Berdasarkan Gen 16S rRNA
Abstrak (Bhs. Indonesia)Pencemaran laut oleh minyak bumi yang mengandung Polycyclic Aromatic
Hydrocarbons (PAH) merupakan masalah lingkungan global yang bersifat
persisten, toksik, dan karsinogenik, sementara informasi mengenai mikroba
pendegradasi PAH asal methane seep di Perairan Sabang masih terbatas.
Penelitian ini bertujuan untuk mengetahui potensi bakteri methane seep sebagai
pendegradasi PAH serta mengidentifikasi spesiesnya berdasarkan gen 16S
rRNA. Metode yang digunakan adalah eksperimental terhadap 75 isolat bakteri
koleksi Laboratorium Mikrobiologi Laut BRIN yang diskrining menggunakan
sublimation test terhadap senyawa dibenzothiophene, fluorene, dan phenanthrene,
dilanjutkan dengan pewarnaan Gram, ekstraksi DNA, amplifikasi PCR gen 16S
rRNA, serta analisis BLAST dan pohon filogenetik menggunakan MEGA12.
Hasil penelitian menunjukkan bahwa dari 75 isolat tersebut, diperoleh 8 isolat
pendegradasi dibenzothiophene, 6 isolat fluorene, dan 6 isolat phenanthrene yang
membentuk zona bening, dengan 4 isolat terbaik (MBAC24-PAH-26, -27, -31, dan -32) yang mampu mendegradasi ketiga senyawa tersebut. Isolat MBAC24-PAH
26 teridentifikasi sebagai Bacillus firmus dengan kemiripan 99,65% dan nilai
bootstrap 98. Penelitian ini menyimpulkan bahwa methane seep Perairan Sabang
menyimpan bakteri Gram positif berpotensi tinggi sebagai agen bioremediasi
PAH di lingkungan laut.
Abtrak (Bhs. Inggris)Marine pollution caused by petroleum containing Polycyclic Aromatic Hydrocarbons
(PAH) was a global environmental problem that remained persistent, toxic, and
carcinogenic, while information on PAH-degrading microbes originating from methane
seeps in Sabang Waters was still limited. This study aimed to determine the potential of
methane seep bacteria as PAH degraders and to identify their species based on the 16S
rRNA gene. The method used was experimental, applied to 75 bacterial isolates from the
collection of the Marine Microbiology Laboratory, BRIN, which were screened using a
sublimation test against dibenzothiophene, fluorene, and phenanthrene, followed by
Gram staining, DNA extraction, PCR amplification of the 16S rRNA gene, and BLAST
and phylogenetic tree analysis using MEGA12. The results showed that among the 75
isolates, 8 isolates degraded dibenzothiophene, 6 isolates degraded fluorene, and 6 isolates
degraded phenanthrene, as indicated by the formation of clear zones, with 4 of the best
isolates (MBAC24-PAH-26, -27, -31, and -32) being able to degrade all three compounds.
Isolate MBAC24-PAH-26 was identified as Bacillus firmus with a 99.65% similarity and
a bootstrap value of 98. This study concluded that the methane seep in Sabang Waters
harbored Gram-positive bacteria with high potential as PAH bioremediation agents in the
marine environment.
