| Judul Artikel | PENGARUH MEDIUM GRADIEN PUTIH TELUR DAN LAMA INKUBASI TERHADAP MOTILITAS DAN ABNORMALITAS SPERMATOZOA HASIL SEPARASI SEKS KAMBING PERANAKAN ETAWAH |
| Abstrak (Bhs. Indonesia) | Penelitian ini bertujuan untuk mengetahui pengaruh medium gradien putih telur dan perbedaan lama inkubasi terhadap motilitas dan abnormalitas spermatozoa kambing Peranakan Etawah. Materi yang digunakan yaitu semen dari 2 ekor pejantan kambing Peranakan Etawah, NaCl fisiologis, medium putih telur, dan eosin-negrosin. Penelitian ini dilakukan dengan menggunakan Rancangan Acak Kelompok (RAK) pola faktorial 2x3 dengan ulangan sebanyak 3 kali dan terdiri dari dua faktor yaitu faktor gradien 10:30 dan 20:40 serta lama inkubasi 30, 45, dan 60 menit. Data penelitian yang telah diperoleh dianalisis variansi dan dengan uji lanjut Duncan’s Multiple Range Test (DMRT). Hasil penelitian menunjukkan bahwa tidak ada interaksi antara medium putih telur dan perbedaan lama inkubasi terhadap motilitas (P>0,05), namun setiap faktor memberikan pengaruhnya masing- masing. Terdapat interaksi antara medium putih telur dan perbedaan lama inkubasi terhadap abnormalitas. Beradasarkan faktor gradien putih telur pada fraksi atas didapatkan nilai motilitas spermatozoa 67,08 ± 24,33% – 75,91 ± 11,7%, dan pada fraksi bawah nilai motilitas 62,3 ± 27,26% – 69,65 ± 11,87%. Nilai motilitas berdasarkan faktor lama inkubasi pada fraksi atas yaitu 66,14 ± 22,37% - 77,29 ± 4,35%, dan fraksi bawah yaitu 59,64 ± 24,5% - 72,60 ± 2,82%. Berdasarkan faktor gradien dan lama inkubasi didapatkan hasil nilai abnormalitas fraksi atas yaitu 11,67 ± 1,53% - 20 ± 2,00%, dan pada fraksi bawah yaitu 6,78 ± 1,12% - 11,85 ± 1,51%. Kesimpulan dari penelitian ini adalah motilitas spermatozoa kambing Peranakan Etawah hasil separasi seks terbaik pada gradien 10:30 dengan lama inkubasi 45 menit dan abnormalitas spermatozoa hasil separasi seks terbaik pada gradien 10:30 dengan lama inkubasi 45 menit. |

Inggris) | This study aims to determine the effect of white egg gradient medium and differences in incubation time on the motility and abnormalities of spermatozoa in Etawah Peranakan goats. The materials used were semen from 2 male Etawah crossbreed goats, white egg medium, physiological NaCl, and eosin-negrosin. This research was carried out using a Randomized Block Design (RAK) with a 2x3 factorial pattern with 3 replications and consisting of two factors, namely gradient factors of 10:30 and 20:40 and incubation times of 30, 45 and 60 minutes. The research data that was obtained was analyzed for variance and further tested by Duncan's Multiple Range Test (DMRT). The results showed that there was no interaction between egg white medium and differences in incubation time on motility (P>0.05), but each factor had its own influence. There was an interaction between egg white medium and differences in incubation time on abnormality. Based on the egg white gradient factor in the upper fraction, the sperm motility value was 67.08 ± 24.33% - 75.91 ± 11.7%, and in the lower fraction the motility value was 62.3 ± 27.26% - 69.65 ± 11.87%. The motility value based on the incubation time factor in the upper fraction was 66.14 ± 22.37% - 77.29 ± 4.35%, and the lower fraction was 59.64 ± 24.5% - 72.60 ± 2.82%. Based on the gradient factor and incubation time, the abnormality value of the upper fraction was 11.67 ± 1.53% - 20 ± 2.00%, and the lower fraction was 6.78 ± 1.12% - 11.85 ± 1.51%. The conclusion of this study is that the motility of Etawah crossbred goat spermatozoa from the best sex separation results in a gradient of 10:30 with an incubation time of 45 minutes and the abnormality of spermatozoa from the best sex separation results in a gradient of 10:30 with an incubation time of 45 minutes. |
