| Abstrak (Bhs. Indonesia) | Latar Belakang: Sambiloto (Andrographis paniculata) mengandung andrographolide yang memiliki bioavailabilitas rendah sehingga diformulasikan dalam bentuk nanopartikel. Andrographolide mampu melawan radikal bebas yang dapat memicu terjadinya kematian sel. Penelitian ini dilakukan untuk mengetahui aktivitas nefroprotektif nanopartikel sambiloto berbasis PLGA pada sel vero. Metodologi: Nanopartikel sambiloto (50, 100, 250, 500, dan 1000 ppm) dilakukan uji sitotoksisitas dan uji nefroprotektif. Uji nefroprotektif terdiri atas uji proteksi akut, proteksi kronis, dan recovery menggunakan konsentrasi 100, 250, dan 500 ppm. Pada uji proteksi, nanopartikel diberikan dan diikuti pemberian H2O2. Sedangkan uji recovery dilakukan pemberian H2O2, diikuti pemberian nanopartikel. Uji akut menggunakan inkubasi 24 jam, sedangkan uji kronis selama 72 jam. Keseluruhan uji tersebut menggunakan metode MTT assay dan dilakukan analisis data menggunakan one-way ANOVA. Hasil Penelitian: nanopartikel sambiloto (50 – 1000 ppm) tidak menimbulkan efek toksik terhadap sel vero. Nanopartikel memberikan efek nefroprotektif dengan trend dose-dependent. Nanopartikel konsentrasi tinggi (500 ppm) mampu memberikan efek proteksi akut lebih baik daripada vitamin E. Nanopartikel 500 ppm memberikan efek recovery (p<0,05) yang lebih baik daripada vitamin E. Pada uji proteksi kronis, vitamin E memberikan efek lebih baik dibandingkan nanopartikel 500 ppm. Kesimpulan: nanopartikel sambiloto dengan konsentrasi 500 ppm memberikan aktivitas nefroprotektif terhadap sel vero yang mengalami kerusakan akibat induksi H2O2. |
| Abtrak (Bhs. Inggris) | Background: Sambiloto (Andrographis paniculata) contains andrographolide that has low bioavailability so it is formulated in the form of nanoparticles. Andrographolide ables to fight free radicals that cause cell death. This study was conducted to determine nephroprotective activity of PLGA-based Sambiloto nanoparticles in vero cell line. Methodology: Sambiloto nanoparticles (50, 100, 250, 500, and 1000 ppm) were tested for cytotoxicity and nephroprotective activity. Nephroprotective tests consisted of acute protection, chronic protection, and recovery tests using concentrations of 100, 250 and 500 ppm. The protective effect was tested by administering nanoparticles, followed by H2O2. The recovery effect was tested by administering H2O2, followed by nanoparticles. All tests were carried out using the MTT assay method and data analysis was performed using one-way ANOVA. Results: Sambiloto nanoparticles (50 – 1000 ppm) did not have any toxic effect on vero cell line. Nanoparticles provide nephroprotective activity based on concentration-dependent trend. High concentration nanoparticles (500 ppm) were able to provide better acute protective effect than vitamin E. Nanoparticles 500 ppm provided a better recovery effect (p<0,05) than vitamin E, but the chronic protection effect were lower than vitamin E. Conclusion: Sambiloto nanoparticles with concentration of 500 ppm provide effective nephroprotective activity against H2O2-induced toxicity in vero cells line. |
