Abstrak (Bhs. Indonesia)Ikan nilem (Osteochilus vittatus) merupakan salah satu organisme yang berpotensi digunakan sebagai bioindikator pencemaran perairan karena sensitivitasnya terhadap berbagai polutan lingkungan. Salah satu polutan yang berbahaya adalah Potasium dikromat (K₂Cr₂O₇) yang mengandung kromium heksavalen (Cr(VI)), senyawa neurotoksik yang dapat memicu pembentukan reactive oxygen species (ROS), menyebabkan stres oksidatif, kerusakan DNA, dan gangguan fungsi sel. Penelitian ini bertujuan untuk mengevaluasi pengaruh paparan Potasium dikromat terhadap pertumbuhan dan viabilitas sel cerebrum O. vittatus secara in vitro serta menentukan konsentrasi yang berpotensi menimbulkan efek toksik.
Penelitian ini menggunakan Rancangan Acak Kelompok (RAK) dengan lima konsentrasi K₂Cr₂O₇ (0; 0,125; 0,250; 0,375; dan 0,500 ppm) sebagai perlakuan dan jenis kelamin ikan sebagai kelompok. Sel cerebrum O. vittatus diperoleh melalui kultur fragmen menggunakan medium DMEM yang disuplementasi dengan FBS 10%, penicillin-streptomycin-antimycotic 5%, dan L-glutamine 5%. Pengamatan meliputi densitas, viabilitas, kurva pertumbuhan, Population Doubling Time (PDT), dan Population Doubling Level (PDL). Hasil penelitian menunjukkan bahwa kultur fragmen cerebrum menghasilkan sel dengan viabilitas tinggi (99–100%) dan kemampuan proliferasi yang baik. Kurva pertumbuhan menunjukkan fase adaptasi pada awal kultur yang diikuti peningkatan proliferasi hingga hari keempat. Laju proliferasi sel-sel cerebrum ikan jantan lebih lambat (PDT= 25,58 jam) dibanding sel-sel cerebrum ikan betina (PDT= 19,60 jam). Paparan K₂Cr₂O₇ menyebabkan penurunan densitas sel secara dose-dependent dan berpengaruh nyata terhadap densitas sel (p < 0,05), terutama pada konsentrasi 0,375 ppm dan 0,500 ppm, namun tidak berpengaruh nyata terhadap viabilitas sel (p > 0,05).
Berdasarkan hasil penelitian dapat disimpulkan bahwa potasium dikromat tidak berpengaruh terhadap viabilitas sel cerebrum O. vittatus, tetapi menghambat pertumbuhan sel secara dose-dependent. Konsentrasi ≥ 0,375 ppm mampu menimbulkan efek toksik terhadap sel cerebrum O. vittatus. Hasil penelitian ini menunjukkan bahwa kultur primer cerebrum O. vittatus berpotensi digunakan sebagai model in vitro untuk kajian neurotoksisitas logam berat dan penilaian risiko pencemaran perairan.
Abtrak (Bhs. Inggris)The bonylip barb (Osteochilus vittatus) is a freshwater fish which is potential as a bioindicator of aquatic pollution due to its sensitivity to the environmental contaminants. One of the hazardous pollutant is potassium dichromate (K₂Cr₂O₇), which contains hexavalent chromium (Cr(VI)). This ion is a neurotoxic compound capable of generating reactive oxygen species (ROS), inducing oxidative stress, causing DNA damage, and disrupting cellular functions. This study aimed to evaluate the effects of potassium dichromate exposure on the growth and viability of O. vittatus cerebrum cells in vitro and to determine concentrations capable of inducing toxic effects.
This study was conducted using randomized block design (RBD) with five concentrations of K₂Cr₂O₇ (0; 0.125; 0.250; 0.375; and 0.500 ppm), and fish sex as the block. O. vittatus cerebrum fragments were obtained through fragment culture using DMEM supplemented with 10% FBS, 5% penicillin-streptomycin-antimycotic, and 5% L-glutamine. The data included cell density, cell viability, growth curves, Population Doubling Time (PDT), and Population Doubling Level (PDL). The results showed that cerebrum fragment culture successfully produced outgrowth with high viability (99–100%) and good proliferative capacity. The growth curve showed a lag phase at the first day of the culture, followed by exponential phase untuil the fourth day. The PDT values for male and female cells were 25.58 hours and 19.60 hours, respectively, indicating a higher proliferation rate in female cerebral cells. Exposure to K₂Cr₂O₇ caused a dose-dependent decrease in cell density and had a significant effect on cell density (p < 0.05), particularly at concentrations of 0.375 ppm and 0.500 ppm; however, it had no significant effect on cell viability (p > 0.05).
Based on the findings of this study, it can be concluded that potassium dichromate did not affect the viability of O. vittatus cerebrum cells but inhibited cell growth in a dose-dependent manner. Concentrations of ≥ 0.375 ppm were capable of inducing toxic effects on O. vittatus cerebrum cells. These findings indicate that primary cerebrum cell culture of O. vittatus has the potential to serve as an in vitro model for investigating the neurotoxicity of heavy metals and for assessing the risks associated with aquatic environmental pollution.
