| Abstrak (Bhs. Indonesia) | Ganoderma lucidum telah banyak digunakan sebagai obat tradisional karena senyawa bioaktif yang terkandung di dalamnya seperti triterpenoid. Senyawa bioaktif tersebut dapat digunakan sebagai immunomodulator, antivirus, antioksidan, antikanker, antitumor, dan antiinflamasi. Rheumathoid arthritis merupakan penyakit autoimun yang ditandai dengan inflamasi sistemik dan progresif pada sendi. Inflamasi merupakan respon sistem imun terhadap infeksi atau luka jaringan yang bertujuan untuk mencegah penyebaran infeksi lebih lanjut serta mengatur perbaikan jaringan. Saat terjadinya inflamasi kandungan asam urat akan meningkat karena aktivitas enzim xantin oksidase (XO). Neutrofil merupakan leukosit pertama yang bermigrasi ke lokasi inflamasi untuk memfagosit bahan asing. Perawatan RA seringkali menggunakan obat golongan Nonsteroidal anti-inflammatory drugs (NSAIDs) yang sering kali menimbulkan efek samping seperti komplikasi gastrointestinal dan hipertensi. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian ekstrak etanol G. lucidum terhadap kadar asam urat dan neutrofil tikus model RA. Penelitian dilakukan secara eksperimental dengan Rancangan Acak Lengkap yang terdiri atas 6 perlakuan yakni Kontrol Sehat tikus tanpa induksi CFA, Kontrol Negatif tikus diinduksi CFA, Kontrol Positif tikus diinduksi CFA dan diberi natrium diklofenak 0,012 g.200g-1 BB, Perlakuan 1 tikus diinduksi CFA dan diberikan dosis ekstrak etanol G. lucidum 250mg.kg-1 BB, Perlakuan 2 tikus diinduksi CFA dengan dosis ekstrak etanol G. lucidum 500mg.kg-1 BB dan Perlakuan 3 tikus diinduksi CFA dengan dosis ekstrak etanol G. lucidum 750 mg.kg-1 BB. Setiap perlakuan dilakukan sebanyak 5 kali ulangan. Variabel bebas yang digunakan yaitu variasi dosis ekstrak G. lucidum dan variabel terikat yaitu perubahan kadar asam urat dan jumlah neutrofil darah tikus. Parameter yang diamati dalam penelitian ini adalah kadar asam urat dan jumlah neutrofil darah tikus. Pada hari ke-21 dilakukan pengambilan darah tikus untuk mengukur kadar asam urat dan menghitung jumlah total neutrofil. Data hasil pengukuran kadar asam urat dan neutrofil dianalisis menggunakan ANOVA dengan tingkat kesalahan 5% kemudian dilanjutkan dengan uji Duncan. Hasil penelitian menunjukkan bahwa semua dosis ekstrak etanol G. lucidum dapat menurunkan kadar asam urat dan jumlah neutrofil pada tikus model Rheumathoid arthritis (RA). Ekstrak etanol G. lucidum dengan dosis 250 mg.kg-1 BB merupakan dosis yang paling efektif dalam menurunkan kadar asam urat sebesar 17% dengan rata-rata 3,69 ± 0,23 mg.dL-1 dan dosis 750 mg.kg-1 BB merupakan dosis yang efektif dalam menurunkan neutrofil sebesar 62% dengan jumlah total 2.126 ± 735 sel/µL. |
| Abtrak (Bhs. Inggris) | Ganoderma lucidum has been widely used as a traditional medicine due to its bioactive compounds, such as triterpenoids. These bioactive compounds can be used as immunomodulators, antivirals, antioxidants, anticancer, antitumor, and anti-inflammatory agents. Rheumatoid arthritis is an autoimmune disease characterized by systemic and progressive inflammation of the joints. Inflammation is an immune system response to infection or tissue injury, aimed at preventing further spread of infection and regulating tissue repair. During inflammation, uric acid levels increase due to the activity of the enzyme xanthine oxidase (XO). Neutrophils are the first leukocytes to migrate to the site of inflammation to phagocytize foreign substances. RA treatment often uses nonsteroidal anti-inflammatory drugs (NSAIDs), which often cause side effects such as gastrointestinal complications and hypertension. This study aims to determine the effect of ethanol extract of G. lucidum on uric acid level and neutrophil count in RA model rats. The study was conducted experimentally with a Completely Randomized Design consisting of 6 treatments of Healthy Control, rats induced by CFA as negative control, rats induced by CFA with Na-diclofenac administration as Positive Control, and rats induced by CFA with G. lucidum extract administration with a dose of 250, 500, 750 mg.kg-1 BW. Each treatment was repeated 5 times. The independent variable used was the variation of the dose of G. lucidum extract, while dependent variables were the changes in uric acid levels and the number of neutrophils in rat blood. The parameters observed in this study were uric acid levels and the number of neutrophils in rats. On the 21st day, blood was collected from the rats to measure uric acid levels and count the total number of neutrophils. Uric acid and neutrophil measurement data were analyzed using ANOVA with a 5% margin of error, followed by Duncan's test. The results showed that all doses of G. lucidum ethanol extract reduced uric acid levels and neutrophil counts in RA model rats. The 250 mg.kg-1 BW of G. lucidum ethanol extract was the most effective dose in reducing uric acid levels by 17% with an average of 3,69 ± 0,23 mgdL-1 and a dose of 750 mg.kg-1 BW is an effective dose in reducing neutrophil count by 62% with a total of 2.126 ± 735 cell/µL. |
