| Abstrak (Bhs. Indonesia) | Pencemaran logam kadmium pada lahan pertanian bawang merah di Brebes diketahui melebihi ambang batas 0,05 ppm sehingga berdampak pada kematian tanaman. Untuk meminimalisir cemaran dapat dilakukan remediasi lahan menggunakan mikroorganisme seperti bakteri. Penelitian ini bertujuan 1) mengetahui identitas isolat bakteri resisten kadmium indigenus perakaran bawang merah melalui analisis bioinformatika berbasis sekuen 16S rRNA; 2) hubungan kekerabatan isolat bakteri dengan bakteri lainnya, 3) karakteristik biokimia isolat bakteri indigenus perakaran bawang merah dan 4) respon perkecambahan dan pertumbuhan tanaman tomat. Penelitian dilaksanakan di Laboratorium Agroekologi Fakultas Pertanian, Universitas Jenderal Soedirman, Purwokerto, Jawa Tengah, dan PT. Genetika Science Indonesia. Penelitian dilaksanakan pada bulan Maret-November 2022. Percobaan ini menggunakan Rancangan Acak Lengkap (RAL) dengan faktor tunggal, yaitu inokulasi isolat bakteri terdiri atas: (P0) = perendaman dengan air steril, (P1) = perendaman dengan isolat bakteri BR2, dan (P2) = perendaman dengan isolat bakteri SJ3. Variabel yang diamati dalam penelitian meliputi, daya kecambah, potensi tumbuh maksimum, First Count Test, Kecepatan Tumbuh, Keserempakan Tumbuh, Bobot Kering Kecambah Normal, Karakter agronomi, pertumbuhan vegetatif. Hasil penelitian menunjukkan identitas masing-masing isolat bakteri SJ3 dan BR2 adalah Bacillus cereus dan Bacillus tropicus. Isolat bakteri SJ3 memiliki hubungan kekerabatan dengan spesies B. cereus dengan identity 98,51%, sedangkan isolat bakteri BR2 memiliki kekerabatan dengan spesies B. tropicus dengan identity 98,13%. Isolat bakteri BR2 dan SJ3 bersifat negatif terhadap hidrolisis urea, hidrolisis CMC, pelarut fosfat, penghasil IAA, dan bersifat positif dalam menghasilkan gelatinase. Hasil penambahan isolat bakteri berpengaruh nyata terhadap bobot kering kecambah normal, tinggi tanaman, dan panjang akar dengan p value < 0,05, tetapi tidak berpengaruh nyata terhadap daya kecambah, potensi tumbuh maksimal, first count test, kecepatan tumbuh, keserempakan tumbuh, dan jumlah daun (p value > 0,05). |
| Abtrak (Bhs. Inggris) | Cadmium metal pollution on shallot farming land in Brebes is known to exceed the threshold of 0.05 ppm, resulting in plant death. Therefore, to minimize cadmium metal contamination, land remediation can be carried out using microorganisms such as bacteria. This study aims to 1) determine the identity of cadmium-resistant bacterial isolates to shallot roots through bioinformatics analysis based on 16S rRNA sequences; 2) the kinship of bacterial isolates with other bacteria recorded in GenBank, 3) biochemical characteristics of indigenous bacterial isolates of shallot roots and 4) response of tomato plant germination and growth through the application of indigenous bacterial isolates of shallot roots. The research was carried out at the Agroecology Laboratory of the Faculty of Agriculture, Jenderal Soedirman University, Purwokerto, Central Java and PT. Genetika Science Indonesia. The research was conducted from March-November 2022. This study uses a completely randomized design (CRD) with a single factor named bacterial isolate inoculation. It consists of: (P0) = soaking with sterile water, (P1) = soaking with BR2 bacterial isolate, and (P2) = soaking with SJ3 bacterial isolate. Variables observed in this study include germination, maximum growth potential, first count test, growth speed, growth uniformity, dry weight of normal sprouts, agronomic characters, vegetative growth. The results obtained the identity of each bacterial isolate SJ3 and BR2 based on N-BLAST results are Bacillus cereus and Bacillus tropicus. The bacterial isolate SJ3 has a kinship with species B. cereus with an identity of 98,51%, while the bacterial isolate BR2 has a kinship with species B. tropicus with identity of 98,13%. Bacterial isolates BR2 and SJ3 are negative for urea hydrolysis, CMC hydrolysis, phosphate solvent, IAA producer, and are positive in producing gelatinase. The results of the addition of bacterial isolates had a significant effect on sprout dry weight, plant height, and root length with p value <0.05, but had no significant effect on germination, maximum growth potential, first count test, growth speed, growth uniformity, and number of leaves (p value> 0.05). |
