| Abstrak (Bhs. Indonesia) | Penelitian ini bertujuan untuk mengetahui pengaruh medium gradien percoll dan perbedaan lama inkubasi terhadap motilitas dan abnormalitas spermatozoa kambing Peranakan Etawah. Materi yang digunakan yaitu semen dari 2 ekor pejantan kambing Peranakan Etawah, medium percoll, NaCl fisiologis, dan eosin-negrosin. Penelitian ini dilakukan dengan menggunakan Rancangan Acak Kelompok (RAK) pola faktorial 2x3 dengan ulangan sebanyak 3 kali dan terdiri dari dua faktor yaitu faktor gradien 40:55 dan 45:60 serta lama inkubasi 30, 45, dan 60 menit. Data penelitian yang telah diperoleh dianalisis variansi dan dengan uji lanjut Duncan’s Multiple Range Test (DMRT). Hasil penelitian menunjukkan bahwa tidak ada interaksi antara medium gradien percoll dan perbedaan lama inkubasi terhadap motilitas dan abnormalitas (P>0,05), namun setiap faktor memberikan pengaruhnya masing-masing. Beradasarkan faktor gradient percoll pada fraksi atas (Y) didapatkan nilai motilitas dan abnormalitas spermatozoa masing-masing 64,4 – 72,36% dan 8,46 – 9,28%. Adapun nilai motilitas dan abnormalitas spermatozoa fraksi bawah (X) masing-masing yaitu 61,63 – 68,70% dan 7,27 – 8,35%. Nilai motilitas dan abnormalitas spermatozoa Y berdasarkan faktor lama inkubasi yaitu 64,16 – 74,27% dan 8,32 – 8,85%, sedangkan untuk spermatozoa X yaitu 60,44 – 71,70% dan 6,24 – 8,77%. Kesimpulan yang didapat dari penelitian ini adalah motilitas dan abnormalitas spermatozoa kambing Peranakan Etawah hasil separasi seks terbaik pada gradien percoll 40:55 dengan lama inkubasi 30 menit pada suhu 37oC. |

Inggris) | This study aims to determine the effect of Percoll gradient medium and differences in incubation time on the motility and abnormalities of spermatozoa in Etawah Peranakan goats. The materials used were semen from 2 male Etawah crossbreed goats, Percoll medium, physiological NaCl, and eosin-negrosin. This research was carried out using a Randomized Block Design (RAK) with a 2x3 factorial pattern with 3 replications and consisting of two factors, namely gradient factors of 40:55 and 45:60 and incubation times of 30, 45 and 60 minutes. The research data that was obtained was analyzed for variance and further tested by Duncan's Multiple Range Test (DMRT). The results showed that there was no interaction between Percoll gradient medium and differences in incubation time on motility and abnormalities (P>0,05), but each factor had its own influence. Based on the Percoll gradient factor in the upper fraction (Y), the spermatozoa motility and abnormality values were 64,4 – 72,36% and 8,46 – 9,28%, respectively. The motility and abnormality values for lower fraction (X) spermatozoa were 61,63 – 68,70% and 7,27 – 8,35% respectively. The motility and abnormality values for Y spermatozoa based on incubation time factors are 64,16 – 74,27% and 8,32 – 8,85%, while for X spermatozoa they are 60,44 – 71,70% and 6,24 – 8,77 %. The conclusion obtained from this research is the motility and abnormalities of spermatozoa in Etawah crossbreed goats with the best sex separation results on a Percoll gradient of 40:55 with an incubation time of 30 minutes at a temperature of 37oC. |
