Indonesia) | Penggunaan obat agen pengkhelat besi memiliki beberapa kelemahan seperti memiliki cakupan terapi sempit, harganya mahal, memiliki beberapa efek samping, dan kepatuhan pasien yang buruk. Senyawa flavonoid dapat mengkhelat zat besi untuk mengurangi akumulasi zat besi. Tanaman yang memiliki kandungan flavonoid yaitu tanaman rimpang C. manga, C. heyneana, dan C. zedoaria. Penelitian ini dilakukan dengan tujuan menentukan kadar flavonoid total dan aktivitas khelasinya serta Penelitian ini merupakan penelitian laboratorium eksperimental menggunakan ekstrak etanol rimpang C. manga, C. heyneana, dan C. zedoaria. Penetapan Kandungan flavonoid total diukur menggunanakan spektrofotometri UV-Vis menggunakan metode AlCl3. Sedangkan pengukuran kadar agen pengkhelat dilakukan menggunakan spektrofotometri UV-Vis dengan reagen ferrozin dan EDTA sebagai standar. Kandungan flavonoid total tertinggi diperoleh dari rimpang C. heyneana yaitu sebesar 16,65 ± 0,53 mg QE/g sedangkan pada C.zedoaria dan C. manga yaitu 10,88 ± 0,33 mg QE/g dan 4,27 ± 0,64 mg QE/g. Aktivitas khelasi besi tertinggi didapat dari C. heyneana yaitu sekitar 43,10 ± 0,54% sedangkan C. manga dan C. zedoaria memiliki kekuatan aktivitas pengkhelat besi 41,22 ± 0,87% dan 41,16 ± 0,51% untuk setiap 5 ppm ekstrak. Inggris) | The use of iron-chelating agent drugs has several disadvantages such as having a narrow therapeutic scope, being expensive, having several side effects, and poor patient compliance. Flavonoid compounds can chelate iron to reduce iron accumulation. Plants that contain flavonoids are C. manga, C. heyneana, and C. zedoaria rhizomes. This study was conducted with the aim of determining the total flavonoid content and chelation activity as well as the correlation between flavonoid content and chelation activity in curcuma rhizomes. This study is an experimental laboratory study using ethanol extracts of C. manga, C. heyneana, and C. zedoaria rhizomes. Determination of total flavonoid content was measured using UV-Vis spectrophotometry using the AlCl3 method. While the measurement of chelating agent content was carried out using UV-Vis spectrophotometry with ferrozine and EDTA reagents as standards. The highest total flavonoid content was obtained from the rhizome of C. heyneana, which was 16.65 ± 0.53 mg QE/g, while in C. zedoaria and C. manga, it was 10.88 ± 0.33 mg QE/g and 4.27 ± 0.64 mg QE/g. The highest iron chelation activity was obtained from C. heyneana, which was around 43.10 ± 0.54%, while C. manga and C. zedoaria had iron chelator activity strengths of 41.22 ± 0.87% and 41.16 ± 0.51% for every 5 ppm of extract.
