Abstrak (Bhs. Indonesia)Ikan gurami (Osphronemus gouramy) merupakan salah satu komoditas ikan air tawar yang memiliki nilai ekonomi penting, tetapi memiliki laju pertumbuhan yang relatif lambat, terutama pada fase larva. Fase tersebut merupakan periode kritis karena fungsi pencernaan larva masih belum berkembang secara optimal. Efisiensi pencernaan sangat dipengaruhi oleh aktivitas enzim protease dan amilase, yang terutama dipengaruhi oleh suhu lingkungan sebagai regulator utama metabolisme pada organisme poikilotermik. Namun, informasi mengenai suhu optimal yang dapat meningkatkan efisiensi aktivitas enzim pada larva gurami masih terbatas, sehingga pertumbuhan dan kelangsungan hidup pada praktik pembenihan belum optimal. Penelitian ini bertujuan untuk menentukan suhu terbaik dalam meningkatkan aktivitas enzim dan pertumbuhan somatik larva gurami secara efektif. Penelitian dilakukan secara eksperimental menggunakan Rancangan Acak Lengkap (RAL) dengan empat perlakuan suhu dan lima ulangan. Perlakuan terdiri atas suhu tidak terkontrol yang berfluktuasi antara 24–27 °C, serta suhu terkontrol 27 ± 1 °C, 30 ± 1 °C, dan 33 ± 1 °C. Parameter yang diamati meliputi aktivitas protease, aktivitas amilase, dan faktor kondisi yang diukur pada umur 15, 25, 35, dan 45 hari setelah menetas (HSM). Aktivitas enzim diukur menggunakan spektrofotometer, dengan aktivitas protease ditentukan berdasarkan konsentrasi tirosin, sedangkan aktivitas amilase ditentukan berdasarkan konsentrasi maltosa. Faktor kondisi ditentukan berdasarkan bobot dan panjang total larva. Data dianalisis menggunakan Analysis of Variance (ANOVA) yang dilanjutkan dengan Duncan's Multiple Range Test (DMRT). Hasil penelitian menunjukkan bahwa aktivitas protease dipengaruhi secara signifikan oleh interaksi antara suhu pemeliharaan dan waktu pengamatan, dengan aktivitas tertinggi pada suhu tidak terkontrol pada umur 45 HSM. Sebaliknya, aktivitas amilase dipengaruhi secara terpisah oleh suhu dan waktu pengamatan, dengan rata-rata aktivitas tertinggi pada suhu 30 °C. Sementara itu, faktor kondisi relatif stabil dan tidak menunjukkan perbedaan yang signifikan pada seluruh perlakuan. Berdasarkan hasil penelitian, enzim pencernaan pada larva gurami memiliki suhu optimum yang berbeda; suhu 30 °C mengoptimalkan aktivitas amilase, sedangkan aktivitas protease mencapai nilai maksimum pada profil suhu yang berbeda. Perbedaan suhu tersebut tidak memberikan dampak negatif terhadap proporsionalitas tubuh larva.
Abtrak (Bhs. Inggris)The gourami (Osphronemus gouramy) is an economically significant freshwater species noted for its comparatively slow growth rate, especially during the larval stage. This phase signifies a crucial interval during which digestive functions remain inadequately developed. Digestive efficiency is significantly dependent on the activities of proteases and amylases, which are primarily regulated by environmental temperature, the main regulator of metabolism in poikilothermic species. Nevertheless, data concerning the precise thermal optima to enhance enzymatic efficiency in gourami larvae remains scarce, resulting in unsatisfactory growth and survival rates in hatchery practices. This research aimed to identify the best temperature for effectively enhancing enzymatic activity and somatic growth in gourami larvae. The research applied an experimental approach utilizing a Completely Randomized Design (CRD) that featured four temperature treatments with five replicates each. These treatments consisted of an uncontrolled temperature fluctuating between 24 and 27 °C, alongside controlled temperatures of 27 ± 1 °C, 30 ± 1 °C, and 33 ± 1 °C. Measured parameters encompass protease activity, amylase activity, and condition factor, assessed monthly at 15, 25, 35, and 45 days after hatching (DAH). Enzyme activities were quantified using a spectrophotometer; protease activity was assessed by tyrosine concentrations, whilst amylase activity was evaluated through maltose concentrations. Simultaneously, the condition factor was determined by measuring larval weight and total length. Data are examined utilizing Analysis of Variance (ANOVA) succeeded by Duncan's Multiple Range Test. The results demonstrated that protease activity was significantly influenced by the interaction of rearing temperature and observation time, reaching its peak at uncontrolled temperature on 45 DAH. Conversely, amylase activity was independently affected by temperature and time, with the highest mean activity recorded at 30 °C. Meanwhile, the condition factor remained relatively stable and showed no significant differences across all treatments. In conclusion, the digestive enzymes of gourami larvae exhibit distinct thermal optima; while 30 °C optimizes amylase production, a different temperature profile is required to maximize protease activity, all of which do not negatively impact the body proportionality of the larvae.
