| Abstrak (Bhs. Indonesia) | Latar Belakang : Pencarian kandidat antibakteri dari jamur endofit Penicillium citrinum semakin pesat karena kemampuanya menghasilkan keragaman senyawa bioaktif. Pendekatan ko-kultur P. citrinum dengan Sinomicrobium sp. PAP.21 telah mampu menghasilkan diversitas metabolit sekunder, seperti senyawa golongan alkaloid, flavonoid, terpenoid, dan steroid. Akan tetapi, adanya urgensi mengidentifikasi struktur kimia senyawa tersebut, sedangkan tahap isolasi dan identifikasi senyawa membutuhkan waktu yang lama. Maka dari itu, dilakukan kajian untargeted metabolomic dengan LC-MS/MS dalam pencarian senyawa yang berpotensi sebagai antibakteri dan mengevaluasi aktivitas antibakteri secara in vitro terhadap Staphylococcus epidermidis. Metodologi : Jamur P. citrinum, bakteri Sinomicrobium sp. PAP.21, dan ko-kultur keduanya yang difermentasi pada media beras selama 40 hari. Sampel diekstraksi dengan etil asetat dan sampel ekstrak ko-kultur difraksinasi dengan metanol dan n-heksan. Keempat sampel diukur dengan LC-MS/MS menghasilkan puncak kromatogram dan m/z untuk analisis metabolomik. Selanjutnya, semua sampel diuji aktivitas antibakterinya dengan metode difusi cakram terhadap S. epidermidis. Hasil Penelitian : Analisis metabolomik menghasilkan tujuh puncak khas yang hanya terdapat pada ekstrak ko-kultur, meliputi enam senyawa terdereplikasi dan satu senyawa unknown. Ekstrak etil asetat dan fraksi metanol ko-kultur juga menunjukkan aktivitas antibakteri terhadap S. epidermidis yang didominasi senyawa potensial antibakteri dari fraksi metanol ko-kultur berdasarkan penelusuran literatur. Kesimpulan : Pendekatan metode ko-kultur mampu menghasilkan keberagaman senyawa yang terbukti memiliki aktivitas antibakteri terhadap bakteri Gram positif |
| Abtrak (Bhs. Inggris) | Background : The search for new antibacterial compounds from the endophytic fungus Penicillium citrinum is emerging due to its ability to produce bioactive compounds. The co-culture of P. citrinum and Sinomicrobium sp. PAP.21 has successfully diversified secondary metabolites, including alkaloids, flavonoids, terpenoids, and steroids Furthermore, it is necessary to determine the chemical structure of the compounds, however the isolation and identification of those are time-consuming. Accordingly, this study applies untargeted metabolomics by using LC-MS/MS in order to determine antibacterial compounds from that co-culture, and to evaluate in vitro antibacterial activity against Staphylococcus epidermidis. Methods : P. citrinum, Sinomicrobium sp. PAP.21, and both co-cultures were fermented on solid rice media for 40 days. Subsequently, samples were extracted with ethyl acetate and co-culture extract was fractionated with methanol and n-hexane. All samples were employed to LC-MS/MS yielding chromatogram peaks and m/z for further metabolomics analysis. Then, samples were assayed for its antibacterial activity against S. epidermidis. Results : Metabolomic analysis revealed seven unique peaks appeared only in the co-culture extract, including six dereplicated compounds and one unknown compound. The ethyl acetate extract and methanol fraction of co-culture also showed antibacterial activity against S. epidermidis. Furthermore, the most putative antibacterial compounds based on the literature study were found in the methanol fraction of co-culture. Conclusion : The co-culture approach is able to diversify fungal bioactive compounds, as evident by those that reveal antibacterial potential against Gram-positive bacteria. |
