| Judul Artikel | ISOLASI BAKTERI POTENSI REMEDIATOR PESTISIDA SINTETIK ABAMEKTIN DARI TANAH PERTANAMAN PADI (Oryza sativa L.) DI KABUPATEN PEMALANG |
| Abstrak (Bhs. Indonesia) | Isolat bakteri yang digunakan berasal dari lahan pertanaman padi di Kabupaten Pemalang, Kecamatan Moga, Desa Mndiraja dengan kode isolat yaitu, KMAba, KTAba1, KTAba2, KBAba1 dan KBAba2. Tahapan penelitian meliputi pengambilan sampel, pengukuran pH tanah, pengenceran sample tanah hingga 10-5 , penghitungan jumlah dan pengamatan morfologi koloni, isolasi bakteri, pewarnaan gram dan endospora, pengujian kurva pertumbuhan, resistensi, dan remediasi. Tujuan dilakukan penelitian ini adalah untuk mendaptkan isolat resisten terhadap pestisida abamektin, melakukan karakteristik isolat bakteri resisten serta mengetahui kemampuan remediasi dari isolat resisten pestisida abamektin. Kadar pH di lima lokasi pengambilan sampel tergolong asam menggunakan metode KCl dan menggunakan metode H2O. Hasil isolasi dari lima lokasi diperoleh lima isolat bakteri dominan reisten abamektin, yaitu KMAba (Dukuh Kembang), KTAba1 (Krajan Timur 1), KTAba2 (Krajan Timur 2), KBAba1 (Krajan Barat 1), dan KBAba2 (Krajan Barat 2). Berdasarkan karakterstik morfologi koloni, kelima isolat mempunyai bentuk bundar, berwarna putih, berukuran kecil (2-3 mm) dan elevasi datar. Berdasarkan pewarnaan gram dan pewarnaan spora, kelima isolat merupakan bakteri gram negatif dan memiliki kemampuan membentuk endospora. Pengamatan kurva pertumbuhan pada medium NB tanpa cemaran abamektin menunjukkan waktu generasi isolat paling baik adalah KTAba1 (Krajan Timur 1), yaitu 2 jam. Pengamatan kurva pertumbuhan di medium cair MSM yang diperkaya abamektin 5 ppm menunjukkan waktu generasi isolat paling baik adalah KBAba1 (Krajan Barat 1), yaitu 9 jam 19 menit. Uji resistensi pada medium MSM yang diperkaya abamektin hingga 20 ppm menunjukkan bahwa isolat KBAba1 (Krajan Barat 1) mempunyai kemampuan resisten tertinggi. Uji remediasi abamektin pada medium MSM cair yang diperkaya 5 ppm menunjukkan bahwa isolat KBAba1 (Krajan Barat 1) mempunyai hasil reduksi tertinggi dengan total penurunan cemaran pestisida sebesar 99%. |

Inggris) | The bacterial isolates used came from rice plants in Pemalang District, Moga District, Mndiraja Village with isolate codes namely Kembang (KMAba), East Krajan 1 (KTAba1), East Krajan 2 (KTAba2), West Krajan 1 (KBAba1) and Krajan West 2 (KBAba2). The stages of the research included sampling, measuring soil pH, diluting the soil sample to 10-5 , counting the number and observing colony morphology, isolating bacteria, gram staining and endospores, testing growth curves, resistance and remediation. The aims of this study were to obtain isolates that were resistant to abamectin pesticides, to characterize resistant bacterial isolates and to determine the remediation ability of isolates that were resistant to abamectin pesticides. The pH levels at the five sampling locations were classified as acidic by the KCl method and by the H2O method. The results of isolation from five locations obtained five dominant bacterial isolates namely KMAba (Dukuh Kembang), KTAba1 (East Krajan 1), KTAba2 (East Krajan 2), KBAba1 (West Krajan 1), and KBAba2 (West Krajan 2). Based on the morphological characteristics of the colonies, the five isolates were round in shape, white in color, small in size (2-3 mm) and at an even elevation. Based on gram staining and spore staining, the five isolates were gram negative bacteria and had the ability to form endospores. Observation of the growth curve on NB media without abamectin contamination showed that the best isolate generation time was KTAba1 (Krajan Timur 1), which was 2 hours. Observation of the growth curve in MSM liquid media enriched with 5 ppm abamectin showed that the best isolate generation time was KBAba1 (Krajan Barat 1) which was 9 hours 19 minutes. Resistance test on MSM media enriched with abamectin up to 20 ppm showed that isolate KBAba1 (Krajan Barat 1) had the highest resistance ability. Abamectin remediation test in 5 ppm enriched liquid MSM media showed that isolate KBAba1 (Krajan Barat 1) had the highest reduction results with a total reduction of pesticide contamination of 99%. |
