Abstrak (Bhs. Indonesia)Penyakit degeneratif adalah penyakit yang disebabkan oleh penurunan fungsi organ
tubuh karena paparan radikal bebas berlebih yang melebihi kapasitas antioksidan
endogen sehingga diperlukan antioksidan alami. Minyak atsiri daun pala (Myristica
fragrans) diketahui mengandung senyawa aktif sebagai antioksidan alami, namun
bersifat hidrofobik sehingga diperlukan teknologi nanoemulsi untuk meningkatkan
kelarutan senyawa aktif. Penelitian ini bertujuan untuk mengetahui fraksi hasil
destilasi fraksinasi pengurangan tekanan yang memiliki aktivitas antioksidan
teraktif dan menentukan formula nanoemulsi terbaik. Metode penelitian meliputi
destilasi fraksinasi pengurangan tekanan minyak atsiri daun pala, analisis senyawa
menggunakan Gas Chromatography-Mass Spectrometry (GC-MS), uji aktivitas
antioksidan menggunakan metode DPPH, formulasi menggunakan surfaktan
Tween 80 dan ko-surfaktan propilen glikol, karakterisasi nanoemulsi serta uji
aktivitas antioksidan nanoemulsi. Hasil destilasi fraksinasi diperoleh tiga fraksi
dengan hasil GC-MS berturut-turut yaitu fraksi 1 (84,33%) didominasi golongan
monoterpen, fraksi 2 (6,17%) didominasi golongan monoterpen, dan fraksi 3
(3,42%) didominasi golongan fenilpropanoid yaitu eugenol dan miristisin. Uji
aktivitas antioksidan menunjukkan fraksi 3 memiliki aktivitas antioksidan teraktif
dengan nilai IC50 sebesar 12,67 ppm, kemudian diformulasi menjadi nanoemulsi
dengan variasi konsentrasi 0%, 1%, 3%, dan 5% menggunakan surfaktan Tween 80
(20%) dan ko-surfaktan propilen glikol (15%). Hasil karakterisasi menunjukkan FN
1 dan FN2 merupakan formula terbaik dengan nilai pH 6,9 dan 6,8, bertipe oil in
water (o/w), viskositas 14,6 dan 38,8 mPa.s, persen transmitan 99,6 dan 99,3%,
ukuran partikel dominan 14,28 nm dan 16,51 nm, serta stabil. Aktivitas antioksidan
nanoemulsi lebih aktif daripada non-nanoemulsi. Nilai IC50 FR3 (1%), FR3 (3%),
FN1, dan FN2 berturut-turut yaitu 13,19; 11,66; 12,42; dan 8,81 ppm yang termasuk
dalam kategori sangat kuat

excessive exposure to free radicals that exceeds the body’s endogenous antioxidant
capacity, thereby necessitating the use of natural antioxidants. Essential oil from
nutmeg leaves (Myristica fragrans) is known to contain active compounds that act
as natural antioxidants; however, these compounds are hydrophobic, so
nanoemulsion technology is required to enhance their solubility. This study aims to
identify the fraction from pressure-reduced fractional distillation that exhibits the
highest antioxidant activity and to determine the optimal nanoemulsion formulation.
The research methods included pressure-reduced fractional distillation of nutmeg
leaf essential oil, compound analysis using Gas Chromatography-Mass
Spectrometry (GC-MS), antioxidant activity testing using the DPPH method,
formulation using surfactant Tween 80 and co-surfactant propylene glycol,
characterization nanoemulsion, and antioxidant activity testing of the
nanoemulsions. The fractional distillation yielded three fractions with the following
GC-MS results: fraction 1 (84.33%) was dominated by monoterpenes, fraction 2
(6.17%) was dominated by monoterpenes, and fraction 3 (3.42%) was dominated
by phenylpropanoids, namely eugenol and myristicin. Antioxidant activity testing
showed that fraction 3 had the highest antioxidant activity with an IC50 value of
12.67 ppm; it was then formulated into nanoemulsions with concentrations of 0%,
1%, 3%, and 5% using the surfactant Tween 80 (20%) and the co-surfactant
propylene glycol (15%). Characterization results showed that FN1 and FN2 were
the best formulations, with pH values of 6.9 and 6.8, an oil-in-water (o/w) type,
viscosities of 14.6 and 38.8 mPa.s, transmittance percentages of 99.6% and 99.3%,
dominant particle sizes of 14.28 nm and 16.51 nm, and were stable. The antioxidant
activity of the nanoemulsions was higher than that of the non-nanoemulsions. The
IC50 values for FR3 (1%), FR3 (3%), FN1, and FN2 were 13.19; 11.66; 12.42; and
8.81 ppm, respectively, which fall into the very strong category.
