| Judul Artikel | Efek Ekstrak Etanolik Kayu Manis (Cinnamomum burmannii) Terhadap Histopatologi Pankreas dan Ekspresi GLUT 4 Tikus Terinduksi Pakan Tinggi Lemak dan Streptozotocin |
| Abstrak (Bhs. Indonesia) | Latar Belakang: Diabetes Melitus tipe II ditandai dengan hiperglikemia. Penurunan fungsi atau kerusakan sel pankreas menyebabkan gangguan sekresi insulin. Tanaman yang mengandung senyawa polifenol, flavonoid, steroid dan terpenoid seperti kayu manis (Cinnamomum burmannii) berfungsi sebagai antioksidan dan anti diabetes melitus. Penelitian ini bertujuan mengetahui gambaran histopatologi dan ekspresi GLUT 4 pada pankreas tikus yang diinduksi pakan tinggi lemak dan STZ. Metodologi: Penelitian ini merupakan ekperimen laboratorium Post-Test Control Group Design menggunakan 25 ekor tikus putih jantan yang dibagi ke dalam 5 kelompok. Kelompok I (kontrol normal), kelompok II (metformin), kelompok III (kayu manis dosis 100 mg/kgBB), kelompok IV (kayu manis dosis 200 mg/kgBB), kelompok V (kayu manis dosis 400 mg/kgBB). Dilakukan pengamatan pada perubahan histopatologi pankreas dengan metode skoring setelah dilakukan pewarnaan Hemaatoxylin-Eosin serta data dianalisis secara deskriptif setelah dilakukan pewarnaan Imunohistokimia. Hasil Penelitian: Hasil penelitian menunjukkan bahwa pemberian pakan tinggi lemak dan streptozotocin dapat menyebabkan kondisi diabetes melitus pada tikus. Pemberian kayu manis dosis 100 mg/kgBB memberikan kenaikan berat badan tikus paling tinggi dibandingkan pemberian metformin, kayu manis dosis 200 mg/kgBB, dan 400 mg/kgBB. Dosis 100 mg/kgBB dapat memperbaiki pankreas paling tinggi ditunjukkan dengan batas sel yang jelas, tidak adanya nekrosis serta peningkatan ekspresi GLUT 4 pada sitoplasma sel pankreas dengan warna coklat kegelapan pekat, lebih tegas dan seragam. Kesimpulan: Pemberian ekstrak etanolik kayu manis (Cinnamomum burmannii) berpotensi memperbaiki profil histopatologi organ pankreas yang terdapat kemiripan dengan kontrol sehat serta meningkatkan terjadinya ekspresi GLUT 4 pada sitoplasma sel organ pankreas yang terdapat kemiripan dengan kelompok pemberian metformin. |
| Abtrak (Bhs. Inggris) | Background: Type II Diabetes Melitus is characterized by hyperglycemia. Reduced function or damage to pancreatic beta-cells leads to impaired insulin secretion. Plants containing compounds such as polyphenols, flavonoids, steroids, and terpenoids, such as cinnamon (Cinnamomum burmannii), function as antioxidant and antidiabetic agents. This research aims to determine the histopathological features and glut-4 expression in the pancreas of male wistar rats induced with a high fat diet and streptozotocin (STZ). Methodology: This study used a laboratory experiment design with Post-Test Control Group. Twenty-five male wistar rats were utilized and divided into five groups: Group I (normal control), Group II (metformin treatment), Group III (cinnamon at a dose of 100 mg/kg body weight), Group IV (cinnamon at a dose of 200 mg/kg body weight), and Group V (cinnamon at a dose of 400 mg/kg body weight). Observation were conducted on pancreatic histopathological changes using a scoring method following after Hematoxylin-Eosin staining, and data were analyzed descriptively after Immunohistochemisstry staining. Result: The results from the study indicate that the administration of a high fat and streptozotocin can cause diabetes mellitus condition in rats. Cinnamon administration at a dosage of 100 mg/kg body weight resulted in the highest increase of body weight in rats compared to the administration of metformin and cinnamon at dosages of 200 mg/kg and 400 mg/kg body weight. The dosage of 100 mg/kgBB demonstrated the most significant improvement in pancreatic tissue characterized by distinct cell borders, the absence of necrosis, and cellular regeneration. In addition, the increased expression of GLUT 4 in the cytoplasm of pancreatic cells was characterized by a strong, dark brown staining that appeared more distinct and uniform. Conclusion: The administration of ethanolic extract of Cinnamomum burmannii demonstrates potential; in improving the histopathological profile of pancreatic tissue, closely resembling that of the healthy control group, and in enhancing GLUT 4 expression within the cytoplasm of pancreatic cells, comparable to the effects observed in the metformin treated group. |
