Abstrak (Bhs. Indonesia)Cell and tissue culture has advanced and developed in a short time but still needs serum to support cell growth and survival. This research intended to obtain more information on kidney-derived cell culture from bony lip barb kidney (Osteochilus vittatus), which is still less explored and needs further research. A healthy bony lip barb sized of 12 to 15 cm were purchased from Balai Benih Ikan Pandak, Baturaden. The development of primary cell culture from the kidney fragments was performed in cell culture medium composed of DMEM Low Glucose (1 g/l), with L-Glutamine, with Sodium Pyruvate supplemented with various concentrations (0%, 5%, 10%, and 15%) of Fetal Bovine Serum (Research Grade), Penicillin-Streptomycin, and 2Mm L-Glutamine, and incubated at 29°C with 0.8% CO2. The treatments were done with six replications, so there were 24 experimental units. Primary cell culture achieved confluence on day four and subsequently subcultured. While the primary cell culture development from kidneys gone well, the subcultured cells did not adhere to the substrate. The cell density and cell survival did not significantly affected by the addition of 5% and 15% fetal bovine serum. The cell morphology was evaluated using a phase-contrast microscope and showed two dominant cell types for both primary and subcultures, i.e. erythrocytes and small-rounded cells. The results showed that addition of 10% serum concentration showed the highest cell yield with PDT and PDL for 69 h 23 min and 0.47 times, respectively. Statistical analysis indicated that serum addition significantly increased cell density of primary culture (p<0.05), but did not significantly affect cell density of primary culture nor cell viability in both primary and subculture (p>0.05). A better understanding of serum supplementation to culturing success is essential to establish an auspicious cell line.
