| Abstrak (Bhs. Indonesia) | Oral squamous cell carcinoma (OSCC) merupakan kanker pada rongga mulut dengan etiologi multifaktor. Ekspresi berlebihan mRNA murine double minute 2 (MDM2) diketahui meningkatkan degradasi tumor protein p53 (TP53) dan berkontribusi dalam tumorigenesis. Perubahan ekspresi mRNA MDM2 pada kejadian OSCC dengan paparan rokok sebagai salah satu faktor risiko kanker belum diketahui hingga saat ini. Penelitian ini bertujuan untuk mengetahui perbedaan ekspresi mRNA MDM2 pada pasien OSCC perokok dan bukan perokok. Penelitian dilakukan secara observasional analitik dengan rancangan cross-sectional study pada 20 sampel FFPE pasien OSCC dari RSUD Prof. Dr. Margono Soekarjo yang diambil dengan teknik purposive sampling. Metode penelitian, yaitu two step PCR dan penghitungan ekspresi relatif mRNA MDM2 menggunakan metode Livak (2-ΔΔCT). Analisis statistik independent t-test dilakukan untuk melihat perbedaan ekspresi pada kedua kelompok. Rerata ΔCT MDM2 pada kelompok OSCC perokok dan bukan perokok secara berurutan sebesar 3,66±2,30 dan 4,74±1,99. Nilai fold change yang didapatkan sebesar 2,12 (p>0,05). Simpulan dari penelitian ini adalah tidak terdapat perbedaan ekspresi mRNA MDM2 pada pasien OSCC perokok dan bukan perokok, namun pada pasien OSCC perokok mengalami peningkatan ekspresi relatif mRNA MDM2 sebesar 2,12 kali dibandingkan pasien OSCC bukan perokok. |
| Abtrak (Bhs. Inggris) | Oral squamous cell carcinoma (OSCC) is a cancer of the oral cavity with a multifactorial etiology. Overexpression of murine double minute 2 (MDM2) mRNA is known to increase the degradation of the tumor protein p53 (TP53) and contribute to tumorigenesis. Changes in MDM2 mRNA expression in the incidence of OSCC with cigarette exposure as a risk factor are currently unknown. This study aimed to determine the differential mRNA expression of MDM2 between smokers and non-smokers OSCC patients. The study was conducted in an analytical observational with a cross-sectional study design on 20 FFPE samples of OSCC patients from RSUD Prof. Dr. Margono Soekarjo taken by purposive sampling technique. The research method used is two step PCR and MDM2 mRNA relative expression calculation using the Livak’s method (2-ΔΔCT). Statistical analysis of independent t-test was conducted to see the differential expression in the two groups. The mean ΔCT MDM2 in the OSCC group of smokers and non-smokers were 3.66±2.30 and 4.74±1.99, respectively. The fold change value obtained is 2.12 (p>0.05). This study concluded that there was no difference of MDM2 mRNA expression between smokers and non-smokers OSCC patients, however there was an increase in the relative expression of MDM2 mRNA by 2.12-fold in smoker OSCC patients compared to non-smoker OSCC patients. |
