| Abstrak (Bhs. Indonesia) | Penelitian ini bertujuan untuk mengetahui pengaruh penggunaan percoll sebagai medium separasi seks dan lama inkubasi terhadap konsentrasi dan mortalitas spermatozoa pejantan Kambing Peranakan Etawah. Materi yang digunakan dalam penelitian adalah kambing Peranakan Etawah jantan berusia 3 tahun yang berasal dari experimental farm, Universitas Jenderal Soedirman. Bahan yang digunakan yaitu NaCl fisiologis, eosin-nigrosin, gradien percoll. Alat yang digunakan yaitu, centrifuge, tabung centrifuge, mikroskop, cover glass, object glass, waterbath, termos es, vagina tiruan, mikropipet, pipet, glass beaker, spuit, microtube, stopwatch, kertas label, Erlenmeyer, pemanas Bunsen, dan bilik thoma. Metode penelitian yang digunakan adalah rancangan acak kelompok RAK) pola faktorial 2x2x3 dengan faktor fraksi atas dan bawah, faktor gradien dua level (g1:40:55 dan g2:45:60) dan lama inkubasi yang terdiri dari tiga level (30,45, dan 60 menit). Metode analisis menggunakan analisis variansi dan dilanjutkan dengan uji Duncan’s Multiple Range test. Hasil penelitian Spermatozoa dengan konsentrasi paling tinggi berada di fraksi atas dengan gradien 45:60 dan Lama inkubasi 30 -60 menit. Spermatozoa dengan persentase mortalitas paling rendah berada di fraksi bawah dengan gradien 45:60 dan Lama inkubasi 45 menit, Spermatozoa Kambing Peranakan Etawah hasil separasi seks memiliki konsentrasi dan persentase mortalitas yang tidak jauh berbeda dan layak digunakan untuk program inseminasi buatan. kesimpulan dari penelitian ini Spermatozoa dengan konsentrasi tertinggi berada di fraksi atas pada gradien 45:60% dengan waktu inkubasi 30-60 menit, dan hasil ini masih sesuai dengan standar inseminasi buatan. Sementara itu, spermatozoa dengan persentase mortalitas terendah terdapat di fraksi bawah pada gradien 45:60% dengan waktu inkubasi 45 menit. |

Inggris) | This study aims to determine the effect of using Percoll as a sex separation medium and the incubation duration on the concentration and mortality of male Etawah Crossbred Goat spermatozoa. The material used in the research is a 3-year-old male Etawah crossbred goat from the experimental farm of Jenderal Soedirman University. The materials used are physiological saline, eosin-nigrosin, and Percoll gradient. The tools used are a centrifuge, centrifuge tubes, microscope, cover glass, object glass, water bath, ice thermos, artificial vagina, micropipette, pipette, glass beaker, syringe, microtube, stopwatch, label paper, Erlenmeyer flask, Bunsen burner, and Thoma chamber. The research method used is a randomized block design (RBD) with a 2x2x3 factorial pattern, with upper and lower fraction factors, two-level gradient factors (g1:40:55 and g2:45:60), and incubation time consisting of three levels. (30,45, dan 60 menit). The analysis method uses variance analysis followed by the Duncan’s Multiple Range test. Research results show that spermatozoa with the highest concentration are found in the upper fraction with a gradient of 45:60 and an incubation period of 30-60 minutes. Spermatozoa with the lowest mortality percentage are found in the lower fraction with a gradient of 45:60 and an incubation period of 45 minutes. Spermatozoa from Etawah Crossbreed goats resulting from sex separation have similar concentration and mortality percentage and are suitable for artificial insemination programs. The conclusion of this study is that spermatozoa with the highest concentration are found in the upper fraction at a gradient of 45:60% with an incubation time of 30-60 minutes, and these results are still in accordance with artificial insemination standards. Meanwhile, spermatozoa with the lowest mortality percentage are found in the lower fraction at a gradient of 45:60% with an incubation time of 45 minutes. |
