| Abstrak (Bhs. Indonesia) | Tujuan penelitian ini yaitu mengetahui Pengaruh Penggunaan Bovine Serum Albumin Sebagai Medium Gradien Separasi Seks dan Lama Inkubasi Terhadap Viabilitas dan Tudung Akrosom Utuh (TAU) Spermatozoa Kambing Peranakan Etawah. Materi penelitian yang akan digunakan spermatozoa kambing Peranakan Etawah yang berasal dari Experimental Farm, Bahan yang digunakan yaitu NaCl fisiologis, eosin-nigrosin,gradien Bovine Serum Albumin (BSA). Alat yang digunakan yaitu, sentrifuge, tabung sentrifuge, mikroskop, cover glass, object glass, waterbath, termos, vagina tiruan, mikropipet, pipet, glass beaker, spuit, microtube, stopwatch, kertas label, Erlenmeyer, thermometer, pemanas bunsen, kompor, dan panci. Penelitian dilaksanakan di Laboratorium Kesehatan Ternak dan Experimental Farm Fakultas Peternakan Universitas Jenderal Soedirman. Metode penelitian yang akan digunakan adalah metode experimental dengan menggunakan uji RAK Faktorial 2x2x3 dengan blok frekuensi penyadapan sebanyak 3 kali dan terdiri dari tiga faktor, yaitu fraksi(F) terdiri dari dua level (atas dan bawah), gradien (G) terdiri dari dua level (g1 = 5:15 dan g2 = 10:20) dan faktor lama inkubasi (T) terdiri dari tiga level (30, 45, dan 60 menit). Hasil penelitian menunjukkan bahwa ada interaksi berpengaruh sangat nyata antara fraksi, medium gradien BSA dan perbedaan lama waktu inkubasi terhadap viabilitas dan tudung akrosom utuh (P<0,01). Berdasarkan hasil penelitian bahwa dapat disimpulkan viabilitas spermatozoa kambing Peranakan Etawah hasil separasi seks yang terbaik secara keseluruhan terdapat pada perlakuan fraksi atas gradien 5:15 lama inkubasi 60 menit dan juga perlakuan fraksi atas gradien 10:20 lama inkubasi 60 menit. Tudung akrosom utuh spermatozoa kambing Peranakan Etawah hasil separasi seks yang terbaik secara keseluruhan pada perlakuan fraksi atas gradien 10:20 lama inkubasi 30 menit, perlakuan fraksi atas gradien 10:20 lama inkubasi 60 menit dan perlakuan fraksi bawah gradien 10:20 lama inkubasi 45 menit. |

Inggris) | The purpose of this study was to determine the effect of the use of Bovine Serum Albumin as a sex separation gradient medium and the length of incubation on the viability and intact acrosome cap (IAC) of Peranakan Etawah goat spermatozoa. The research material that will be used is Peranakan Etawah goat spermatozoa from the Experimental Farm, the materials used are physiological NaCl, eosin-nigrosin, and Bovine Serum Albumin (BSA) gradient. The tools used are a centrifuge, centrifuge tube, microscope, cover glass, object glass, water bath, thermos, artificial vagina, micropipette, pipette, glass beaker, syringe, microtube, stopwatch, label paper, Erlenmeyer, thermometer, bunsen heater, stove, and pan. The research was conducted at the Animal Health Laboratory and Experimental Farm, Faculty of Animal Husbandry, Universitas Jenderal Soedirman. The research method that will be used is an experimental method using a 2x2x3 factorial RAK test with 3 times tapping frequency block and consists of three factors, namely fraction (F) consists of two levels (top and bottom), gradient (G) consists of two levels (g1 = 5:15 and g2 = 10:20) and incubation time factor (T) consists of three levels (30, 45, and 60 minutes). The results showed a very significant interaction effect between fractions, BSA gradient medium, and different lengths of incubation time on viability and intact acrosome cap (P < 0.01). Based on the results of the study, it can be concluded that the viability of Peranakan Etawah goat spermatozoa from the best sex separation overall is in the treatment of fractions on a 5:15 gradient for 60 minutes incubation and also the treatment of fractions on a 10:20 gradient for 60 minutes incubation. The best overall intact acrosome cap of sex-separated Peranakan Etawah goat spermatozoa in the treatment of the upper fraction of the gradient 10:20 incubation duration of 30 minutes, the treatment of the upper fraction of the gradient 10:20 incubation duration of 60 minutes and the treatment of the lower fraction of the gradient 10:20 incubation duration of 45 minutes. |
