| Abstrak (Bhs. Indonesia) | Latar Belakang — Artritis reumatoid (RA) merupakan penyakit autoimun kronis yang ditandai dengan adanya inflamasi pada sendi. Patofisiologi RA melibatkan TNF-α yang berperan dalam proses inflamasi dan proses osteoklastogenesis. Salah satu pengobatan yang saat ini sering digunakan dalam terapi RA adalah natrium diklofenak. Jahe, kunyit, dan seledri mengandung senyawa aktif yang dapat berpotensi menurunkan produksi TNF-α sehingga bahan tersebut dapat menurunkan progresivitas penyakit RA. Tujuan — Mengetahui efek kombinasi ekstrak kunyit, jahe, dan seledri terhadap kadar TNF-α pada tikus putih yang diinduksi Complete Freund’s Adjuvant (CFA). Metode Penelitian — Penelitian ini merupakan penelitian eksperimental dengan desain post-test only with control group. Penelitian ini menggunakan 28 tikus yang dibagi secara random menjadi 4 kelompok, yaitu kelompok A (kontrol sehat), kelompok B (kontrol sakit, diberikan CFA 0,1 ml intraplantar), kelompok C (kontrol positif, diberikan CFA 0,1 ml intraplantar kemudian diberikan natrium diklofenak 2 ml per oral selama 14 hari), dan kelompok D (perlakuan, diberikan CFA 0,1 ml intraplantar kemudian diberikan kombinasi ekstrak jahe 200 mg/kgBB, kunyit 60 mg/kgBB, dan seledri 250 mg/kgBB selama 14 hari). Kadar TNF-α diukur menggunakan ELISA dan dianalisis menggunakan One-way ANOVA dilanjutkan dengan uji Post-hoc LSD. Hasil — Hasil penelitian menunjukkan bahwa rerata peningkatan TNF-α yang paling tinggi didapatkan pada kelompok D (227,10), diikuti kelompok B (193,90), kelompok C (180,85), dan kelompok A (109,25). Hasil uji Post-hoc LSD menunjukkan bahwa terdapat perbedaan yang signifikan antara kelompok 1 dengan kelompok lainnya (p<0,05). Sedangkan antar kelompok 2, 3, dan 4 tidak memiliki perbedaan signifikan (p>0,05). Kesimpulan — Tidak terdapat pengaruh yang signifikan pemberian kombinasi ekstrak jahe 200 mg/kgBB, kunyit 60 mg/kgBB, dan seledri 250 mg/kgBB terhadap kadar TNF-α pada tikus putih yang diinduksi Complete Freund’s Adjuvant (CFA). |

| Abtrak (Bhs. Inggris) | Background — Rheumatoid arthritis (RA) is a chronic autoimmune disease characterized by inflammation of the joints. The pathophysiology of RA involves TNF-α which plays a role in the inflammatory process and osteoclastogenesis. One of the treatments currently often used in RA therapy is sodium diclofenac. Ginger, turmeric, and celery contain active compounds that can potentially reduce the production of TNF-α so that these ingredients can reduce the progression of RA. Objective— To determined the effect of a combination of turmeric, ginger, and celery extracts on TNF-α levels in white mice induced by Complete Freund's Adjuvant (CFA). Research Method — This study was an experimental study with a post-test only with control group design. This study used 28 rats randomly divided into 4 groups, namely group A (healthy control), group B (sick control, given 0.1 ml CFA intraplantar), group C (positive control, given 0.1 ml CFA intraplantar then given 2 ml diclofenac sodium orally for 14 days), and group D (treatment, given 0.1 ml CFA intraplantar then given a combination of ginger extract 200 mg/kgBW, turmeric 60 mg/kgBW, and celery 250 mg/kgBW for 14 days). TNF-α levels were measured using ELISA and analyzed using the One-way ANOVA followed by the Post-hoc LSD test. Results— The results showed that the highest average increase in TNF-α was obtained in group D (227,10), followed by group B (193,90), group C (180,85), and group A (109,25). The results of the Post-hoc LSD test showed that there was a significant difference between group 1 and the other groups (p<0,05). Meanwhile, there was no significant difference between groups 2, 3, and 4 (p>0,05). Conclusion — There was no effect of giving a combination of ginger extract 200 mg/kgBW, turmeric 60 mg/kgBW, and celery 250 mg/kgBW on TNF-α levels in white mice induced by Complete Freund's Adjuvant (CFA). |
