| Abstrak (Bhs. Indonesia) | Latar Belakang. Periodontitis merupakan penyakit inflamasi kronis yang menyebabkan kerusakan pada jaringan periodontal. Gold standard perawatan periodontitis adalah scalling and root planning (SRP) dan gel asam hialuronat 0,2% sebagai terapi adjuvant. Gel asam hialuronat 0,2% yang digunakan berkepanjangan, akan menimbulkan reaksi alergi. Daun katuk mampu dijadikan sebagai alternatif terapi adjuvant karena mengandung senyawa fitokimia seperti flavonoid, saponin, tanin, alkaloid, dan steroid, yang memiliki sifat antiinflamasi. Tujuan. Penelitian ini bertujuan untuk mengetahui pengaruh topikal gel ekstrak etanol daun katuk konsentrasi 6%, 8%, dan 10% terhadap jumlah fibroblas dan kepadatan kolagen pada proses penyembuhan periodontitis tikus Wistar hari ke-1, ke-3, dan ke-7. Metode. Penelitian true experimental laboratoris secara in vivo dengan posttest control group design. Sampel sebanyak 62 ekor tikus Wistar jantan. Sampel dibagi menjadi 5 kelompok yaitu kontrol negatif gel CMC-Na tanpa ekstrak daun katuk (KN), kontrol positif gel asam hialuronat 0,2% (KP), perlakuan gel ekstrak etanol daun katuk konsentrasi 6% (P1), 8% (P2), dan 10% (P3). Pengamatan dilakukan pada hari ke-1, 3, dan 7. Data jumlah fibroblas dilakukan uji Two-Way ANOVA dan uji Post Hoc LSD. Data kepadatan kolagen dilakukan uji Kruskal-Wallis dan uji Post-Hoc Mann Whitney. Data Jumlah fibroblas dan kepadatan kolagen dilakukan uji Pearson Correlation. Hasil. Data jumlah fibroblas terdapat perbedaan signifikan (p≤0,05) antara kelompok P1, P2, dan P3 dengan kelompok KN pada hari ke-1, 3, dan 7. Data kepadatan kolagen menunjukkan terdapat perbedaan signifikan (p≤0,05) antara kelompok P1, P2, dan P3 dengan kelompok KN pada hari ke-7. Hasil uji korelasi menunjukkan bahwa terdapat hubungan bermakna (p≤0,05) antara jumlah fibroblas dan kepadatan kolagen. Simpulan. Topikal gel ekstrak etanol daun katuk konsentrasi 6% (P1) memiliki efektivitas paling baik dalam meningkatkan jumlah fibrolas dan kepadatan kolagen pada hari ke-1, 3, dan 7. |
| Abtrak (Bhs. Inggris) | Background. Periodontitis is a chronic inflammatory disease that causes damage to the periodontal tissue. The gold standard for periodontitis treatment is scaling and root planning (SRP) and 0.2% hyaluronic acid gel as adjuvant therapy. Prolonged use of 0.2% hyaluronic acid gel will cause allergic reactions. Katuk leaves can be used as an alternative adjuvant therapy because they contain phytochemical compounds such as flavonoids, saponins, tannins, alkaloids, and steroids, which have anti-inflammatory properties. Purpose. This study aims to determine the effect of topical katuk leaf ethanol extract gel with concentrations of 6%, 8%, and 10% on the number of fibroblasts and collagen density in the healing process of periodontitis in Wistar rats on days 1, 3, and 7. Method. True experimental laboratory research in vivo with posttest control group design. Samples were 62 male Wistar rats. The samples were divided into 5 groups, namely negative control CMC-Na gel without katuk leaf extract (KN), positive control hyaluronic acid gel 0.2% (KP), treatment of katuk leaf ethanol extract gel concentration of 6% (P1), 8% (P2), and 10% (P3). Observations were made on days 1, 3, and 7. Data on the number of fibroblasts were tested using Two-Way ANOVA and Post Hoc LSD tests. Data on collagen density were tested using Kruskal-Wallis and Post-Hoc Mann Whitney tests. Data on the number of fibroblasts and collagen density were tested using Pearson Correlation tests. Results. Data on the number of fibroblasts showed a significant difference (p≤0.05) between groups P1, P2, and P3 with the KN group on days 1, 3, and 7. Data on collagen density showed a significant difference (p≤0.05) between groups P1, P2, and P3 with the KN group on days 7. The results of the correlation test showed that there was a significant relationship (p≤0.05) between the number of fibroblasts and collagen density. Conclusion. Topical gel of ethanol extract of katuk leaves with a concentration of 6% (P1) had the best effectiveness in increasing the number of fibroblasts and collagen density on days 1, 3, and 7. |
