Indonesia) | Metode Kjeldahl merupakan metode pengujian kadar protein yang telah diakui Association of Official Agricultural Chemists (AOAC) dinilai kurang efisien dan tersedia metode alternatif yaitu metode gelombang ultrasonik. Tujuan penelitian ini untuk mengetahui perbedaan antara metode Kjeldahl dengan gelombang ultrasonik pada pengujian kadar protein yang diperoleh, tingkat akurasi, presisi, dan linearitas nya. Materi penelitian yang digunakan adalah 30 sampel susu sapi segar dari Kecamatan Baturraden dan Kecamatan Karanglewas serta sampel standar dengan kadar protein 2%, 6%, dan 10%. Materi penelitian dilakukan pengujian kadar protein dengan dua metode tersebut dan triplikasi pada sampel standar. Data penelitian yang diperoleh dilakukan uji analisis deskriptif, dan uji T. Rataan pengujian kadar protein susu segar menunjukkan metode Kjeldahl dan Gelombang Ultrasonik berbeda tidak nyata (2.97±0.37 vs.3.06±0,14%; p>0.05). Hasil uji validasi antara metode Kjeldahl dan Gelombang Ultrasonik menunjukkan akurasi yang berbeda nyata (101,71±5,45vs.85,74±0,99%; p<0,05) dan presisi yang berbeda tidak nyata (0,61vs.0,58%; p>0,05). Uji linearitas menunjukkan respon linear dari kedua metode tersebut (R² = 0,99). Kesimpulan nya, kedua metode dapat digunakan untuk pengujian kadar protein namun metode Kjeldahl lebih akurat dibandingkan metode gelombang ultrasonik. |
| Abtrak (Bhs. Inggris) | Kjeldahl is a method to measure protein in milk that has been recognized by many official organization, but has many disadvantages. A saver and more efficient method is available, namely ultrasonic wave method that proposed as an alternative. The purpose of this research was to determine the accuracy, precision, and linearity as well as real milk protein measurement of both methods. The materials used were 30 samples of fresh cow's milk and standardized milk samples with 2%, 6%, and 10% of protein content. These samples were measured by both methods and triplicated for standardized milk. The obtained data were analyzed by descriptive and t test for comparing both methods. The average of protein by Kjeldahl and ultrasonic while measuring fresh samples were not significantly different (2.97±0.37 vs.3.06±0,14%; p>0.05). The result of Kjeldahl and ultrasonic in the accuracy was significantly different (101.71±5.45vs.85.74±0.99%; p>0.05), but had not significantly different precision level (0,61vs.0.58%; p<0.05). The linearity test showed good response of both methods (R² = 0.998). Finally, both methods can be used in protein measurement but Kjeldahl had more accurate than ultrasonic. |
