| Abstrak (Bhs. Indonesia) | Penelitian dilaksanakan untuk mengetahui preferensi Pentalonia nigronervosa dan potensinya sebagai vektor Banana bunchy top virus pada famili Zingiberaceae. Penelitian dilaksanakan selama empat bulan di screen house dan Laboratorium Perlindungan Tanaman, Fakultas Pertanian, Universitas Jenderal Soedirman. Penelitian dilakukan dalam dua tahap. Tahap pertama bertujuan untuk mendapatkan informasi mengenai tingkat preferensi P. nigronervosa pada enam jenis tanaman meliputi kencur, jahe, kunyit, temulawak, kapulaga, dan lengkuas dengan menggunakan Rancangan Acak Kelompok Lengkap (RAKL). Parameter yang diamati berupa keberadaan P. nigronervosa pada tanaman uji ditinjau dari perilaku makan, natalitas, dan mortalitas. Tahap kedua bertujuan untuk menguji potensi BBTV pada tanaman Zingiberaceae berdasarkan tingkat preferensi tertinggi. Percobaan ini menggunakan Rancangan Acak Lengkap (RAL) dengan parameter yang diamati meliputi masa inkubasi, kejadian penyakit, intensitas penyakit, analisis biokimia, dan analisis jaringan. Data hasil pengujian dianalisis menggunakan sidik ragam ANOVA dan apabila berpengaruh nyata maka dilanjutkan uji Duncan’s Multiple Range Test (DMRT) pada taraf 5%. Hasil penelitian menunjukan bahwa preferensi P. nigronervosa tertinggi terdapat pada tanaman kunyit, lengkuas, dan kapulaga, sedangkan tanaman kencur, temulawak, dan jahe tingkat preferensinya rendah. P. nigronervosa banyak ditemukan pada daun muda dan batang tanaman Zingiberaceae. Infeksi BBTV ditandai dengan munculnya gejala pada tanaman kunyit (masa inkubasi 18 HSI) serta kapulaga dan lengkuas (masa inkubasi masing-masing 24 HSI). Infeksi BBTV juga ditandai dengan reaksi ketahanan tanaman berupa peningkatan kandungan fenol dan penurunan kadar klorofil serta karotenoid. Analisis jaringan tanaman terinfeksi menunjukan terjadinya penebalan lapisan epidermis, pembengkakan floem, xylem mengkerut, dan perubahan struktur parenkim palisade. |
| Abtrak (Bhs. Inggris) | The research was conducted to determine the preference of Pentalonia nigronervosa and its potential as a vector of Banana bunchy top virus in the Zingiberaceae family. The research was conducted for four months at the screen house and Plant Protection Laboratory, Faculty of Agriculture, Universitas Jenderal Soedirman. The research was conducted in two stages. The first stage aimed to obtain information on the level of preference of P. nigronervosa for six types of plants including aromatic ginger, ginger, turmeric, curcuma, cardamom, and galangal using a Randomized Complete Group Design (RCGD). Parameters observed were the presence of P. nigronervosa in the test plants in terms of feeding behavior, natality, and mortality. The second stage aims to test the potential of BBTV on Zingiberaceae plants that have the highest level of preference. This experiment used a completely randomized design (CRD) with observed parameters including incubation period, disease incidence, disease intensity, biochemical analysis, and tissue analysis. Data from the test results were analyzed using ANOVA variance analysis and if there was a significant effect, it was followed by Duncan's Multiple Range Test (DMRT) at the 5% level. The results showed that the highest preference of P. nigronervosa was found in turmeric, galangal, and cardamom plants, while aromatic ginger, curcuma, and ginger plants had low preference levels. P. nigronervosa is mostly found on the young leaves and stems of Zingiberaceae plants. The BBTV infection was characterized by the appearance of symptoms on turmeric (incubation period 18 HSI) and cardamom and galangal (incubation period 24 HSI each). The BBTV infection was also characterized by plant resistance reactions in the form of increased phenol content and decreased chlorophyll and carotenoid levels. Tissue analysis of infected plants showed thickening of the epidermal layer, phloem swelling, xylem contraction, and changes in palisade parenchyma structure. |
