Abstrak (Bhs. Indonesia)Isolat BAL LG50 asal sedimen mangrove Pantai Logending berpotensi digunakan sebagai agensia probiotik karena menunjukkan aktivitas antimikroba dan memiliki karakteristik yang sesuai dengan genus Lactobacillus. Syarat sebagai agensia probiotik isolat LG50 harus memiliki ketahanan terhadap cairan asam lambung dan cairan usus halus, salah satunya dengan terenkapsulasi yoghurt. Enkapsulasi merupakan proses menjebak suatu zat (zat aktif) ke dalam zat lain (bahan dinding). Tujuan penelitian ini yaitu mengetahui bagaimana viabilitas isolat BAL LG50 pada kondisi pH saluran pencernaan (lambung dan usus halus) ketika dienkapsulasi pada minuman fermentasi yoghurt. Penelitian ini dilakukan dengan metode Rancangan Acak Lengkap (RAL) pola faktorial yang dilakukan secara terpisah dengan 2 faktor. Faktor pertama yaitu jenis inokulum BAL dan faktor kedua yaitu variasi pH. Masing-masing perlakuan dilakukan pengulangan sebanyak tiga kali sehingga terdapat 36 unit percobaan. Data hasil persentase viabilitas pada simulasi asam lambung dan simulasi usus halus dianalisis menggunakan Analysis of Variance (ANOVA) pada tingkat kepercayaan 95% dan dilanjutkan dengan uji Beda Nyata Terkecil (BNT). Hasil penelitian menunjukkan inokulum bakteri A1 (campuran LG50, L. acidophilus dan S. thermophilus), A2 (campuran L. acidophilus dan S. thermophilus) dan A3 (isolat LG50) menunjukkan viabilitas yang tinggi pada simulasi asam lambung dan usus halus. Total BAL LG50 setelah terenkapsulasi yoghurt sebesar 3,1 × 10^9 CFU/mL.Viabilitas isolat BAL LG50 terenkapsulasi yoghurt pada simulasi asam lambung yaitu memiliki viabilitas sebesar 84,93% pada pH 3 dan sebesar 85,76% pada pH 4. Viabilitas isolat BAL LG50 terenkapsulasi yoghurt pada simulasi usus halus yaitu memiliki viabilitas sebesar 90% pada pH 7 dan sebesar 89,1% pada pH 8.

Inggris)LAB isolate LG50 from logending beach mangrove sediments has the potential to be used as a probiotic agent because it shows antimicrobial activity and has characteristics that correspond to the genus Lactobacillus. The requirements for a probiotic agent, LG50 isolate, must be resistant to gastric acid and small intestine fluids, one of which is encapsulated yoghurt. Encapsulation is the process of trapping a substance (active substance) into another substance (wall material). The aim of this research is to determine the viability of LAB isolate LG50 at the pH conditions of the digestive tract (stomach and small intestine) when encapsulated in fermented yoghurt drinks. This research using the Completely Randomized Design (CRD) factorial pattern method which was carried out separately with 2 factors. The first factor is the type of LAB inoculum and the second factor is pH variation with 4 test levels on gastric pH (P1:pH 3 and P2:pH 4) and small intestine pH (P3:pH 7 and P4:pH 8). Each treatment was repeated three times so that there were 36 experimental units. The independent variables observed were the type of LAB inoculum and pH variations, while the dependent variable observed was the viability of the LAB isolate. The main parameter measured was the percentage of viability of LAB isolates and the supporting parameter was the number of cells of LAB isolates. Data on the percentage of viability results in the stomach acid simulation and small intestine simulation were analyzed using Analysis of Variance (ANOVA) at a confidence level of 95% and continued with the Least Significant Difference (LSD) test. The results showed that bacterial inoculum A1 (a mixture of LG50, L. acidophilus and S. thermophilus), A2 (a mixture of L. acidophilus and S. thermophilus) and A3 (isolate of LG50) showed high viability in simulated stomach and small intestine acids. The total LAB LG50 after being encapsulated in yoghurt was
3,1 × 10^9 CFU/mL. The viability of LAB LAB LG50 isolates encapsulated in yoghurt in simulated stomach acid had a viability of 84.93% at pH 3 and 85.76% at pH 4 . The viability of the yoghurt-encapsulated LAB LG50 isolate in the small intestine simulation had a viability of 90% at pH 7 and 89.1% at pH 8.
