Abstrak (Bhs. Indonesia)Kappaphycus striatus merupakan rumput laut merah yang mengandung berbagai senyawa metabolit bioaktif. Profil metabolit yang dihasilkan dapat berbeda bergantung pada metode budidaya, masa panen, kondisi sampel, metode ekstraksi, dan jenis pelarut yang digunakan. Penelitian ini bertujuan menganalisis profil metabolomik K. striatus berdasarkan perbedaan metode budidaya, masa panen, kondisi sampel, metode ekstraksi, dan jenis pelarut menggunakan Liquid Chromatography–High Resolution Mass Spectrometry (LC-HRMS) berbasis molecular networking, serta mengidentifikasi senyawa yang terdereplikasi. Sampel berasal dari Teluk Ekas, Nusa Tenggara Barat, dengan perlakuan metode budidaya long line apung dan long line dasar, masa panen hari ke-0, 25, 35, dan 45, kondisi segar dan kering, metode ekstraksi Microwave-Assisted Extraction (MAE) dan maserasi bertingkat, serta pelarut n-heksana, etil asetat, dan metanol. Data dianalisis menggunakan GNPS, Cytoscape, Compass DataAnalysis, dan RStudio. Sebanyak 84 fitur metabolit terdeteksi dan enam senyawa berhasil terdereplikasi, yaitu pheophorbide A, 10S-hydroxypheophorbide a, cholecalciferol, cholesterol, erucamide, dan 13-docosenamide. Perbedaan masa panen menunjukkan variasi profil metabolomik, dengan jumlah node tertinggi diperoleh pada hari ke-25. Sementara itu, metode budidaya, kondisi sampel, metode ekstraksi, dan jenis pelarut tidak menunjukkan perbedaan jumlah node yang signifikan. Senyawa yang berhasil terdereplikasi dilaporkan memiliki potensi sebagai antikanker, antiinflamasi, dan antimikroba, serta berperan dalam metabolisme, sistem imun, dan stabilitas membran
Abtrak (Bhs. Inggris)Kappaphycus striatus is a red seaweed containing various bioactive metabolites. Its metabolomic profile may vary depending on cultivation method, harvest time, sample condition, extraction method, and solvent type. This study aimed to analyze the metabolomic profile of K. striatus under different cultivation methods, harvest times, sample conditions, extraction methods, and solvent types using Liquid Chromatography–High Resolution Mass Spectrometry (LC-HRMS) combined with a molecular networking approach, as well as to identify dereplicated compounds. Samples were collected from Ekas Bay, West Nusa Tenggara, Indonesia, and included floating and bottom long-line cultivation methods, harvest times of 0, 25, 35, and 45 days, fresh and dried sample conditions, Microwave-Assisted Extraction (MAE) and sequential maceration extraction methods, and n-hexane, ethyl acetate, and methanol as extraction solvents. The data were analyzed using GNPS, Cytoscape, Compass DataAnalysis, and RStudio. A total of 84 metabolite features were detected, and six compounds were successfully dereplicated: pheophorbide A, 10S-hydroxypheophorbide a, cholecalciferol, cholesterol, erucamide, and 13-docosenamide. Differences in harvest time were associated with variations in the metabolomic profile, with the highest number of nodes observed at day 25. In contrast, cultivation method, sample condition, extraction method, and solvent type did not result in significant differences in the number of nodes. The dereplicated compounds have been reported to possess anticancer, anti-inflammatory, and antimicrobial potential, as well as roles in metabolism, immune function, and membrane stability.
