven from data of erythrocyte dimension which was highly significant different (P<0.01) among treatment. Erythrocytes dimension composed of the major axes, minor axes, surface area and volume from erythrocytes cell of triploid juvenile group which were much bigger that from the diploid control one. Fertility rate and juvenile body length were highly significant different (P<0.01) among treatments. However, the hatching rate, larvae abnormalities percentage, as well as the juvenile survival rate were no significant (P>0.05) among treatments. This is the first cold shock triploid successful protocol and ready for aquaculture application.
