Abstrak (Bhs. Indonesia)Diabetes melitus (DM) merupakan gangguan metabolisme yang ditandai hiperglikemia akibat kekurangan atau resistensi insulin, memicu inflamasi. Salah satu mediator inflamasi adalah sitokin IL-1ß. Penelitian ini bertujuan mengetahui pengaruh ekstrak etanol jamur tiram coklat (Pleurotus cystidiosus) terhadap kadar IL-1ß pada tikus model diabetes, serta seberapa besar penurunan kadar IL-1ß yang terjadi. Penelitian menggunakan metode eksperimental dengan Rancangan Acak Lengkap. Tikus dibagi menjadi enam kelompok: kontrol sehat (KS), kontrol negatif (KN, diinduksi STZ), kontrol positif (KP, diinduksi STZ + metformin), serta tiga kelompok perlakuan (P1, P2, P3) yang mendapatkan ekstrak etanol P. cystidiosus pada dosis 250, 500, dan 750 mg/kg BB. Variabel bebas adalah variasi dosis ekstrak, sedangkan variabel terikat adalah kadar IL-1ß. Parameter utama yang diukur adalah kadar IL-1ß, didukung berat badan tikus, kadar gula darah, dan identifikasi senyawa bioaktif jamur. Hasil penelitian menunjukkan ekstrak etanol P. cystidiosus dosis 500 mg/kg BB (P2) efektif menurunkan kadar IL-1ß secara signifikan, dengan rata-rata 11,46 ng/L. Nilai ini mendekati kadar normal kelompok sehat (KS: 10–35,07 ng/L). Temuan ini membuktikan ekstrak etanol P. cystidiosus mampu menurunkan inflamasi pada tikus model diabetes melalui penurunan kadar IL-1ß.
Abtrak (Bhs. Inggris)Diabetes mellitus (DM) is a macromolecular metabolic disorder characterized by hyperglycemia due to insulin deficiency or resistance, which in turn triggers inflammation. Interleukin-1β (IL-1β) is one of the key inflammatory mediators in DM. This study aims to investigate the effects of ethanol extract from brown oyster mushrooms (Pleurotus cystidiosus) on IL-1β levels in diabetic mouse models and the extent of its reduction. The research method employed a Completely Randomized Design (CRD) with six groups of mice: healthy control (HC), negative control (NC) induced by streptozotocin (STZ), positive control (PC) induced with STZ and administered metformin, and three treatment groups (P1, P2, P3) receiving ethanol extract of P. cystidiosus at doses of 250, 500, and 750 mg/kg body weight (BW). The variation in extract dose was the independent variable, while IL-1β levels were the primary dependent variable. Supporting parameters included body weight, blood glucose levels in mice, and identification of bioactive compounds in the mushroom. The results showed that administration of P. cystidiosus ethanol extract at a dose of 500 mg/kg BW (P2) significantly reduced IL-1ß levels to an average of 11.46 ng/L, approaching the normal levels of healthy mice, which range around 10 ng/L. In comparison, the healthy control group (KS) had IL-1ß levels of 35.07 ng/L. This reduction indicates the potential of ethanol extract from brown oyster mushrooms in reducing inflammation in diabetic mouse models. In conclusion, P. cystidiosus ethanol extract effectively reduces IL-1ß inflammatory cytokine levels, thereby acting as an anti-inflammatory agent in diabetes mellitus. This study opens up opportunities for the development of brown oyster mushrooms as a supportive therapeutic agent for controlling inflammation in DM.
