Penelitian ini berjudul Penggunaan Medium BSA (Bovine Serum Albumin) dengan Lama Inkubasi yang Berbeda Terhadap Morfometri dan Membran Plasma Utuh (MPU) Spermatozoa X Hasil Separasi Seks Kambing Peranakan Etawah. Tujuan penelitian dilakukan untuk mengkaji interaksi gradien BSA dan lama inkubasi terhadap morfometri dan Membran Plasma Utuh (MPU) Spermatozoa X hasil separasi seks kambing Peranakan Etawah (PE). Materi yang digunakan untuk penelitian yaitu semen kambing Peranakan Etawah (PE) jantan sebanyak 2 ekor dengan umur 3-4 tahun. Rancangan penelitian dilakukan menggunakan Rancangan Acak Kelompok (RAK) pola faktorial 2 x 3 dengan ulangan frekuensi penyadapan sebanyak 3 kali. Hasil analisis variansi perlakuan yang berpengaruh nyata kemudian dilakukan uji lanjut dengan uji Duncan’s Multiple Range Test (DMRT). Lokasi penelitian berada di Laboratorium Kesehatan Ternak dan Experimental Farm Fakultas Peternakan, Universitas Jenderal Soedirman. Hasil pengukuran morfometri rata-rata spermatozoa fraksi atas 33,91 µm2 dan fraksi bawah 30,88 µm2. Hasil dari penelitian keutuhan membran plasma spermatozoa X diperoleh rataan sebesar 70,30%. Berdasarkan analisis variasi pada pengamatan morfometri diperoleh bahwa tidak ada interaksi antara faktor gradien dan lama inkubasi (P>0,05) terhadap morfometri spermatozoa X, namun faktor gradien berpengaruh nyata (P<0,05) dan faktor lama inkubasi berpengaruh sangat nyata (P<0,01). Berdasarkan analisis variasi pada pengamatan MPU tidak ada interaksi antara gradien dan lama inkubasi (P>0,05) terhadap MPU fraksi atas namun faktor gradien dan lama inkubasi berpengaruh sangat nyata (P<0,01). Kesimpulan yang didapat dari penelitian ini adalah kualitas spermatozoa kambing PE hasil separasi seks dengan hasil terbaik untuk spermatozoa X adalah menggunakan medium BSA gradien 10:20% dengan lama waktu inkubasi 30 menit ditinjau berdasarkan MPU dan 45 menit berdasarkan keakuratan morfometri.

This study is entitled The Use of BSA (Bovine Serum Albumin) Medium with Different Incubation Times on the Morphometry and Intact Plasma Membrane (IPM) of Sperm X Results of Sex Separation of Etawah Crossbred Goats. The purpose of this study was to examine the interaction of BSA gradient and incubation times on the morphometry and Intact Plasma Membrane (IPM) of Sperm X results of sex separation of Etawah Crossbred (PE) goats. The material used for the study was 2 male Etawah Crossbred (PE) goat semen aged 3-4 years. The study design was carried out using a Randomized Block Design (RAK) factorial pattern 2 x 3 with tapping frequency replications 3 times. The results of the analysis of variance of treatments that had a significant effect were then further tested using the Duncan's Multiple Range Test (DMRT). The location of the study was in the Animal Health Laboratory and Experimental Farm, Faculty of Animal Husbandry, Jenderal Soedirman University. The results of the average morphometric measurements of the upper fraction spermatozoa were 33.91 µm2 and the lower fraction was 30.88 µm2. The results of the study on the integrity of the plasma membrane of spermatozoa X obtained an average of 70.30%. Based on the analysis of variations in morphometric observations, it was found that there was no interaction between the gradient factor and incubation time (P>0.05) on the morphometry of spermatozoa X, but the gradient factor had a significant effect (P<0.05) and the incubation time factor had a very significant effect (P<0.01). Based on the analysis of variations in the MPU observation, there was no interaction between the gradient and incubation time (P>0.05) on the MPU of the upper fraction, but the gradient factor and incubation time had a very significant effect (P<0.01). The conclusion obtained from this study is that the quality of PE goat spermatozoa from sex separation with the best results for X spermatozoa is using 10:20% gradient BSA medium with an incubation time of 30 minutes reviewed based on MPU and 45 minutes based on morphometric accuracy.
