Abstrak (Bhs. Indonesia)Eksplorasi metabolit sekunder Penicillium citrinum terus berkembang karena potensinya dalam menghasilkan senyawa kandidat antibiotik. Strategi OSMAC (One Strain Many Compounds) melalui penambahan NaBr terbukti memperkaya profil metabolit dan menghasilkan senyawa aktif baru. Untuk mengidentifikasi senyawa potensial, digunakan pendekatan metabolomik tidak tertarget berbasis LC-MS/MS, serta evaluasi aktivitas antibakteri terhadap Staphylococcus epidermidis. P. citrinum difermentasi pada media beras selama 30 hari dalam dua kondisi: dengan dan tanpa OSMAC, kemudian diekstraksi dengan etil asetat. Ekstrak difraksinasi menggunakan metanol dan n-heksan, dianalisis menggunakan LC-MS/MS, serta diuji aktivitas antibakterinya secara in vitro. Senyawa antibakteri potensial diidentifikasi melalui kajian literatur. Hasil menunjukkan hanya terdapat tiga senyawa yang sama antara ekstrak etil asetat dari kedua perlakuan, menandakan keberhasilan OSMAC dalam memperkaya metabolit. Seluruh sampel menunjukkan aktivitas antibakteri dengan zona hambat 8,33–13,5 mm. Berdasarkan literatur, fraksi metanol OSMAC mengandung senyawa antibakteri terbanyak. Strategi OSMAC terbukti meningkatkan keragaman metabolit dan menghasilkan ekstrak dan fraksi yang aktif terhadap S. epidermidis.
Abtrak (Bhs. Inggris)The exploration of secondary metabolites from Penicillium citrinum continues to grow due to its potential to produce antibiotic candidate compounds. The OSMAC (One Strain Many Compounds) strategy, involving the addition of NaBr, has been proven to enrich metabolite profiles and generate novel active compounds. A non-targeted metabolomic approach based on LC-MS/MS was used to identify potential compounds, alongside antibacterial evaluation against Staphylococcus epidermidis. P. citrinum was fermented on rice medium for 30 days under two conditions: with and without OSMAC, then extracted with ethyl acetate. The extract was fractionated using methanol and n-hexane, analyzed by LC-MS/MS, and tested for antibacterial activity in vitro. Potential antibacterial compounds were identified through literature review. Results showed only three shared compounds between ethyl acetate extracts of both treatments, indicating successful metabolite enrichment by OSMAC strategy. All samples exhibited antibacterial activity with inhibition zones of 8.33 to 13.5 mm. Based on the literature review, the OSMAC methanol fraction contained the highest number of antibacterial compounds. OSMAC strategy effectively enhanced metabolite diversity and yielded active extracts and fractions against S. epidermidis.
