Abstrak (Bhs. Indonesia)Lahan salin belum dimanfaatkan secara optimal karena tanaman pangan sulit untuk bertahan pada kondisi salin. Salah satu tanaman yang peka terhadap salinitas yaitu kedelai. Penelitian ini bertujuan untuk mengetahui pengaruh kadar garam terhadap karakter anatomi dan kandungan prolin daun kedelai [Glycine max (L.) Merrill] Kultivar Grobogan. Rancangan yang digunakan yaitu Rancangan Acak Lengkap (RAL). Perlakuan kadar garam dengan kadar garam 0, 25, 50, 75 dan 100 mM masing-masing perlakuan diulang 5 kali. Parameter yang diamati yaitu panjang dan lebar stomata, kerapatan stomata dan trikomta per 1 mm2 luas daun, tebal lapisan kutikula atas dan bawah, tebal jaringan epidermis atas dan bawah, tebal mesofil, kandungan prolin, bobot basah dan bobot kering. Data hasil penelitian dianalisis menggunakan analisis ragam (ANOVA) dengan tingkat kesalahan 5% dan dilanjutkan Uji Beda Nyata Jujur (BNJ) dengan tingkat kesalahan 5%. Hasil penelitian menunjukkan kadar garam 100 mM dapat menurunkan ukuran panjang dan lebar stomata, kerapatan stomata dan tebal mesofil selain itu dapat meningkatkan tebal epidermis dan kutikula daun kedelai Kultivar Grobogan. Kadar garam 100 mM juga menurunkan bobot basah dan bobot kering tanaman dimana nilai bobot basah yang diperoleh yaitu 5,0080 g dan bobot kering yaitu 1,9060 g serta berpengaruh terhadap peningkatan akumulasi senyawa osmotik terlarut yaitu kandungan prolin dengan kadar yang diperoleh yaitu 0,0060 μ/mol.
Abtrak (Bhs. Inggris)Saline land has not been used optimally because food crops are difficult to survive in saline conditions. One of the plants that are sensitive to salinity is soybean. This study aimed to determine the effect of salt content on the anatomical character and proline content of soybean leaves [Glycine max (L.) Merrill] Grobogan Cultivar. The design used is Completely Randomized Design (CRD). Treatment of salt content with a salt content of 0, 25, 50, 75 and 100 mM each treatment was repeated 5 times. Parameters observed were stomata length and width, stomata and trichome density per 1 mm2 leaf area, upper and lower cuticle thickness, upper and lower epidermal tissue thickness, mesophyll thickness, proline content, wet weight and dry weight. The research data were analyzed using analysis of variance (ANOVA) with an error rate of 5% and continued with the Honest Significant Difference Test (HSD) with an error rate of 5%. The results showed that 100 mM salt content could decrease the length and width of stomata, stomata density and mesophyll thickness, besides that it could increase the thickness of the epidermis and cuticle of soybean leaves of Grobogan cultivar. The salt content of 100 mM also reduced the wet weight and dry weight of the plant obtained, namely 5.0080 g and dry weight, namely 1.9060 g and had an effect on increasing the accumulation of dissolved osmotic compounds, namely proline content with the obtained level of 0.0060 μ/mol.
