| Judul Artikel | Transesterifikasi Minyak Biji Nyamplung, Formulasi Nanoemulsi, dan Uji Aktivitas Antioksidannya Menggunakan Metode DPPH |
| Abstrak (Bhs. Indonesia) | Minyak biji nyamplung (Calophyllum inophyllum L.) memiliki aktivitas antioksidan yang berpotensi menghambat radikal bebas penyebab penuaan dan berbagai penyakit degeneratif. Penelitian ini bertujuan untuk mengevaluasi aktivitas antioksidan minyak biji nyamplung dan produk transesterifikasinya, serta mengembangkan produk transesterifikasi dengan aktivitas antioksidan tertinggi ke dalam sediaan emulsi dan nanoemulsi. Metil ester disintesis melalui reaksi transesterifikasi pada suhu 60 ºC selama 6 jam dan dikarakterisasi menggunakan GC-MS. Aktivitas antioksidan minyak biji nyamplung dan metil ester dievaluasi dengan metode DPPH. Metil ester diformulasikan ke dalam sediaan emulsi dengan variasi konsentrasi 0, 1, 3, 5, dan 7%, kemudian formula dengan karakteristik terbaik dikembangkan menjadi sediaan nanoemulsi. Karakterisasi emulsi meliputi pengujian pH, viskositas, dan stabilitas fisik, sedangkan nanoemulsi dikarakterisasi menggunakan PSA. Aktivitas antioksidan sediaan juga dievaluasi menggunakan metode DPPH. Hasil analisis GC-MS menunjukkan komponen utama metil ester adalah metil oleat, metil palmitat, metil linoleat, dan metil stearat. Transesterifikasi meningkatkan aktivitas antioksidan minyak biji nyamplung yang ditunjukkan dengan nilai IC50 metil ester sebesar 398,5040 ppm. Formula emulsi terbaik diperoleh pada FE3 yang mengandung 3% metil ester dengan pH 6,4; viskositas 23,5 mPa.s; serta stabilitas fisik yang baik selama penyimpanan, ditandai dengan tidak adanya perubahan warna signifikan, tetap jernih, homogen, dan tidak beraroma tengik. Nanoemulsi FE3 memiliki ukuran partikel 14,16 nm dengan distribusi 100% dan nilai PDI 0,1198. Uji aktivitas antioksidan menunjukkan bahwa nilai IC50 nanoemulsi FE3 (283,316 ppm) lebih rendah dibandingkan emulsi FE3 (362,745 ppm), yang mengindikasikan aktivitas antioksidan lebih kuat. Inggris) | Nyamplung seed oil (Calophyllum inophyllum L.) possesses antioxidant activity that has the potential to inhibit free radicals, which cause aging and various degenerative diseases. This study aims to evaluate the antioxidant activity of nyamplung seed oil and its transesterification products, as well as to develop the transesterification product with the highest antioxidant activity into emulsion and nanoemulsion formulations. The methyl ester was synthesized via a transesterification reaction at 60 °C for 6 hours and then characterized using GC- MS. The antioxidant activity of nyamplung seed oil and the methyl ester was evaluated using the DPPH method. The methyl esters were formulated into emulsion formulations with varying concentrations of 0, 1, 3, 5, and 7%; the formulation with the best characteristics was then developed into a nanoemulsion formulation. Characterization of the emulsions included testing for pH, viscosity, and physical stability, while the nanoemulsions were characterized using PSA. The antioxidant activity of the formulations was also evaluated using the DPPH method. GC-MS analysis results showed that the main components of the methyl esters were methyl oleate, methyl palmitate, methyl linoleate, and methyl stearate. Transesterification enhanced the antioxidant activity of nyamplung seed oil, as indicated by an IC50 value of 398.5040 ppm for the methyl esters. The best emulsion formula was obtained for FE3, which contained 3% methyl ester, had a pH of 6.4 and a viscosity of 23.5 mPa·s, and exhibited good physical stability during storage, as indicated by the absence of significant color changes; it remained clear, homogeneous, and free of rancid odors. The FE3 nanoemulsion has a particle size of 14.16 nm with a 100% distribution and a PDI value of 0.1198. Antioxidant activity testing showed that the IC50 value of the FE3 nanoemulsion (283.316 ppm) was lower than that of the FE3 emulsion (362.745 ppm), indicating stronger antioxidant activity.
