| Abstrak (Bhs. Indonesia) | Sedimen mangrove kaya akan karbon organik, di antaranya lignin yang terdapat pada serasah daun, batang, dan akar tanaman mangrove yang membusuk. Mikroorganisme lingkungan mangrove mampu menghasilkan enzim ligninase yang dapat mendegradasi lignin. Bakteri asal sedimen mangrove pantai Logending Kebumen belum diketahui kemampuan ligninolitik dan aktivitasnya pada pH dan suhu yang berbeda. Penelitian ini bertujuan untuk mengetahui kemampuan ligninolitik bakteri asal sedimen mangrove pantai Logending Kebumen, mengetahui pengaruh suhu dan pH terhadap aktivitas ligninase yang dihasilkan dan mengetahui identitas bakteri ligninolitik berdasarkan karakteristik fenetik. Penelitian dilakukan dengan metode eksperimental menggunakan rancangan penelitian Rancangan Acak Lengkap (RAL). Perlakuan pengaruh suhu terhadap aktivitas ligninase berupa variasi suhu (30ºC, 35ºC, dan 40ºC) dan perlakuan pengaruh pH terhadap aktivitas ligninase berupa variasi pH (6,0, 7,0, dan 8,0). Masing-masing perlakuan dilakukan pengulangan sebanyak 6 kali. Parameter utama yaitu aktivitas enzim ligninase dan paramater pendukung terdiri atas indeks ligninolitik, jumlah sel bakteri, dan identitas bakteri ligninolitik. Analisis data menggunakan one way ANOVA (P = 0,95) dan identifikasi bakteri ligninolitik mengacu pada Bergey's Manual of Systematics Bacteriology. Hasil penelitian menunjukkan bahwa isolat dengan indeks ligninolitik tertinggi LG113 sebesar 0,256. Suhu optimum aktivitas ligninase isolat LG113 adalah 40ºC sebesar 3,40 U.mL-1 dan pH optimum adalah 7 sebesar 1,02 U.mL-1. Jumlah total sel bakteri ligninolitik pada akhir masa produksi adalah 1,52 x 103 CFU.mL-1. Identitas isolat bakteri LG113 asal sedimen mangrove pantai Logending yaitu termasuk spesies anggota genus Bacillus. |
| Abtrak (Bhs. Inggris) | Mangrove sediments are rich in organic carbon, including lignin found in leaf litter, stems, and rotting roots of mangrove plants. Mangrove environmental microorganisms can produce ligninase enzymes which can degrade lignin. Bacteria from mangrove sediments at the Logending Kebumen beach have not been known yet for their ligninolytic ability and activity at different pH and temperatures. This research aimed to determine the ligninolytic ability of bacteria isolated from mangrove sediments at Logending Kebumen beach, the effect of temperature and pH on the ligninase activity produced, and the identity of ligninolytic bacteria based on phenetic characteristics. The research employed an experimental approach with a completely random design (CRD). Treatment of the effect of temperature on ligninase activity in the form of temperature variations (30ºC, 35ºC, and 40ºC) and treatment of the effect of pH on ligninase activity in the form of variations in pH (6,0, 7,0, and 8,0). Each treatment was repeated 6 times. The principal parameters were ligninase enzyme activity, and supporting parameters consist of the ligninolytic index, number of bacterial cells, and identity of ligninolytic bacteria. Data analysis utilised one-way (ANOVA) and identified ligninolytic bacteria refers to Bergey's Manual of Systematic Bacteriology. The results indicated that the isolate LG113 with the highest ligninolytic index value of 0,256. The optimum temperature for the ligninase activity of the LG113 isolate was 40ºC with the value of 3,40 U.mL-1 , and the optimum pH was 7 with the value of 1,02 U.mL-1. The total number of ligninolytic bacterial cells at the end of the production period was 1,52 x 103 CFU.mL-1. The identity of the bacterial isolate LG113 from mangrove sediments Logending Kebumen beach belonged to species member of the genus Bacillus. |
