Abstrak (Bhs. Indonesia)Water spinach, a plant species belongs to the Convolvulaceae, is distributed and cultivated world widely for its high economic value; such as protein, calcium, phosphorus, iron, carotene, sitosterol, and vitamins A, B, and C. The current research objectives are to identify specific molecular markers with these species through RAPD analysis. Additionally, it seeks to investigate how RAPD profiles can reveal genetic polymorphism among the different Ipomoea spp. species.

This research uses a survey method with a random sampling technique. The PCR-RAPD administering 5 different primers OPA-01 OPA-10 OPB-03 OPAM-07 UBC-210. The parameters for PCR are DNA concentrations, DNA quality, and the polymorphic bands; use a broader range of RAPD primers and increase sample size. Incorporate additional Purposive Random Sampling techniques markers for validation, and apply statistical tools for in-depth analysis.

The result of the study shows that the RAPD profile of Ipomoea reptans and Ipomoea aquatica. reveal that polymorphism detected by each primer are OPA-01 produce three bands with 100% polymorphism, OPA-10 produce 1 bands with 100% polymorphism, OPB-03 produce one bands with 100% polymorphism, OPAM-7 produce three bands with 33,3% polymorphism, UBC-210 produce eleven bands with 83,3% polymorphism, With the total polymorphism, is 78,57%.
The results showed a number of amplified bands obtained from all primers is 14 bands, which consists of 3 monomorphic bands and 11 polymorphic bands, representing 83,32% of total polymorphism. This polymorphism is considered highest, generally indicates significant genetic diversity and reflects the primers ability to detect a broad range of genetic variations within the sample.
