| Abstrak (Bhs. Indonesia) | Kantong semar [Nepenthes mirabilis (Lour.) Druce] dikenal sebagai tanaman unik karena memiliki kantong yang berfungsi sebagai perangkap serangga. Saat ini N. mirabilis berada dalam daftar Appendix II CITES sehingga di Indonesia ditetapkan sebagai tumbuhan dilindungi. Upaya perbanyakan N. mirabilis melalui kultur in vitro dapat dipilih karena mampu menghasilkan bibit dalam jumlah banyak dengan waktu relatif singkat. Penelitian dilakukan untuk menentukan formulasi media dan konsentrasi agar yang paling baik dan ekonomis pada perbanyakan tunas mikro N. mirabilis melalui kultur in vitro. Penelitian dilaksanakan dengan Rancangan Acak Lengkap (RAL) pola faktorial. Faktor pertama adalah formulasi media yang terdiri atas empat taraf yaitu media MS full-strength, MS half-strength, MS half-strength + AB Mix, dan AB Mix. Faktor kedua adalah konsentrasi agar yang terdiri atas tiga taraf yaitu 6 g.L-1, 8 g.L-1, dan 10 g.L-1 sehingga menghasilkan 12 kombinasi perlakuan dan setiap perlakuan diulang sebanyak 5 kali. Parameter yang diukur meliputi waktu muncul tunas, jumlah tunas, jumlah daun, dan tinggi tunas. Data dianalisis dengan Analysis of Variance (ANOVA) taraf kepercayaan 95% dan 99%, dilanjutkan dengan uji Duncan taraf kepercayaan 95%. Hasil penelitian menunjukkan pertambahan jumlah tunas, jumlah daun, dan tinggi tunas N. mirabilis dipengaruhi oleh formulasi media, sedangkan waktu muncul tunas hanya dipengaruhi oleh konsentrasi agar. Media MS half-strength menghasilkan jumlah tunas, jumlah daun, dan tinggi tunas terbaik. Sementara, konsentrasi agar 8 g.L-1 menghasilkan waktu muncul tunas tercepat. |
| Abtrak (Bhs. Inggris) | The pitcher plant [Nepenthes mirabilis (Lour.) Druce] is a unique plant because it has a pitcher that functions as an insect trap. Currently N. mirabilis is on the Appendix II CITIES list, therefore it designated as a protected plant in Indonesia. Propagation of N. mirabilis through in vitro culture can be proposed, which can produce large numbers of seedlings in a relatively shorter time. This study has been caried out with a view to determine the best and most economical media formulation and agar concentration for the propagation of N. mirabilis microshoots in in vitro culture.This research using a completely randomized design on a factorial treatment pattern. The first factor was the media formulation, which consisted of four levels: MS full-strength, MS half-strength, MS half-strength + AB Mix, and AB Mix. The second factor was agar concentration, which consisted of three levels: 6 g.L-1, 8 g.L-1, and 10 g.L-1, resulting in 12 treatment combinations and each treatment was repeated five times. The parameters measured including shoot emergence time, number of shoots, number of leaves, and shoot length. The data were analysed using analysis of variance (ANOVA) at 95% and 99% levels of confidence, followed by Duncan's multiple range test at 95% level of confidence. The results showed that the increase of Nepenthes mirabilis (Lour.) Druce number of shoot, number of leaves, and shoot length were controlled by media formulation, whereas the shoot emergence time was controlled by the agar concentration. Half-strength MS resulted in the highest number of shoots and leaves, as well as the longest shoot length. In addition, 8 g.L-1 agar resulted in the fastest shoot emergence time. |
