Indonesia) | Plastik hitam jenis polikarbonat banyak digunakan sebagai kemasan pangan dan kebutuhan rumah tangga, namun mengandung bahan kimia tambahan seperti bisphenol A, ftalat, serta logam berat (timbal, kadmium, dan kromium) yang dapat bermigrasi ke dalam makanan atau minuman saat dipanaskan atau digunakan berulang sehingga berpotensi menimbulkan stres oksidatif dan kerusakan hati. Kerusakan tersebut ditandai dengan meningkatnya aktivitas aspartate aminotransferase dan alanine aminotransferase dalam darah. Salah satu upaya untuk menekan dampak toksik tersebut adalah pemberian senyawa alami yang memiliki aktivitas antioksidan tinggi, seperti tomat (Solanum lycopersicum L.) yang mengandung likopen, flavonoid, dan vitamin C. Penelitian ini bertujuan untuk mengetahui aktivitas aspartate aminotransferase dan alanine aminotransferase pada tikus putih jantan yang dipaparkan air rebusan plastik hitam dan diberi ekstrak tomat, serta menentukan dosis ekstrak tomat yang paling efektif dalam menurunkan aktivitas kedua parameter tersebut. Penelitian ini menggunakan rancangan acak lengkap dengan lima kelompok perlakuan dan lima ulangan. Perlakuan yang dicobakan: P01 = kontrol negative, tikus diberi akuades; P02 = kontrol positif, tikus diberi air rebusan plastik hitam; P1 = tikus diberi air rebusan plastik hitam dan 5 mg/kg BB ekstrak tomat; P2 = tikus diberi air rebusan plastik hitam dan 10 mg/kg BB ekstrak tomat; dan P3 = tikus diberi air rebusan plastik hitam dan 20 mg/kg BB ekstrak tomat. Air rebusan plastik hitam dibuat dengan merebus 5 g plastik dalam 500 mL air selama 3 jam, sedangkan ekstrak tomat diperoleh melalui metode maserasi 1 kg tomat menggunakan etanol 96% dan diencerkan dengan minyak jagung sesuai dosis. Air rebusan plastik hitam diberikan terlebih dahulu, diikuti ekstrak tomat setelah jeda 3 jam. Perlakuan diberikan secara oral menggunakan sonde dengan volume 2 mL per ekor setiap hari selama tujuh hari. Pada hari ke-8 dilakukan pengambilan darah melalui vena retro-orbital, kemudian disentrifugasi dalam tabung EDTA. Aktivitas aspartate aminotransferase dan alanine aminotransferase diukur menggunakan spektrofotometer pada panjang gelombang 340 nm. Data dianalisis menggunakan ANOVA dengan taraf kesalahan 5%, dan apabila terdapat perbedaan nyata (p < 0,05), analisis dilanjutkan dengan uji Duncan karena antar perlakuan berbeda nyata. Hasil penelitian menunjukkan bahwa paparan air rebusan plastik hitam meningkatkan aktivitas aspartate aminotransferase dan alanine aminotransferase secara signifikan dibandingkan kontrol negatif. Pemberian ekstrak tomat mampu menurunkan aktivitas kedua enzim tersebut. Dosis 10 mg/kgBB dan 20 mg/kgBB menunjukkan penurunan yang mendekati kondisi normal dan tidak berbeda nyata, sedangkan dosis 5 mg/kgBB belum optimal. Berdasarkan hasil tersebut, ekstrak tomat berpotensi sebagai agen hepatoprotektif alami, dengan dosis 10 mg/kgBB sebagai dosis paling efektif dan efisien dalam menurunkan aktivitas aspartate aminotransferase dan alanine aminotransferase akibat paparan air rebusan plastik hitam. |

Inggris) | Black plastic made from polycarbonate is widely used for food packaging and household needs, but it contains additives such as bisphenol A, phthalates, and heavy metals (lead, cadmium, and chromium) that can migrate into food or beverages when heated or reused, potentially causing oxidative stress and liver damage. This damage is indicated by increased activity of aspartate aminotransferase and alanine aminotransferase in the blood. One effort to reduce these toxic effects is the administration of natural compounds with high antioxidant activity, such as tomato (Solanum lycopersicum L.), which contains lycopene, flavonoids, and vitamin C. This study aimed to determine the activity of aspartate aminotransferase and alanine aminotransferase in male white rats exposed to boiled black plastic water and given tomato extract, as well as to determine the most effective dose of tomato extract in reducing the activity of these parameters. This study used a completely randomized design with five treatment groups and five replications. The treatments were: P01 = negative control, rats were given distilled water; P02 = positive control, rats were given boiled black plastic water; P1 = rats were given boiled black plastic water and 5 mg/kg BW of tomato extract; P2 = rats were given boiled black plastic water and 10 mg/kg BW of tomato extract; and P3 = rats were given boiled black plastic water and 20 mg/kg BW of tomato extract. Boiled black plastic water was prepared by boiling 5 g of plastic in 500 mL of water for 3 hours, while tomato extract was obtained through maceration of 1 kg of tomatoes using 96% ethanol and diluted with corn oil according to the dosage. Boiled black plastic water was administered first, followed by tomato extract after a 3-hour interval. Treatments were given orally using a gavage at a volume of 2 mL per rat daily for seven days. On day 8, blood samples were collected via the retro-orbital vein and centrifuged in EDTA tubes. The activity of aspartate aminotransferase and alanine aminotransferase was measured using a spectrophotometer at a wavelength of 340 nm. Data were analyzed using ANOVA at a 5% significance level, and if significant differences were found (p < 0.05), the analysis was continued with Duncan’s test. The results showed that exposure to boiled black plastic water significantly increased the activity of aspartate aminotransferase and alanine aminotransferase compared to the negative control. Administration of tomato extract was able to reduce the activity of both enzymes. Doses of 10 mg/kg BW and 20 mg/kg BW showed reductions close to normal conditions and were not significantly different, while the 5 mg/kg BW dose was not optimal. Based on these results, tomato extract has potential as a natural hepatoprotective agent, with a dose of 10 mg/kg BW being the most effective and efficient in reducing aspartate aminotransferase and alanine aminotransferase activity due to exposure to boiled black plastic water. |
