Abstrak (Bhs. Indonesia)Penyakit antraknosa menyebabkan kerugian hasil panen mencapai hingga 80% jika kondisi lingkungan mendukung pertumbuhan patogen. Salah satu cara untuk mengendalikan penyakit ini yaitu dengan menggunakan agensia hayati berupa Pseudomonas fluorescens yang berasal dari tanah sekitar tanaman putri malu. Penelitian ini bertujuan untuk mengisolasi bakteri rhizosfer Pseudomonas fluorescens, mengetahui potensi sebagai antagonis patogen, dan sebagai Plant Growth Promoting Rizobacteria, serta efeketivitasnya dalam mengendalikan penyakit antraknosa. Penelitian dimulai dengan eksplorasi pada tanah sekitaran putri malu, kemudian di isolasi pada medium padat dan dikarakter berdasarkan bentuk koloni, sel, dan biokimia. Isolat Pseudomonas fluorescens kemudian diuji terhadap Colletotrichum capsici, Fusarium oxysporum, dan Rhizoctonia solani serta bakteri Xanthomonas oryzae dan Ralstonia solanacearum. Pengujian PGPR dilakukan pada benih mentimun dengan 3 isolat terbaik diaplikasikan pada buah cabai rawit, diamati kemampuan menghambat penyakit dengan 5 ulangan dan kontrol serta fungisida. Rancangan yang digunakan adalah RAL. Variabel berupa karakterisasi biokimia, uji antagonis, PGPR data in vivo cabai berupa luas serangan, indeks sampah, dan kadar gula. Hasil eksplorasi diperoleh 13 isolat dengan karakter sel berbentuk batang, Gram negatif, uji katalase dan oksidase positif. Uji antagonis dan PGPR diperoleh 3 isolat P. fluorescens terbaik. Perlakuan Clp 1.1; Clp 1.2; dan Clp 1.4 memiliki persentase efektivitas panjang akar terhadap kontrol masing-masing 25,10; 46,6; 42,3; dan 57,35. Isolat Clp 1.1 menunjukkan hasil paling efektif dalam mengendalikan antraknosa pada buah cabai rawit pascapanen dengan menekan masa inkubasi sebesar 68,71 %, luas serangan efektivitas sebesar 67,34 %, serta indeks sampah 71,41 % dibandingkan kontrol.
Abtrak (Bhs. Inggris)Anthracnose is a major disease in chili that can cause up to 80% yield loss under favorable environmental conditions. This study aimed to isolate rhizospheric Pseudomonas fluorescens from the soil surrounding sensitive-creeping plant, evaluate its potential as an antagonist to plant pathogens, its role as Plant Growth-Promoting Rhizobacteria (PGPR), and its effectiveness in controlling postharvest anthracnose on bird’s eye chili fruit (Capsicum frutescens). Soil samples were collected, and bacterial isolation was performed on solid medium, followed by morphological and biochemical characterization. The isolates were tested against fungal pathogens (Colletotrichum capsici, Fusarium oxysporum, Rhizoctonia solani) and bacterial pathogens (Xanthomonas oryzae, Ralstonia solanacearum). PGPR activity was assessed using cucumber seeds. The three most promising isolates (Clp 1.1, Clp 1.2, Clp 1.4) were applied to chili fruit to assess their postharvest disease suppression ability, using a Completely Randomized Design with five replications, a control, and a fungicide treatment. Variables observed included biochemical characterization, antagonistic ability, PGPR parameters, and in vivo data from chili fruits such as disease severity, waste index, and sugar content. Thirteen isolates were obtained, characterized as Gram-negative rods with positive catalase and oxidase activity. Clp 1.1, Clp 1.2, and Clp 1.4 showed pathogen inhibition above 50% and promoted root growth by 25.10%, 46.6%, and 42.3%, respectively. Clp 1.1 showed the strongest inhibition against C. capsici (66.65%) and was the most effective in controlling postharvest anthracnose, reducing the incubation period by 68.71%, disease severity by 67.34%, and waste index by 71.41% compared to the control.
